BACKGROUND
Amaranthus palmeri is an aggressive annual weed native to the United States, which has become invasive in some European countries. Populations resistant to acetolactate synthase (ALS) inhibitors have been recorded in Spain and Italy, but the evolutionary origin of the resistance traits remains unknown. Bioassays were conducted to identify cross-resistance to ALS inhibitors and a haplotype-based genetic approach was used to elucidate the origin and distribution of resistance in both countries.RESULTS
Amaranthus palmeri populations were resistant to thifensulfuron-methyl and imazamox, and the 574-Leu mutant ALS allele was found to be the main cause of resistance among them. In two Spanish populations, 376-Glu and 197-Thr mutant ALS alleles were also found. The haplotype analyses revealed the presence of two and four distinct 574-Leu mutant haplotypes in the Italian and Spanish populations, respectively. None was common to both countries, but some mutant haplotypes were shared between geographically close populations or between populations more than 100 km apart. Wide genetic diversity was found in two very close Spanish populations.CONCLUSION
ALS-resistant A. palmeri populations were introduced to Italy and Spain from outside Europe. Populations from both countries have different evolutionary histories and originate from independent introduction events. ALS resistance then spread over short and long distances by seed dispersal. The higher number and genetic diversity among mutant haplotypes from the Spanish populations indicated recurrent invasions. The implementation of control tactics to limit seed dispersal and the establishment of A. palmeri is recommended in both countries. © 2023 The Authors. A biotype of Aster squamatus (Sprengel) Hieronymus with suspected resistance to the ALS-inhibiting herbicide imazapyr was detected in a chicken farm in the province of Seville, Spain, which had been treated once a year with imazapyr for 10 years. Resistance to imazapyr in this biotype was studied using dose-response experiments, absorption and translocation assays, metabolism studies and ALS activity assays. The rate of imazapyr required to inhibit A squamatus growth by 50% (ED50) was 15 times higher for the R (resistant) than for the S (susceptible) biotype. Cross-resistance existed for the ALS-inhibitors imazamox, imazethapyr, amidosulfuron, nicosulfuron, rimsulfuron, triasulfuron and tribenuron, but not for bensulfuron. Control of A squamatus using alternative herbicides was poor with clopyralid, intermediate with quinclorac, amitrole and MCPA, and excellent with 2,4-D, glufosinate and glyphosate. Absorption of [14C]imazapyr increased over time for both the R and S biotypes, and translocation from the treated leaf to shoots and roots was similar in both biotypes, with most of the radioactivity remaining in the treated leaf. No metabolites of imazapyr were detected in either biotype. Sensitivity of the ALS enzyme (target site) to imazapyr was lower for the R biotype (I50(R) = 4.28 x I50(S)). The mechanism of imazapyr resistance in this R biotype appears to be an altered ALS conferring decreased sensitivity to imazapyr at the whole-plant level. 相似文献

A new immunocapture technique has been applied to the diagnosis of ovine brucellosis under experimental conditions. The tests were made on a serum bank derived from both young and adult ewes vaccinated conjunctivally with the Rev 1 strain at a dose of 10(8) to 10(9) colony-forming units. Adult ewes were infected experimentally two-and-a-half years after they had been vaccinated and the results were compared with an unvaccinated control group. The condition of each animal in terms of infection with Brucella melitensis was determined by clinical and bacteriological investigations. The development of the immune response was compared by the rose bengal test, the complement fixation test, the Coombs' test and the immunocapture technique for 180 days after the vaccination and for 410 days after the experimental infection, that is, the two following gestations. The results suggest that the new technique is more specific in animals vaccinated conjunctivally, regardless of their age when they were vaccinated. After the experimental infection, significantly (P < 0.05) fewer of the vaccinated sheep which were free of clinical signs and were not excreting B melitensis reacted positively to the test. 相似文献

An experiment was conducted to determine how an E. coil challenge and dietary clays affect the intestinal barrier of pigs. Two groups of 32 pigs (initial BW: 6.9 ± 1.0 kg) were distributed in a 2 x 4 factorial arrangement of a randomized complete block design (2 challenge treatments: sham or E. coil, and 4 dietary treatments: control, 0.3% smectite A, 0.3% smectite B and 0.3% zeolite), with 8 replicates total. Diarrhea score, growth performance, goblet cell size and number, bacterial translocation from intestinal lumen to lymph nodes, intestinal morphology, and relative amounts of sulfo and sialo mucins were measured. The E. coli challenge reduced performance, increased goblet cell size and number in the ileum, increased bacterial translocation from the intestinal lumen to the lymph nodes, and increased ileal crypt depth. One of the clays (smectite A) tended to increase goblet cell size in ileum, which may indicate enhanced protection. In conclusion, E. coli infection degrades intestinal barrier integrity but smectite A may enhance it. 相似文献

A study was conducted to measure residues of 13 organophosphorus (OP) pesticides, widely used as dairy cattle ectoparasiticides or in crops used for animal feed, in homogenized and pasteurized Mexican milk samples. Four different milk brands with high distribution were collected biweekly during a 12 month period (n = 96) in supermarkets. OP pesticide residues were measured by gas chromatography with a flame photometric detector. Approximately 39.6% of the samples contained detectable levels of OP pesticide residues. Eight samples contained residues exceeding established maximum residue limits (MRL), and the OP pesticides present in these samples were dichlorvos (five samples), phorate, chlorpyrifos, and chlorfenvinphos (one sample, respectively). Average residues of 13 OP pesticides measured were below established MRLs ranging between 0.0051 and 0.0203 ppm. 相似文献

This research compared the toxic effects of aflatoxin B1 and monocrotaline, the active principle of Crotalaria spectabilis, and the additive effect between aflatoxin B1 and monocrotaline in turkey poults. It was of interest whether selenium fed at dosage levels of 0.1, 5, or 10 micrograms/g of feed would protect against the toxic effect of aflatoxin and/or monocrotaline, and whether the toxicants would result in detectable residues in poult tissues. A total of 180 healthy 1-day-old male turkey poults was assigned at random to 12 treatment groups (15 birds/group). Body and liver weight losses, and low serum concentrations in total protein (TP), albumin (A), alpha-globulin (alpha G), and beta-globulin (beta G), as well as high values in gamma-globulin (gamma G), were produced in the groups fed crotalaria. Pathologic changes were induced by monocrotaline with no protection afforded by the added selenium. Low values in TP, A, alpha G, and beta G and in body and liver weights were observed in groups given the combination of aflatoxin plus crotalaria. Gross lesions were associated with an additive toxic effect and a lack of protective effect of selenium against this combination. However, higher values in TP, A, alpha G, and beta G, and liver weights in groups fed aflatoxin B1 plus selenium indicated that selenium had a protective effect against aflatoxin toxicity. Residues of aflatoxin B1 and aflatoxin M1 were found in the kidneys of poults fed aflatoxin B1; also, dehydroretronecine (the metabolite of monocrotaline) was detected in livers of poults fed Crotalaria spectabilis seeds. 相似文献

A 12‐week feeding trial was conducted to examine the replacement of fish meal with pet‐grade poultry by‐product meal (PBM‐PG) in the spotted rose snapper Lutjanus guttatus diet. Five experimental diets were formulated to contain graded levels of PBM‐PG at proportion of 250, 500, 75 or 900 g kg?1. The control diet contained sardine fish meal as the main protein source. Four groups of 15 randomly assigned L. guttatus juveniles were fed to satiation 3 times day?1. Except for the fish fed the PBM‐PG90 diet, the growth performance, survival and feed utilization efficiency of the experimental fish were not significantly lower than those of the control fish. The dietary level of PBM‐PG did significantly affect the haematological characteristics (P < 0.05). The dietary dry matter and protein apparent digestibility coefficients (ADCs) decreased with increasing dietary PBM‐PG. High values for lipid ADCs were observed in all diets, with significant differences among the dietary treatments. The fish whole‐body protein, moisture, lipid and ash contents were not affected by the inclusion of dietary PBM. These results indicate that high‐quality terrestrial PBM can successfully replace more than half of the marine fish meal protein in the L. guttatus diet. 相似文献

Two experimental modules with different stocking densities (M1 = 70 and M2 = 120 shrimp /m2) were examined weekly over a culture cycle in tanks with low‐salinity water (1.9 g/L) and zero water exchange. Results showed survival rates of 87.7 and 11.9% in M1 and M2, respectively. Water temperature, pH, dissolved oxygen, electrical conductivity and chlorophyll a were not significantly (p > .05) different between modules. In contrast, the concentrations of nitrogen compounds were significantly (p < .05) different between modules, except nitrite‐N (M2 were 2.31 ± 1.38 mg/L N‐TAN, 0.18 ± 0.49 mg/L N‐NO2? and 6.83 ± 6.52 mg/L N‐NO3?; in M1: 0.97 ± 0.73 mg/L N‐TAN, 0.05 ± 0.21 mg/L N‐NO2? and 0.63 ± 0.70 mg/L N‐NO3?). When waters of both modules reached higher levels of ammonia and nitrite, histological alterations were observed in gills. In this study, we investigated the ability of matrix‐assisted laser desorption/ionization mass spectrometry (MALDI‐MS) to characterize the lipid contents of canine spermatozoa. For that, samples of pure semen were analysed. Indeed, quite comprehensive lipid coverage was observed, and the most abundant phospholipid ions detected were from four phosphatidylcholines, that is those of m/z 760.6; 782.6; 808.6; and 830.6 and one of m/z 725.6 from a sphingomyelin. In conclusion, MALDI‐MS was found to offer an easy, fast, accurate, and sensitive analytical method for lipid profiling in canine spermatozoa and could be used as a tool to select sires by assessing the relationship between sperm lipid profiles and variables such as age and breeding history as well as to study the effects of cryopreservation on lipid contents. 相似文献
