ABSTRACT

1. The objective of this study was to investigate the effect of supplementing broiler diets with xylanase or xylo- oligosaccharide (XOS) on growth performance, the concentration of non-starch polysaccharide (NSP) hydrolysis products in the ileum and concentration of short chain fatty acids (SCFA) in the caeca of broiler chickens.

2. In total, 500 male Ross 308 broilers were used in this 29-day (d) study. The treatments were organised into a 2 × 2 plus 1 factorial arrangement consisting of two additives (xylanase or XOS) at two levels (low or high) plus a control treatment with no additives. This gave five treatments with 100 birds in each treatment group. The diets were slightly deficient in protein by 20 g/kg and energy by 1 MJ/kg.

3. On d 14 and 28, two birds per pen were euthanised, the caeca content collected and analysed for short chain fatty acid (SCFA) concentration. On d 29, six birds per pen were euthanised and ileal digesta were collected and analysed for the concentration of NSP fractions.

4. On d 14, caecal acetic acid, iso-butyric acid, iso-valeric acid, n-valeric acid and total SCFA concentrations were significantly greater (P ≤ 0.05) when diets were supplemented with XOS compared with xylanase.

Ileal concentration of arabinose, galactose and glucuronic acid (GlucA2) were significantly greater (P ≤ 0.05) in the insoluble NSP fraction when diets were supplemented with a high level of xylanase, compared with the control treatment. Ileal concentration of fructose was significantly greater (P ≤ 0.05) in the water soluble NSP when a high level of xylanase or low level of XOS were included in the diet compared with the control.

6. It was concluded that xylanase and XOS had similar effects on NSP concentration and SCFA in the caeca, although there was little effect on performance. This observation demonstrated further benefits of xylanase supplementation in wheat-based broiler diets beyond digesta viscosity reduction and the release of extra nutrients.  相似文献

Total alkalinity and total hardness are familiar variables in aquatic animal production. Aquaculturists—both scientists and practitioners alike—have some understanding of the two variables and of methods for adjusting their concentrations. The chemistry and the biological effects of alkalinity and hardness, however, are more complex than generally realized or depicted in the aquaculture literature. Moreover, the discussions of alkalinity and hardness—alkalinity in particular—found in water chemistry texts are presented in a rigorous manner and without explanation of how the two variables relate to aquaculture. This review provides a thorough but less rigorous discussion of alkalinity and hardness specifically oriented toward aquaculture. Alkalinity and hardness are defined, their sources identified, and analytical methods explained. This is followed by a discussion of the roles of the two variables in aquaculture, including their relationships with carbon dioxide, pH, atmospheric pollution, ammonia, and other inorganic nitrogen compounds, phytoplankton communities, trace metals, animal physiology, and clay turbidity. Liming and other practices to manage alkalinity and hardness are explained. Changes in alkalinity and hardness concentrations that occur over time in aquaculture systems are discussed. Emphasis is placed on interactions among alkalinity, hardness, water quality, and aquacultural production.  相似文献

Superintensive shrimp culture in zero‐exchange, biofloc‐dominated production systems is more biosecure and sustainable than traditional shrimp farming practices. However, successful application of this technology depends upon optimizing dietary formulations, controlling Vibrio outbreaks, and managing accumulative changes in water quality and composition. A 49‐d study investigated the effect of two commercial feeds of differing protein content and an indoor limited‐exchange, biofloc‐dominated culture environment on Litopenaeus vannamei performance and tissue composition, water quality and ionic composition, and Vibrio dynamics. Juveniles (5.3 g) were stocked at 457/m3 into four 40 m3 shallow raceways containing biofloc‐dominated water and fed one of two commercial feeds with differing protein content, 35 or 40%. Shrimp performance, Vibrio populations, and changes in shrimp and culture water composition were monitored. There were no significant differences (P > 0.05) in shrimp performance (survival, weight, growth, specific growth rate, total biomass, yield, feed conversion ratio, and protein efficiency ratio) or proximate composition between feed types. The 40% protein feed resulted in higher culture water nitrate and phosphate concentrations, alkalinity consumption and bicarbonate use, and higher phytoplankton density. The presence of Vibrio, specifically Vibrio parahaemolyticus, reduced shrimp survival. This survival decrease corresponded with increased culture water Vibrio concentrations. Culture water K+ and Mg2+ increased significantly (P < 0.05), and Sr2+, Br?, and Cl? decreased significantly (P < 0.05) over time. While Cu2+ and Zn2+ did increase in shrimp tissue, no heavy metals accumulated to problematic levels in culture water or shrimp tissue. These results demonstrate the importance of monitoring Vibrio populations and ionic composition in limited‐exchange shrimp culture systems.  相似文献

Sweetpotato virus disease (SPVD) is due to the dual infection and synergistic interaction of Sweetpotato feathery mottle potyvirus (SPFMV) and Sweetpotato chlorotic stunt crinivirus(SPCSV), and causes up to 98% yield loss in sweetpotato in East Africa. This study was conducted to determine the inheritance of resistance to SPVD in sweetpotato and to estimate the nature of genetic variance. Ten parental clones varying in reaction to SPVD were crossed in a half diallel mating design to generate 45 full-sib families. The families were graft-inoculated with SPCSV and SPFMV to induce SPVD and evaluated for resistance in a randomized complete block design at two sites in Namulonge, Uganda during 1998–2000. In serological assays for SPFMV and SPCSV,resistance to symptom development and recovery from initial systemic SPVD symptoms, characterised resistant genotypes. Genetic component analysis showed significant effects for both general combining ability (GCA) and specific combining ability (SCA) for resistance to SPVD. GCA to SCA variance component ratios were large (0.51–0.87), hence GCA effects were more important than SCA effects. Resistant parents exhibited high GCA indicating that additive gene effects were predominant in the inheritance of resistance to SPVD and recovery. Narrow-sense heritability (31–41%) and broad-sense heritability (73–98%) were moderate to high, indicating that rapid genetic gains for SPVD resistance could be accomplished by mass selection breeding techniques. Two genotypes, New Kawogo and Sowola, had high negative GCA effects and had several families in specific crosses,which exhibited rapid recovery from SPVD,and are promising parents for enhancement of SPVD resistance and recovery. Effects of harvest moisture and bale wrapping on forage quality and mold formation in orchardgrass (Dactylis glomerata L.) hay have not been investigated. The objectives of this study were to determine the effects of initial bale moisture and plastic wrapping on temperature, forage quality (protein, fiber components, and digestible energy), and mold formation in large round-baled orchardgrass hay. In all, 40 round bales of mature orchardgrass hay measuring 1.2 × 1.5 m2 were baled at three different moisture ranges (eight bales per treatment): 124 to 166 g/kg (low moisture); 180 to 232 g/kg (intermediate moisture); and 259 to 337 g/kg (high moisture). Selected bales within each moisture range were individually wrapped in plastic (16 bales), and temperature sensors were placed in each bale for up to 10 weeks. The lowest (P ≤ .01) maximum temperature and heating degree-day accumulations were observed when initial bale moisture content was 124 g/kg or when hay was wrapped, regardless of initial moisture content. In 2008 and 2009, all wrapped hays resulted in similar forage quality (P ≤ .14) and mold counts (P = .94) compared with 124 g/kg moisture hay. Hay baled at 166 g/kg resulted in fiber (P ≥ .82) and mold (P = .21) components similar to higher moisture bales. Mold counts for hay baled at 166 g/kg and 124 g/kg moisture were 24.8 × 106 and 2.7 × 104 CFU/g, respectively, demonstrating that large round bales are prone to molding at relatively low moisture concentrations. Maintenance of forage quality and reduction in mold growth were achieved by baling dry (124 g/kg moisture) or wrapping round bales of orchardgrass hay up to 337 g/kg moisture.  相似文献

Seasonal and diurnal changes in water-soluble carbohydrates (WSC) and ethanol-soluble carbohydrates (ESC), as well as in chromatographic profiles of the sugars comprising these categories, were studied in orchardgrass pastures at one location in the Piedmont region of Virginia. Grass from four experimental plots was sampled weekly in the morning and afternoon over an 8-week period (early to late spring). Tissue was air-dried to simulate hay, or frozen to preserve the sugar profiles of fresh pasture. WSC and ESC were assayed colorimetrically. To profile sugars, boiled-water extracts were separated by high-performance liquid chromatography coupled to pulsed amperometric detection. Fructan, glucose, fructose, and sucrose were quantified by high-performance liquid chromatography. All sugar concentrations were highest in the early spring in both fresh and dried tissue. Fructan chain length was highest in early spring as well. Significant diurnal differences were observed for WSC and fructan, on some dates (P < .0001 and P = .024, respectively), and for ESC, glucose, and sucrose averaged over the entire period (P = .0002, .004, and <.0001, respectively). Sucrose was barely detectable in fresh tissue but reached 1.6% to 12% dry matter in dried tissue. These results demonstrate changes in orchardgrass carbohydrates over a season, within a day, and between dried and fresh herbage. Understanding these changes may be helpful in the management of horses with a history of insulin resistance and laminitis.  相似文献

Seasonal and diurnal patterns of sugar accumulation in bermudagrass (Cynodon dactylon) pastures were monitored to evaluate risk factors for pasture-associated laminitis of ponies and horses. Bermudagrass was collected from four plots in the morning and afternoon on a weekly basis, from mid-July until late August. Tissue was air-dried to simulate hay, or frozen to retain the sugar profiles of fresh pasture. Samples were analyzed colorimetrically for total water-soluble and ethanol-soluble carbohydrates, and electrochemically for starch. In addition, sugars were separated and quantified by high-performance liquid chromatography coupled to pulsed amperometric detection. The dominant sugars in extracts were glucose, fructose, and sucrose. Some minor peaks, corresponding to tri- and tetrasaccharides, were also detected in some extracts. Starch increased over time in fresh and dried tissue, and concentrations varied diurnally in fresh, but not in dried tissue (P = .021). Sucrose in dried tissue decreased and then increased, with higher concentrations than in fresh tissue on all sampling dates (P = .024). Glucose and fructose exhibited diurnal variation on one and two dates, respectively (P = .034 and .0028, respectively). These results reveal trends in carbohydrate concentrations and profiles that may help to evaluate the likelihood of equine laminitis outbreaks on bermudagrass.  相似文献
