This is the first report of Alternaria leaf spot disease on coriander (Coriandrum sativum L.) in South Africa. Using the agar plate method, Alternaria alternata was isolated from coriander seed lots together with four other fungal genera, which included Aspergillus, Fusarium, Penicillium and Rhizopus. Standard seed germination tests of coriander seed lots infected with seed-borne mycoflora showed a positive correlation with the number of diseased seedlings (r?=?0.239, p?<?0.01). Pathogenicity tests demonstrated that this seed-borne A. alternata was pathogenic on coriander and symptoms on leaves first appeared as small, dark brown to black, circular lesions (<5 mm diam.) that enlarged and coalesced to form dark brown blotches as time progressed. Leaf spot disease was most severe (64%) on wounded leaves inoculated with A. alternata. Re-isolation of A. alternata from diseased coriander plants satisfied the Koch’s postulates, thus confirming it as the causal agent of Alternaria leaf spot disease. Parsimony analysis based on rpb2 (GenBank Accession No. KT895947), gapdh (KT895949) and tef-1α (KT895945) sequences confirmed identity of the Alternaria isolate, which grouped within the A. alternata clade. Alternaria alternata was shown to be transmitted from infected coriander seed to the developing plants.  相似文献

Phytophthora root rot (PRR) of avocado, caused by Phytophthora cinnamomi, is a significant threat to sustainable production wherever the crop is grown. Resistant rootstocks in combination with phosphite applications are the most effective options for managing this disease. Recently, the mechanisms underpinning PRR resistance have been investigated by the avocado community. Here, biochemical assays and confocal and scanning electron microscopy were used to investigate early defence responses in PRR resistant and ‐susceptible avocado rootstocks. Zoospore germination and subsequent hyphal growth for the pathogen were significantly inhibited on the surface of resistant avocado roots. When penetration occurred in the resistant R0.06 rootstock, callose was deposited in the epidermal cells, parenchyma and cortex of roots. In addition, β‐1,3‐glucanase was released early (6 h post‐inoculation, hpi) in response to the pathogen, followed by a significant increase in catalase by 24 hpi. In contrast, susceptible R0.12 roots responded only with the deposition of lignin and phenolic compounds incapable of impeding pathogen colonization. In this study, PRR resistance was attributed to a timely multilayered response to infection by P. cinnamomi.  相似文献

Damping-off and stem rot disease-causing Sclerotium rolfsii has been reported as a destructive soil-borne pathogen of numerous crops, especially in the tropics and subtropics. Trials were conducted to test the efficacy of biocontrol agents alone or combined with Moringa oleifera leaf extracts for the control of the disease. In the laboratory, PDA was amended with Moringa leaf extract, and mycelial growth of S. rolfsii was measured. In the greenhouse and field, Trichoderma Kd 63, Trichoderma IITA 508 and Bacillus subtilis were evaluated as seed treatments, soil drench or sprinkle, separately or combined with Moringa leaf extracts. Percentage disease incidence, severity and control were recorded. In the laboratory, the higher the extract concentration the less the mycelial growth and no mycelial growth occurred on extract at 15 or 20 g leaves 10 ml−1 water. In the greenhouse, the highest disease control was observed at a Moringa extract concentration of 15 kg leaves 10 l−1 water (w/v). Seed treatments using Trichoderma Kd 63, and soil sprinkle using Trichoderma IITA 508 had a significantly (P = 0.05) higher effect on a disease incidence than Bacillus. Disease severity followed the same pattern. Moringa seed treatment combined with Trichoderma soil sprinkle resulted in significantly more than 94% and 70% disease control in the greenhouse and field, respectively, with significant yield increase in the field. This is the first report of Moringa leaf extract combined with Trichoderma as an integrated control for Sclerotium damping-off and stem rot of cowpea in the field.  相似文献

It is widely accepted that biodiversity loss and poverty are linked problems and that conservation and poverty reduction should be tackled together. However, success with integrated strategies is elusive. There is sharp debate about the social impacts of conservation programs and the success of community-based approaches to conservation. Clear conceptual frameworks are needed if policies in these two areas are to be combined. We review the links between poverty alleviation and biodiversity conservation and present a conceptual typology of these relationships.  相似文献

The role of phenolics in plant tolerance to pathogen infection is well documented. The objective of the present preliminary investigation was to study phenolic metabolites involved in the tolerance or susceptibility of cowpea (Vigna unguiculata Walp.) cultivars to Sclerotium rolfsii Sacc. and to use their presence as a possible screening tool. Total, free acid, ester-bound and cell wall-bound phenolics of 10 cowpea cultivars were quantified. In healthy seedlings, the tolerant cultivars displayed the higher phenol content than the susceptible cultivars. In S. rolfsii infected seedlings, the highest increase was found from 48 h after inoculation. The net effect of inoculation was a 630% increase in total phenolics (soluble and insoluble) in the stem of tolerant cultivars while the total phenolic content increased only by 212% in the stems of susceptible cultivars. Although, no significant difference (P = 0.05) was detected among cultivars, in terms of free acid phenolics, the amount of ester-bound and cell wall-bound phenolics significantly increased, therefore demonstrating a similar trend to the one observed for the total phenolic content. These preliminary results showed that the presence of phenolics before and after S. rolfsii infection may be used as a rapid screening method for detection of tolerance to S. rolfsii damping-off and stem rot of cowpea.  相似文献

Trends in weather variables and concentrations of airborne conidia ofAlternaria solani were monitored in a potato field in South Africa during three potato-growing seasons in 2001 and 2002. Distinct seasonal variation was noted, with a drop in spore numbers during winter. Peaks in spore concentration coincided with periods favorable for spore formation and dispersal; most notable was the effect of interrupted wetting periods. Diurnal periodicity of spore dispersal was also observed, with the peak of spore concentrations between 9h00 and 18h00. Few spores were sampled at night, when wind velocity and temperature are lowest and relative humidity is highest. Increased numbers of spores were sampled during days of harvesting or when other ground-operated farm equipment was used. Geminivirus defective interfering DNAs arise spontaneously in mechanically inoculated test plants, and have previously been found with DNA-B of the bipartite cassava mosaic geminiviruses, but not DNA-A. Reported here for the first time is the cloning and characterization of a naturally occurring truncated form of cassava mosaic geminivirus DNA-A, which at 1525 nt is around half the expected full size. Sequence analysis has shown it to be a defective (df) form of East African cassava mosaic virus (EACMV) DNA-A that has retained its cis elements essential for replication by the helper virus, and it has been termed df DNA-A 15. Phylogenetic comparisons placed the df DNA-A 15 molecule close to mild and severe isolates of EACMV-UG2. Biolistic inoculation of Nicotiana benthamiana with infectious df DNA-A 15 clone and East African cassava mosaic Cameroon virus (EACMCV) resulted in symptom amelioration as compared with EACMCV singly inoculated plants, and there was an accumulation of df DNA-A 15 in systemically infected leaves. In addition, the level of EACMV DNA-B accumulation was reduced in the coinoculated plants compared with those inoculated with EACMCV alone. The cultivation of cowpea (Vigna unguiculata) plays a vital role in the livelihood of many subsistence farmers and rural communities in tropical and subtropical countries. The seeds are prone to fungal infestation and mycotoxin contamination during sub-optimal storage conditions. Fumonisin B1 (FB1), produced byFusarium proliferatum, has been detected in cowpea seeds. Surface-disinfected seeds were imbibed in sterile distilled water amended with FB1 at various concentrations. Percentage germination was determined according to the International Seed Testing Association rules. All the toxin concentrations significantly decreased seed germination and the two highest concentrations — 50 and 100μg ml−1 FB1 — inhibited root and shoot elongation. FB1-treated embryonic tissues evinced compaction of the protoplasm and separation of the plasmalemma from the cell wall. Lipid bodies accumulated, which seemed to be lining the cell wall. This is the first study to demonstrate the phytotoxic effects of FB1 on cowpea seeds.  相似文献

Cowpea seed samples from South Africa and Benin were analyzed for seed mycoflora. Fusariumspecies detected were F. equiseti, F. chlamydosporum, F. graminearum, F. proliferatum, F. sambucinum, F. semitectum, and F. subglutinans. Cowpea seed from South Africa and Benin and F. proliferatum isolates from Benin, inoculated onto maize patty medium, were analyzed for fumonisin production. Samples were extracted with methanol/water and cleaned up on strong anion exchange solid phase extraction cartridges. HPLC with precolumn derivatization using o-phthaldialdehyde was used for the detection and quantification of fumonisins. Cowpea cultivars from South Africa showed the presence of fumonisin B(1) at concentrations ranging between 0.12 and 0.61 microg/g, whereas those from Benin showed no fumonisins. This is believed to be the first report of the natural occurrence of FB(1) on cowpea seed. Fumonisin B(1), B(2), and B(3) were produced by all F. proliferatum isolates. Total fumonisin concentrations were between 0.8 and 25.30 microg/g, and the highest level of FB(1) detected was 16.86 microg/g.  相似文献

Damping-off and stem rot are two types of diseases affecting cowpea (Vigna unguiculata (L.) Walp.) in the Quémé Valley, Benin. Of the fungal species isolated from diseased plants in the field during a 2-year experiment (2001 and 2002),Sclerotium rolfsii Sacc. was found to be solely responsible for these diseases. The disease incidence decreased with increasing distance of the field from the river. Measurement ofS. rolfsii initial inoculum, soil moisture and disease incidence in cowpea field plots revealed a positive correlation among these parameters. The multiple regression analysis showed that the disease incidence increase was 0.4% for one unit increase in soil moisture percent, whereas the disease incidence increase was 19.8% for one unit increase of the density of initial inoculum of the pathogen. This is the first comprehensive study of the effects of environmental factors on the incidence of cowpea damping-off and stem rot caused byS. rolfsii in Benin, and shows that the density of the initial inoculum is the main contributing factor of the disease in the field in the Quémé Valley. http://www.phytoparasitica.org posting Dec. 19, 2004.  相似文献
