AFLP markers were used to assess the genetic diversity of 77 breeding lines from three of the world's major canola qualityBrassica juncea breeding programs from Canada (Agriculture and Agri-Food Canada and Saskatchewan Wheat Pool) and Australia (Agriculture Victoria). The objectives of the paper were to assess the genetic diversity within and between these three breeding programs and to assess genetic diversity of the canola quality germplasm as compared to mustard quality B. juncea. Fifteen lines of mustard quality B. juncea from India, China, Russia and Australia were also included in the investigation. Ten EcoR1/Mse1 based primer pairs generated 751 scorable fragments with an average of 26 polymorphic bands per primer pair (35%). Similarity coefficients were calculated using the Simple Matching coefficient and adendrogram was developed using the UPGMA procedure, resulting in germplasm being partitioned into five main groups. Line specific markers were discovered that have potential in enhancing the efficiency of individual breeding programs using breeding techniques like accelerated backcrossing. Further understanding the genetic diversity within and between programs has implications for future breeding and collaboration within and between the three programs. A simple, automated wet digestion procedure was developed for the quantitative determination by atomic absorption spectroscopy (AAS) of arsenic, cadmium, copper, mercury, lead, selenium, and zinc in animal tissue. A commercial digestion block system with automated temperature programming was used. Recoveries of all elements from spiked bovine liver and kidney samples exceed 95%. The analytical results obtained for samples of NBS Bovine Liver (No. 1577a) agree well with certified values. The procedure is safe and requires minimum analyst time.  相似文献

This paper describes poisonings that occurred when two cows were fed, small quantities of minced white clover per rumen fistulam over a two-hour period. One animal died; the other survived following treatment with a specific antidote. The level of cyanide found in the leaf material was 200 /g greentissue. It seemsclear from calculations based on this result that the intake of glucosides was sufficient to account for the results observed. Aspects of these cases are discussed in relation to findings of other workers. These cases demonstrate the potential dangers of the high levels of cyanogenetic glucosides in N.Z. white clovers.  相似文献

Brassicaceae weeds are a widespread problem in Australian oilseed rape crops. The weeds not only compete for resources during crop growth, but also have the potential to reduce both oil and meal quality of the harvested crop. This study investigated oil and meal quality of weedy species from the Brassicaceae family that were collected throughout cropping regions of Australia. Eighty‐nine lines from 19 species were grown and evaluated in the same environment for their potential to contaminate Australian oilseed rape seed lots. Seed and flowering characteristics of each species were also examined. The glucosinolate concentration of most of the weedy species was greater than 100 μmol g?1 of oil‐free meal, well above the threshold for meeting oilseed rape quality. Erucic acid content of 18 of the 19 weedy species also exceeded the oilseed rape quality standard of less than 2% erucic acid. This study highlights the potential of the weedy species to reduce the quality of Australian oilseed rape crops.  相似文献

Tracheal collapse and bilateral laryngeal paralysis were diagnosed in an 8-month-old Cocker Spaniel that had acute onset of dyspnea and cyanosis. Surgical exploration of the mediastinum revealed an abscess involving the ventral wall of the trachea immediately caudal to the thoracic inlet. Both recurrent laryngeal nerves were entrapped in fibrous tissue surrounding the abscess. The dog recovered after tracheal resection and anastomosis and freeing of the entrapped nerves. The peritracheal abscess was suspected to have been the result of esophageal perforation secondary to foreign body penetration.  相似文献

Keratoconjunctivitis sicca is the major cause of chronic or recurrent conjunctivitis in dogs. The diagnosis of KCS is often delayed or mistaken for allergic or bacterial conjunctivitis and inappropriate or insufficient treatment leads to progressive corneal scarring and blindness in many dogs. The cause of KCS in dogs has not been proven, but evidence supports an immune-mediated etiology for the majority of cases. In an attempt to specifically treat the cause of KCS, a new immunosuppressant drug, cyclosporine, has been used in a topical preparation. Treatment results using ophthalmic cyclosporine are dramatic compared with historical treatments. Resolution of corneal scarring and increased lacrimation are seen in most dogs. Investigations of the mechanism of action of ophthalmic cyclosporine suggest the efficacy may have been serendipitous. A local neuroendocrine effect of cyclosporine on the lacrimal gland may actually be responsible for the increased lacrimation. Early diagnosis of KCS and treatment with ophthalmic cyclosporine may avert a major cause of blindness in dogs.  相似文献

An improved method has been developed for the determination of benzyl penicillin in animal tissues. Tissues are fortified with a known amount of penicillin V (internal standard) and extracted with water. The extract is deproteinized with sulfuric acid and sodium tungstate, filtered, and concentrated on a conditioned C18 solid phase extraction column. Penicillin V and benzyl penicillin are then eluted from the column with 1 mL 60% acetonitrile-35% water-5% 0.2M phosphate buffer solution and derivatized with 1 mL 1,2,4-triazole-mercuric chloride solution at 65 degrees C for 30 min. An aliquot of this sample is analyzed by reverse phase liquid chromatography with UV detection at 325 nm. The limit of detection is 5 micrograms/kg (ppb) penicillin G (8.4 IU/kg) in liver, kidney, and muscle tissues).  相似文献

Levels of arsenic, cadmium, copper, mercury and lead were determined in approximately 650 samples of liver and kidney from cattle, swine and poultry slaughtered in Canada during 1979-81. In addition zinc levels were determined in livers and kidneys from swine, and selenium and zinc levels were determined in the livers and kidneys from cattle. Depending on the element several methods of atomic absorption spectroscopy were used to analyze samples including flame, hydride generation, cold vapour generation and graphite furnace atomization. Analyses were also done by plasma emission spectroscopy. Levels of arsenic over 2.0 micrograms/g were detected in 0.9% of swine livers and 0.3% of swine kidneys. Cadmium levels higher than 1.0 micrograms/g were detected in 0.3% of cattle livers, 10.8% of cattle kidneys, 1.8% of swine kidneys, 0.4% of poultry livers and 0.3% of poultry kidneys. Levels of copper over 150 micrograms/g were detected in 0.4% of cattle and swine livers. Levels of lead over 2.0 micrograms/g were detected in 1.4% of poultry livers and 1.6% of poultry kidneys. The highest level of mercury detected in all species was 0.25 micrograms/g and the highest level of selenium was 1.9 micrograms/g. Zinc levels of over 100 micrograms/g were detected in 1.7% of cattle livers, 0.2% of cattle kidneys and 5.0% of swine livers.  相似文献

This study examined the 130 current full-text titles in the BioOne.1 and BioOne.2 collections to determine whether they are peer-reviewed, contain book reviews, and are supplied by document delivery vendors. We identified indexing coverage in 11 major databases in agriculture and life sciences: Aquatic Sciences and Fisheries Abstracts (ASFA), AGRICOLA, Biological Abstracts, BIOSIS Previews, CAB Abstracts, Current Contents, Environmental Sciences and Pollution Abstracts, GeoRef, Scopus, ISI Web of Science, and Zoological Record. We concluded that the majority of the titles in BioOne collections are refereed and scholarly, indexed in several of the major indexing and abstracting journals in the field of agriculture and life sciences, and available via several major document delivery vendors. We find both BioOne collections to be valuable, useful, cost-effective journal aggregations.  相似文献
