N-linked protein glycosylation is the most abundant posttranslation modification of secretory proteins in eukaryotes. A wide range of functions are attributed to glycan structures covalently linked to asparagine residues within the asparagine-X-serine/threonine consensus sequence (Asn-Xaa-Ser/Thr). We found an N-linked glycosylation system in the bacterium Campylobacter jejuni and demonstrate that a functional N-linked glycosylation pathway could be transferred into Escherichia coli. Although the bacterial N-glycan differs structurally from its eukaryotic counterparts, the cloning of a universal N-linked glycosylation cassette in E. coli opens up the possibility of engineering permutations of recombinant glycan structures for research and industrial applications.  相似文献

HJ-1卫星发射升空投入运行以来,已在灾害与环境的快速监测与预报领域发挥了重要作用。HJ-1卫星目前已经成功运行3a多,属于超期服役状态。为了评价延寿期影像质量,该文从影像的几何校正精度、灰度特征和纹理特征3个方面对延寿期HJ-1A-CCD影像进行分析,并与正常寿命运行期的HJ-1ACCD影像、同时期的Landsat7 ETM+影像进行比较,分析结果表明:延寿期HJ-1A-CCD影像质量虽然有一定程度的下降,但其与正常寿命期影像质量接近,仍能较好地描述丰富的地物类型及地物细节信息,可以像正常寿命期影像一样,有效地用于环境监测、灾害监测、国土资源管理、城市规划、农业应用等方面。  相似文献

An 8‐week feeding trial was conducted to evaluate the effects of dietary copper sources on growth performance and intestinal microbial communities of juvenile Pacific white shrimp (Litopenaeus vannamei). Four isonitrogenous (420 g/kg crude protein) and isolipidic (70 g/kg crude lipid) experimental diets were formulated to contain different copper sources, Diet NSC (no‐supplemented copper), Diet CS (copper sulphate), Diet Availa (copper amino acid complex from Availa®Cu100) and Diet M (1:1, copper sulphate + copper amino acid complex). A total of 360 Pacific white shrimp juveniles (initial weight: 1.86 ± 0.03 g) were randomly allocated into four groups, with three replicates per group and 30 shrimps each replicate. An Illumina‐based sequencing method was used to analyse the intestinal bacterial composition of shrimp juveniles. The results revealed that shrimps fed Diet M had significantly higher percent weight gain (PWG) than that fed Diet NSC and Diet CS, while there were no significant differences between Diet M and Diet Availa. Survival was not affected by the dietary copper sources. Proteobacteria and Bacteroidetes were dominant in the intestines of Pacific white shrimp. Shrimps fed Diet M and Diet Availa had higher bacterial richness and diversity than those fed Diet NSC and Diet CS with no significant differences among all treatments. Furthermore, Diet M and Diet Availa which contained copper amino acid complex reduced the relative abundance of Vibrionaceae in shrimp intestinal tract. This study indicated that dietary copper sources could affect the intestinal microbial communities and diets supplemented with Availa®Cu100 could have a positive impact on the growth performance and decrease the potential risk of disease in Pacific white shrimp.  相似文献

The effect of cultivar, season, shelf-life and origin on the accuracy of near infrared (NIR) calibration models for the soluble solids content (SSC) and firmness of apple was studied based on a large spectral data set based on approximately 6000 apple fruit from different cultivars, origins, shelf-life exposure time and seasons. To interpret the variance in the spectra with respect to biological variability, functional analysis of variance (FANOVA) was used. From the FANOVA analysis it was concluded that the effects of cultivar, origin and shelf-life exposure time on the NIR spectra were all significant. The largest differences in the spectra were found around the water absorption peaks (970, 1170 and 1450 nm). External validations using independent data sets showed that the accuracy of the models increased considerably when more variability was included in the calibration data set. In general the RMSEP for predictions of the SSC were in the range 0.6–0.8 °Brix, while for Magness Taylor firmness it was 5.9–8.8 N, depending on the cultivar. It was shown that atypical data can lead to large validation errors. It is, therefore, important to collect a calibration data set which is sufficiently representative for future samples to be analyzed with the developed calibration models and to develop simple procedures for model adaptation during practical use.  相似文献

Abstract.— The present study was conducted to investigate the effects of dietary supplementation of β‐1,3 glucan and a laboratory developed feed stimulant, BAISM, as feed additives for juvenile olive flounder, Paralichthys olivaceus. Eight experimental diets were formulated to be isonitrogenous and isocaloric and to contain 50.0% crude protein and 16.4 kJ of available energy/g with or without dietary β‐1,3 glucan and BAISM supplementation. β‐1,3 glucan (G) and BAISM (B) were provided at 0% in the control diet (G0B0) and at 0.05% G + 0.45% B (G0.05B0.45), 0.05% G + 0.95% B (G0.05B0.95), 0.1% G + 0.90% B (G0.1B0.9), 0.10% G + 1.90% B (G0.1B1.9), 0.15% G + 1.35% B (G0.15B1.35), 0.15% G + 2.85% B (G0.15B2.85), and 0.30% G + 2.70% B (G0.3B2.7) in experimental diets. After the feeding trial, fish fed G0.1B0.9, G0.1B1.9, and G0.15B1.35 diets had higher percent weight gain (WG), feed efficiency ratio (FER), specific growth rate (SGR), protein efficiency ratio (PER), and condition factor (CF) than those fed G0B0, G0.05B0.45, G0.05B0.95, G0.15B2.85, and G0.3B2.7 diets (P < 0.05); however, there was no significant differences among fish fed G0.1B0.9, G0.1B1.9, and G0.15B1.35 diets. Fish fed G0.1B0.9 and G0.1B1.9 diets had higher chemiluminescent responses (CL) than those fed the other diets (P < 0.05). Lysozyme activity of fish fed G0.1B0.9 diet was significantly higher than that of fish fed the other diets (P < 0.05). These results indicated that the optimum dietary supplementation level of β‐1,3 glucan and BAISM could be approximately 0.10% β‐1,3 glucan + 0.90% BAISM (G0.1B0.9) of diet based on WG, FER, SGR, PER, CF, CL, and lysozyme activity in juvenile olive flounder, P. olivaceus.  相似文献

A 10‐wk feeding trial was conducted to evaluate the potential use of fermented fisheries by‐products and soybean curd residues mixture (FFSM) as a partial replacement for fish meal (FM) in the diet of juvenile olive flounder, Paralichthys olivaceus. Five experimental diets were formulated with FFSM replacing 0, 15, 30, 45, and 60% of the FM protein (FFSM0, FFSM15, FFSM30, FFSM45, and FFSM60, respectively). Juvenile olive flounder averaging 5.99 ± 0.08 g (mean ± SD) were randomly distributed into aquaria at 15 fish/aquarium, with three replicate aquaria for each experimental diet. Weight gain (WG) of fish fed FFSM0, FFSM15, and FFSM30 was significantly higher than that of fish fed FFSM45 and FFSM60 (P < 0.05). Also, WG of fish fed FFSM45 was significantly higher than that of fish fed FFSM60 (P < 0.05). There were no significant differences in WG among fish fed FFSM0, FFSM15, and FFSM30 (P > 0.05). Specific growth rate (SGR) of fish fed FFSM15 was significantly higher than that of fish fed FFSM45 and FFSM60 (P < 0.05). Also, SGR of fish fed FFSM0, FFSM15, FFSM30, and FFSM45 was significantly higher than that of fish fed FFSM60 (P < 0.05). There were no significant differences in SGR among fish fed FFSM0, FFSM15, and FFSM30 and among those fed FFSM0, FFSM30, and FFSM45 (P > 0.05). Feed efficiency (FE) and protein efficiency ratio (PER) of fish fed FFSM60 were significantly lower than those of fish fed FFSM0, FFSM15, FFSM30, and FFSM45 (P < 0.05); however, there were no significant differences in FE and PER among fish fed FFSM0, FFSM15, FFSM30, and FFSM45. Hepatosomatic index of fish fed FFSM0, FFSM15, and FFSM30 was significantly higher than that of fish fed FFSM60 (P < 0.05); however, there were no significant differences among fish fed FFSM0, FFSM15, FFSM30, and FFSM45 and among those fed FFSM45 and FFSM60. No significant differences were observed in condition factor and survival rate among all dietary groups tested. The whole‐body proximate composition averaged 75.0 (% dry matter basis [DM]), 8.66 (% DM), 16.38 (% DM), and 76.1%, for crude protein, crude lipid, ash, and moisture, respectively. Based on growth performance, the FFSM could replace up to 30% FM protein by the ANOVA test; however, broken‐line model analysis indicated 28.7% as an optimum replacement level in juvenile olive flounder diets.  相似文献

Tomatoes are consumed and cultivated all over the world for not only their pleasant taste but also their curative properties. Therefore, the challenge for growers is to obtain high-quality crop productions by developing new varieties of tomatoes or new ecofriendly fertilizers. This study was to test a bone glue-based foliar fertilizer on the tomato crop. The experiment was organized in a vegetation house under an original treatment scheme. Four types of foliar fertilizers were tested: macroelements, microelements and glue; macroelements and glue; microeleements and glue; and macroelements and microelements without glue. The fertilizers were applied as diluted solutions (0.5% and 1%) across three treatments applied on nine variants. The treatments with bone glue-based foliar fertilizer led to a high-quality production of healthy tomatoes, a good absorption of nutrients together with a reduced nitrites level in tomatoes and an increase of agricultural productivity. The applied foliar fertilizers tested on the hybrid tomatoes used in the experiment had a significant positive influence on vegetative growth. The nitrate concentration in the fruit did not exceed the maximum accepted level. The agronomic analysis of the mineral composition of the fruit revealed that foliar fertilizers with glue significantly influenced nutrient assimilation during the treatments. The level of nitrogen (N), phosphorus (P), and potassium (K) recovery following the application of bone glue foliar fertilizers was higher as compared to the control and the variant without bone glue. Biometric measurements had shown significant differences favorable to tomatoes treated with this bone glue foliar fertilizer.  相似文献

Mycoplasma bovis is perceived as an emerging cause of mortality in feedlot beef cattle. This study examined the lesions and infectious agents in naturally occurring M. bovis-associated bronchopneumonia and arthritis and the relationship of this condition with bovine viral diarrhea virus (BVDV) infection. Standardized pathologic, immunohistochemical, and microbiologic investigations were conducted on 99 calves that died or were euthanized within 60 days after arrival in 72 feedlots. Cranioventral bronchopneumonia with multiple foci of caseous necrosis was identified in 54 of 99 calves, including 30 with concurrent fibrinosuppurative bronchopneumonia typical of pneumonic pasteurellosis. Mycoplasma bovis was consistently identified in these lesions by culture and immunohistochemistry, but also commonly in healthy lungs and those with pneumonia of other causes. Focal lesions of coagulation necrosis, typical of pneumonic pasteurellosis, were often infected with both Mannheimia haemolytica and M. bovis. Arthritis was present in 25 of 54 (46%) calves with M. bovis pneumonia, and all calves with arthritis had pneumonia. BVDV infection was more common in calves with lesions of bacterial pneumonia than in those dying of other causes, but BVDV infection was not more common in calves with caseonecrotic bronchopneumonia than those with fibrinosuppurative bronchopneumonia. Retrospective analysis identified cases of M. bovis pneumonia in the early 1980s that had milder lesions than the current cases. The findings suggest that, in at least some calves, M. bovis induces caseonecrotic bronchopneumonia within the lesions of pneumonic pasteurellosis.  相似文献
