The objective of this study was to assess fermentability by canine gut microflora to include shortchain fatty acid (SCFA) production, organic matter (OM) disappearance, and gas production of vegetable and fruit fiber sources compared to fiber standards (psyllium, citrus pectin, and Solka Floc). Fiber sources included apple pomace, carrot pomace, flaxseed, fruit blend (mixture of peach, almond, nectarine, and plum), grape pomace, pea hulls, pistachio, and tomato pomace. Substrates were fermented in vitro for 4, 12, and 24 h with fecal flora obtained from three healthy dogs. Citrus pectin had the highest OM disappearance, SCFA production, and gas production at all times of fermentation; psyllium was intermediate and Solka Floc was lowest. A wide variation in fermentability was noted among the vegetable and fruit fiber sources. Apple pomace, carrot pomace, and flaxseed had the greatest fermentability as assessed by OM disappearance. Pea hulls and tomato pomace had intermediate OM disappearances, and fruit blend, grape pomace, and pistachio were poorly fermented. Carrot pomace produced the largest amounts of gas and SCFA. Apple pomace produced high concentrations of gas but intermediate concentrations of SCFA. Pea hulls and tomato pomace produced intermediate concentrations of gas and SCFA, whereas flaxseed, fruit blend, grape pomace, and pistachio produced low amounts of these fermentation products. For all substrates collectively, OM disappearance was highly correlated with both gas production (r2 = 0.782 and 0.723 for 12- and 24-h values, respectively) and SCFA production (r2 = 0.737 and 0.738 for 12- and 24-h values, respectively). In general, OM disappearance, gas production, and SCFA production were related to the insoluble:soluble fiber ratio in the samples; as the insoluble:soluble ratio decreased (increased soluble fiber), the OM disappearance, gas production, and SCFA production increased.  相似文献

Addition of fat to the diet of the equine is a popular method of increasing energy density of the diet while reducing feed intake. Reducing feed intake is of interest to race horse trainers because additional feed is seen as additional weight and, therefore, a hindrance to performance. Limited information is available regarding the interactions of fat with other dietary components, particularly fiber, in the equine digestive system. The effect of dietary fat on in vitro nutrient disappearance in equine cecal fluid was studied in Exp. 1 using a split-plot design within a 2 x 2 Latin square. Two ponies were fed alfalfa (ALF) alone or alfalfa plus 100 g/d corn oil. Five substrates were used to determine in vitro DM disappearance, OM disappearance, NDF disappearance, and total dietary fiber (TDF) disappearance. The substrates included: ALF, tall fescue (TF), red clover (RC), soybean hulls (SBH), and rolled oats (RO). Fat supplementation did not affect in vitro DM, OM, or NDF disappearance. Addition of fat to the diet increased (P < 0.05) the disappearance of NDF in RO. Among substrates, in vitro DM and OM disappearance were highest (P < 0.05) for RO, followed by SBH, ALF, RC, and TF. In vitro NDF and TDF disappearance were highest (P < 0.05) for SBH, followed by RO, ALF, RC, and TF. In Exp. 2, the effects of varying levels of fat on nutrient intake and total tract digestibility were examined using a 4 x 4 Latin square design. Four mature mares were fed a 60% forage-40% concentrate diet containing different concentrations of fat: 0% supplemental fat control (C); 5% supplemental corn oil (5% CO); 10% supplemental corn oil (10% CO); or 15% supplemental corn oil (15% CO). Treatment did not affect intake of the concentrate portion of the diet or CP, gross energy, or NDF intake. Mares consuming the C diet had the highest (P < 0.05) intake of alfalfa cubes, DM, and OM, followed by those on the 10, 5, and 15% CO treatments, respectively. Treatment did not affect nutrient digestibility. Mares consuming the 15% CO diet had the highest (P < 0.05) fat digestibility, whereas those consuming C had the lowest fat digestibility. Fat in the form of CO generally had little effect on in vitro and in vivo nutrient digestibilities in horses.  相似文献

Fructans are fermentable carbohydrates and include short-chain fructooligosaccharides (scFOS), inulin, and hydrolyzed inulin (oligofructose, OF). Two studies with dogs were designed to examine the effects of low concentrations of fructans on nutrient digestibilities, fecal microbial populations, and endproducts of protein fermentation, and fecal characteristics. In Exp. 1, 11 adult male beagles were fed corn-based, kibbled diets supplemented with or without OF to provide 1.9 +/- 0.6 g/d. Dietary inclusion of OF decreased (P < 0.05) nutrient digestibilities, but did not affect fecal characteristics. Increasing OF concentration tended (P < 0.06) to linearly decrease fecal ammonia concentrations, but not those of branched-chain fatty acids (BCFA), amines, indole, or phenols. Fecal concentrations of total short-chain fatty acids (SCFA) and butyrate tended to be higher in OF-supplemented dogs (P < 0.10), as was the ratio of bifidobacteria to total anaerobes (P = 0.15). In Exp. 2, ileally cannulated adult female hounds were fed a meat-based kibbled diet and were assigned to four scFOS treatments (0, 1, 2, or 3 g/d) in a 4 x 4 Latin square design. Ileal nutrient digestibilities tended to increase (P < 0.15) with increasing concentrations of scFOS. On a DMI basis, fecal output tended to decrease linearly (P < 0.10) in response to increasing scFOS supplementation, whereas fecal score tended to exhibit a quadratic response (P = 0.12). In general, fecal concentrations of SCFA, BCFA, ammonia, phenols, and indoles were not altered by supplemental scFOS. Supplementation of scFOS increased fecal concentrations of total aerobes (P < 0.05) and decreased concentrations of Clostridium perfringens (P < 0.05). From these data, it seems that low levels of supplemental fructans have divergent effects on nutrient digestibility and fermentative endproducts, but do not adversely affect nutrient digestibility or fecal characteristics and may improve colonic microbial ecology in dogs.  相似文献

The objectives of this work were to determine the changes in the expression of neuroendocrine markers in Leydig cell by oestradiol treatment, and to determine whether testosterone is able to recover partially the effects of hormonal suppression induced by oestradiol. Adult male rats were injected daily with either 50 microg of oestradiol or oestradiol plus testosterone propionate (25 mg every 3 days) for 15 days. The animals were sacrificed and testicles were dissected and processed by routine histological protocols. FSH and LH serum levels were determined by radioimmunoassay. The visualization of antigens was achieved by the streptavidin-peroxidase immunohistochemical method. Antibodies against chromogranin A (CrA), S-100 protein (S-100), P substance (PS), synaptofisin (SYN), neurofilament protein (NF), gliofibrillary acidic protein (GFAP) and neuron specific enolase (NSE) were used. The mean LH and FSH serum concentrations were consistently suppressed with hormonal treatments. Intermediate filaments (NF and GFAP) showed no difference in their expression. The expression of S-100, NSE and SYN was significantly lower in both hormone-treated groups. In oestradiol-treated rats, the immunoreactivity of CrA and SP decreased significantly but was restored after testosterone supplementation. Although the nature and functions of many of these substances in Leydig cells remain unknown, these results are consistent with the hypothesis that the expression of some neuroendocrine markers is hormonally controlled.  相似文献

The intestinal supply of amino acids (AA) in sheep fed alkaline hydrogen peroxide-treated wheat straw (AHPWS)-based diets supplemented with soybean meal (SBM) or corn grain plus combinations of corn gluten meal (CGM) and blood meal (BM) was measured in a 5 X 5 latin square. Sheep (avg wt 45 kg) with ruminal, duodenal and ileal cannulas were fed diets containing 65% AHPWS supplemented with the following protein sources: soybean meal (SBM), corn gluten meal (CGM), blood meal (BM), 2/3 CGM:1/3 BM and 1/3 CGM:2/3 BM. Total nitrogen (N) flow at the duodenum was not affected (P greater than .05) by protein source. Flows of bacterial N and AA increased (P less than .05) and flows of nonbacterial N and AA decreased (P less than .05) when wethers were fed SBM vs corn plus other protein sources. When diets contained SBM, quantities of total AA at the duodenum were lower (P less than .05) and the profile of AA supplied to the intestine was altered substantially. Total flows of AA at the duodenum and total quantities of AA disappearing from the small intestine were similar (P greater than .05) for all diets containing BM and CGM, but flows and disappearance of valine, histidine, lysine and arginine increased linearly (P less than .05), whereas flows and disappearance of leucine, isoleucine and methionine decreased linearly (P less than .05) as BM replaced CGM in the diets. Results suggest that quantities of individual AA flowing to the duodenum and disappearing from the intestine of wethers fed AHPWS-based diets can be altered by source of dietary protein. Furthermore, feeding protein sources resistant to ruminal degradation in combination may improve the profile of AA supplied to the intestine.  相似文献

Sulfur-containing amino acid requirement of rapidly growing steers   总被引:3,自引:0,他引:3  
Eight ruminally cannulated steers (294 kg, ADG = 1.3 kg/d) were used in a N retention study (8 x 8 latin-square design) to evaluate sulfur-containing (S) amino acid (AA) requirements for growth. Treatments were abomasal infusions of seven levels of L-methionine (0, 3, 6, 9, 12, 15 and 18 g/d) and one level of DL-methionine (6 g/d). All steers were fed a semipurified diet based on ammoniated corn cobs (DMI = 6.56 kg/d) and were abomasally infused with 400 g/d dextrose and 296.4 g/d of crystalline AA that simulated the non-S-AA pattern of casein. Infusion of 3 g/d supplemental L-methionine maximized N retention in steers. Intestinal flows of absorbable S-AA were determined to be 1.89 g/kg DMI. Breakpoint analysis of retained N as a function of total absorbable S-AA yielded a total S-AA requirement of 14.7 g/d. Nitrogen retention for DL-methionine (36.4 g/d) was not different (P greater than .05) from that for 6 g/d L-methionine (38.8 g/d), but because this value was not in the linear response range, the efficacy of DL-methionine in meeting S-AA needs could not be evaluated. Plasma methionine concentrations increased linearly (P less than .05) in response to L-methionine infusion and were greater (P less than .05) for steers infused with 6 g/d DL-methionine (45.3 microM) than for steers receiving 6 g/d L-methionine (30.5 microM). A sheep digestion trial and two feedlot trials with cattle were conducted to study effects of an ionophore, salinomycin, on nutrient digestibility, ruminal metabolism and cattle performance. In trial 1, addition of salinomycin at 5.5, 11 or 22 ppm to 60% concentrate diets fed to ruminal-cannulated rams had no effect (P greater than .05) on apparent digestibility of dry matter (DM), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF) or starch in comparison with control diets. Apparent nitrogen (N) digestibility was increased (P less than .05) in animals fed salinomycin. Salinomycin did not affect total volatile fatty acid (VFA) concentrations in the rumen, but resulted in a linear (P less than .05) increase in molar proportion of propionate and a linear (P less than .05) decrease in molar proportions of acetate and butyrate and in acetate:propionate ratios. Shifts in VFA proportions were fully expressed within 4 d after salinomycin was added to the diet. In trial 2, salinomycin was added at 5.5, 11, 22 or 33 ppm to 85% concentrate diets fed to finishing steers. Salinomycin level had a quadratic effect (P less than .05) on daily weight gain and resulted in a quadratic (P less than .05) decrease in feed intake with a resultant average improvement of 10.3% in feed efficiency. An experiment was conducted to develop a system useful for measuring methionine requirements of growing steers. Seven ruminally cannulated steers (312 kg, gaining .91 kg/d) were fed a diet based on ammoniated corn cobs, corn starch, molasses and urea. Quantities of N and sulfur-containing amino acids disappearing from the small intestine were 96.0 and 14.8 g/d, respectively. Postruminal infusions of Na-caseinate (CAS) resulted in linear (P less than .01) increases in N retention with values increasing from 30.1 g/d with no postruminal CAS infusion to 39.3, 50.8 and 59.2 g/d (averaged across methionine supplementation) when 100, 200 and 300 g/d CAS were infused. Postruminal infusions of a mixture of crystalline L-amino acids (simulating the nonsulfur-containing essential amino acid pattern of casein; SIM) at levels of 100, 200 and 300 g/d also led to linear increases (P less than .01) in N retention with steers retaining 30.9, 38.9 and 50.5 g N/d (averaged across methionine supplementation), respectively. Postruminal infusion of 12 g/d L-methionine across CAS and SIM infusions improved (P less than .01) N retention by 7.6 g/d but infusion of SIM, which is devoid of sulfur amino acids, also increased N retention. Responses to methionine supplementation was greatest when 200 or 300 g/d SIM were abomasally infused. The data are interpreted to demonstrate that, for steers fed a diet containing little true protein, postruminal supplementation with nonsulfur-containing amino acids tended to increase the ability of growing steers to respond to methionine supplementation.  相似文献

The objectives of this study were to examine widely divergent fiber sources for their efficacy as ingredients in a meat-based dog diet and to determine the effects of these fibers on fecal excretion responses and mean retention time of marked fiber in the gastrointestinal tract of the dog. Fiber sources tested included beet pulp (BP), tomato pomace (TP), peanut hulls (PH), wheat bran (WB) and alkaline hydrogen peroxide-treated wheat straw (AHPWS). Diets were isonitrogenous (5.3% N) and iso-total dietary fiber (TDF; 12.5%). Thirty female English Pointers (five/treatment) were used in the experiment. Intakes of DM and OM were similar among treatments. The highest intakes of ether extract (EE) occurred on the TP, PH and WB treatments. Dogs fed PH ingested the most crude fiber (23.6 g/d), NDF (53.5 g/d), ADF (34.3 g/d) and TDF (59.7 g/d). Digestibilities of DM and OM for all fiber treatments were lower than the control (87.6 vs 81.8% for DM; 90.2 vs 85.4% for OM), but values were similar among fiber sources. The highest EE and N digestibilities occurred on the control and AHPWS treatments. No differences were noted among exogenous fiber-containing treatments in fiber component digestibility. Digestible energy and ME values generally were similar among treatments. Among fiber sources, BP resulted in the greatest amount of wet feces excreted (270 g/d) and the lowest fecal DM (30.3%). No differences among fiber sources were noted in frequency of defecation or mean retention time. Iso-TDF diets (containing, on average, 12.5% TDF) appear to be utilized similarly, regardless of the diversity in sources of fiber tested.  相似文献

Two experiments were conducted to evaluate alkaline hydrogen peroxide-treated wheat straw (AHPWS) in cattle growing (Exp. 1) and finishing (Exp. 2) diets. In Exp. 1, 162 crossbred steers (257 kg) were fed 66% roughage diets in an 84-d growth trial to compare AHPWS to corn silage (CS) and to evaluate different supplemental CP sources and levels. A completely randomized design with a 3 x 3 factorial arrangement of treatments was used. Factors were roughage source (CS, a 1:1 mixture of CS:AHPWS [MIX] and AHPWS) and CP treatment (13 and 11% CP with supplemental CP provided by soybean meal [13-SBM] and [11-SBM] and 11% CP with a combination of urea, corn gluten meal, and fish meal [UGF]). Lasalocid was fed at the rate of 200 mg per steer daily. Steers fed AHPWS had decreased (P less than .01) DMI compared with steers fed MIX and CS. This may be due to increased dietary Na from residual Na in AHPWS. With each incremental increase in AHPWS, ADG and gain/feed decreased (P less than .01). Dry matter intakes (kg/d), ADG (kg), and gain/feed for CS, MIX, and AHPWS were 8.0, 1.56, and .19; 8.2, 1.33, and .16; and 7.5, 1.08, and .14, respectively. Decreased performance by steers fed AHPWS may be due, in part, to a negative interaction between the lasalocid and dietary minerals. There were no differences in performance due to CP supplementation.
