Methane oxidation activity (MOA), methanotrophic bacteria (MOB), and CH4 emission in a paddy field fertilized with anaerobically digested slurry were compared between two varieties: a fodder rice variety Leaf star (LS) and a normal rice variety Kinuhikari (KN). Average MOA and MOB per unit dry root were significantly higher in KN (7.1?μmol?g?1 CH4?g?1 dry root h?1 and 3.7?×?107 MPN?g?1 dry root, respectively) than in LS (4.3?μmol?g?1 CH4?g?1 dry root h?1, 2.1?×?107 MPN?g?1 dry root), although MOA in the rice root per whole plant was not significantly different since root biomass of LS (1.5?g dry root plant?1) was significantly higher than that of KN (1.2?g dry root plant?1). MOA in the soil ranged from 0.031 to 0.087?μmol?g?1 CH4?g?1 dry soil h?1, but there were no significant differences in both depths (0–5 and 5–15?cm) between the two rice varieties. MOA in the rhizosphere soil was significantly different between the rice varieties at flowering, but not at tillering. Methane emission in the field was lower and MOA and MOB in the rice root were higher in LS than in KN at tillering, but the reverse results were found at flowering and maturity stages. Total CH4 emission during a growing period was not significantly different between LS (27.4?±?16.9?g CH4?m?2) and KN (24.0?±?19.5?g CH4?m?2). There was a significant negative relationship between CH4 emission and rice root MOB (P?<?0.01) or MOA (P?<?0.05) and significant positive relationship was observed between root MOA and MOB (P?<?0.01). This study revealed that choice of rice variety might be an important environmental issue in paddy cultivation since it can influence MOA and MOB in rice root and rhizosphere soil which relate with CH4 emission.  相似文献

This paper focuses on N balance in a paddy field planted with whole crop rice (Oryza sativa cv. Kusahonami). The experiment was conducted with two treatments during two rice-growing seasons: one was fertilized with N (160 kg N ha–1; 16N plot) and the other unfertilized (0N plot); both plots were fertilized with P and K. The N input from precipitation was 15 and 12 kg N ha–1 in 2002 and 2003, respectively. The N input from irrigation water reached as much as 123 and 69 kg N ha–1 in 2002 and 2003, respectively. This was because irrigation water contained higher NO3 concentrations ranging from 4 to 8 mg N l–1. The N uptake by rice plants was the major output: 118 and 240 kg N ha–1 in the 0N and 16N plots in 2002 and 103 and 238 kg N ha–1 in 2003, respectively. N losses by leaching were 4.8–5.3 and 6.5–7.3 kg N ha–1 in 2002 and in 2003, respectively. Laboratory experiments were carried out to estimate the amounts of N2 fixation and denitrification. Amount of N2 fixation was 43 and 0 kg N ha–1 in the 0N and 16N plots, respectively. Denitrification potential was quite high in both the plots, and 90% of the N input through irrigation water was lost through denitrification. Collectively, the total N inputs were relatively large due to irrigation water contaminated with NO3, but N outflow loading, expressed as leaching–(irrigation water + precipitation + fertilizer), showed large negative values, suggesting that the whole crop rice field might serve as a constructed wetland for decreasing N.  相似文献

Mutants defective in motility or antibiotics production were obtained by Tn5 mutagenesis of a biocontrol agent Pseudomonas fluorescens MelRC2Rif (wt). Tomato or melon seeds were co-inoculated with a Tn5 mutant and wt in a 1:1 ratio and then grown in soil for 10 days. There was no change in ratios of Tn5 mutants defective in antibiosis to wt in the process of rhizoplane colonization, suggesting little contribution of in vitro antibiosis to the rhizoplane competence of P. fluorescens MelRC2Rif. Similar results were also obtained when seeds treated with bacteria were planted in soil artificially infested with fungal pathogens. In contrast, ratios of Tn5 mutants defective in motility to wt significantly decreased, suggesting the contribution of motility to the rhizoplane competence of this bacterium. When a non-motile Tn5 mutant and wt were co-inoculated into soil at a matric potential of pF 2.3 (–20 kPa) and plants were then grown, there was no change in the ratio in rhizoplane colonization, suggesting that motility might have a role in the movement along roots but an insignificant role in the movement from bulk soil towards roots. When they were co-inoculated into 0.2% water agar (WA) instead of soil, a remarkable decline in ratios was detected. Thus it was soil structure that hindered the efficiency of motility. Time course enumeration of rhizoplane colonization of tomatoes grown in WA revealed that motility was an important means of movement towards and/or along roots rather than the multiplication on roots. The effect of soil freeze–thaw cycles on the denitrification potential was examined based on the C2H2 inhibition method. The gross N2O production curve of the soil sample (incubation with C2H2) showed minor changes between the freeze–thaw treatment and the unfrozen control. However, kinetics analysis revealed that the initial production rate, an indicator of the population density of denitrifying communities, decreased (P = 0.043) and the specific growth rate constant, an indicator of the activity of denitrifying communities, increased (P = 0.039) as a result of the freeze–thaw cycles in five of six soil samples examined. The increase in the specific growth rate constant suggested the stimulation of the activity of denitrifying communities that survived after the freeze–thaw cycles and may explain the minor suppression on the gross N2O production in spite of decreasing the population density of denitrifying communities that was suggested by the initial production rate. The net N2O production curve of the soil sample (incubation without C2H2) showed a remarkable change in one out of six soil samples, and in that one soil sample, N2O release to the atmosphere was largely stimulated (7.6 times) by the freeze–thaw cycles. However, the stimulation of the N2O release by the freeze–thaw cycles was even observed in two other selected soil samples (4.6 and 1.8 times), suggesting that an imbalance in the N2O-producing and N2O-reducing activities of denitrifying communities might complementally explain the N2O release stimulated by the freeze–thaw cycles.  相似文献

The effects of fumigation with sodium methyl dithiocarbamate (metam sodium) on the microbial community structure and function in 2 soils were investigated using a variety of techniques. In both soils ca. 50% and 90% of the populations of total and culturable bacteria, respectively, were killed by fumigation, with recovery to levels prevailing in control soils 26 d after cessation of fumigation. The size of the ammonium and nitrite oxidiser populations was reduced by up to 4 orders of magnitude by fumigation, with the latter showing a slight recovery 105 d later. There were substantial changes in the C-utilisation (Biolog GN) profiles in the fumigated soils even 105 d later. The number and pattern of amplified 16S ribosomal DNA restriction analysis (ARDRA) fragments was changed by fumigation, and there was a shift in the %G+C profile toward a greater proportion of lower %G+C classes in treated soils. It appeared that DNA released from killed cells remained for some time after fumigation, and masked the apparent community DNA profiles. This study demonstrates that the effects of fumigation on the soil microbial community structure and function were pronounced and for some parameters very persistent. However, the effects on broad-scale properties such as total or culturable bacterial numbers were less enduring.  相似文献

Epibiotic microorganisms associated with micro crustaceans in the overlying water of Philippine paddy fields were examined by scanning electron microscopy, and their associations were compared with those observed in Japanese paddy fields (Soil Sci. Plant Nutr., 43, 633-641, 651-664, 1997). Various types of microorganisms, including bacteria, protozoa, and algae, colonized microcrustaceans and their kinds and colonizing patterns varied depending on the kinds of microcrustaceans. Common associations between epibionts and respective micro crustaceans were observed in the Philippines and in Japan: e.g., rodshaped bacteria on Cyclopidae and Cypridopsidae; no microorganisms on Simocephalus spp.  相似文献

To study whether or not passive immunity of neonatal calves against Shiga toxin-producing Escherichia coli (STEC) O26, O111, and O157 was obtained by colostrum administration, serum antibodies in calves after the feeding were determined by enzyme-linked immunosorbent assay (ELISA) in comparison with antibodies in colostrum and sera from donor dams. The highest antibody titers to STEC in colostrum from dams were detected soon after parturition. The antibody titers were found to be elevated in sera of neonatal calves (4-9 hr after birth) orally administered with colostrum with high antibody titers, suggesting that passive immunity of neonatal calves to STEC infection may be obtained by feeding colostrum. These results suggest that colostrum administration to neonatal calves may play an important role in elevating serum antibodies against STEC in neonatal calves.  相似文献

In the early 1990s, three egg-laying farms (farms S, T, and B) were thought to have the possibility of Salmonella enteritidis (SE) contamination because positive liquid egg samples originated from those farms. The present study was therefore conducted. The first clarification for SE contamination was the study on the origin of SE contamination including its vertical transmission. The results of SE contamination profiling with dust and manure, food materials, dead embryos, and residual yolks on hatch day in regular monitoring over a few years were clearly negative. Therefore, we concluded the SE transmission/infection was attributed to horizontal infection in the egg-laying farms but not vertical transmission from parental stock, hatcheries, growth, or food materials during a 7-yr experimental period. Second, we attempted to clarify if administration of an SE bacterin (Layermune SE) to growth flocks for the egg-laying farms could reduce SE incidence in liquid egg samples from each egg-laying farm. In the first experiment, we compared SE incidence in liquid egg samples from vaccinated and nonvaccinated flocks (similar age flocks). SE incidence from vaccinated and nonvaccinated flocks showed negative and <2 most probable number (MPN)/100 ml for farm B, <2 and >1600 MPN/100 ml for farm S, and negative and >1600 MPN/100 ml for farm T, respectively. In the second experiment, we compared the SE isolation incidence in the liquid egg samples from nonvaccinated and newly replaced vaccinated flocks in the same chicken houses from each of the three egg-laying farms. SE incidence in the liquid egg samples was similar to that in the first experiment. Therefore, the SE bacterin may play an important role in reducing the SE incidence of liquid egg samples.  相似文献
