Chemical taxonomy based upon the composition of lipids is widely applied to investigate microbial communities and fatty acids have recently been employed to connect soil microbial and faunal food webs as well as to elucidate functional groups at higher trophic levels. The additional use of compound-specific isotopic analysis of 13C/12C ratios in fatty acids allows assessing specific trophic links and belowground carbon fluxes. In this review systematic patterns and processes underlying variations in the composition of fatty acids and their 13C/12C ratio are described. The emphasis is on biomarker fatty acids, their incorporation and modification, effects of pool size, and analytical methods. Further development of the application of fatty acid profiling to soil ecology should include both advances in experimental research and growth of theory. Accordingly, areas in which future experimentation can lead to progress in soil food web analysis are identified. Overall, combining fatty acid biomarker and their isotopic ratios will allow detailed insight into belowground trophic interactions.  相似文献

Stable isotope analysis has been used as a powerful tool in food web studies in terrestrial ecosystems. In addition the occurrence and abundance of fatty acids may serve as indicator for feeding strategies of soil animals. Here we combine both approaches and investigate the fatty acid composition, δ13C values of bulk tissues and individual fatty acids in soil organisms. The fungi Chaetomium globosum and Cladosporium cladosporioides were isotopically labelled by fructose derived from either C3 or C4 plants, and the fungal-feeding nematode Aphelenchoides sp. was reared on C. globosum. Fungi and nematodes were used as diet for the Collembolan Protaphorura fimata. The sugar source was fractionated differently by fungal lipid metabolism in a species-specific manner that points to a sensitivity of physiological processing to the non-random distribution of 13C/12C isotopes in the molecule. As a general trend stearic acid (18:0) was depleted in 13C compared to the precursor palmitic acid (16:0), whereas its desaturation to oleic acid (18:1 ω9) favoured the 13C-rich substrate.Fatty acid profiles of P. fimata varied due to food source, indicating incorporation of dietary fatty acids into Collembolan tissue. Individuals feeding on fungi had lower amounts in C20 fatty acids, with monoenoic C20 forms not present. This pattern likely separates primary consumers (fungivores) from predators (nematode feeders). The isotopic discrimination in 13C for bulk Collembola ranged between −2.6 and 1.4‰ and was dependent on fungal species and C3/C4 system, suggesting differences at metabolic branch points and/or isotope discrimination of enzymes. Comparison of δ13C values in individual fatty acids between consumer and diet generally showed depletion (i.e. de novo synthesis) or no changes (i.e. dietary routing), but the fractionation was not uniform and affected by the type of ingested food. Fatty acid carbon isotopes were more variable than those of bulk tissues, likely due to both the distrimination by enzymes and the different lipid origin (i.e. neutral or polar fraction).  相似文献

Nitrate determination in leachates from farmyard manure by UV absorption A simple, sufficiently sensitive (< 1 ppm NO3?-N) method for nitrate determination in leachates from farmyard manure is described, suitable for routine analysis. The well known determination by UV absorption is preceded by a precipitation of organic matter with ZnSO4BO7. By this, average NO3?-N contents of 48.7 ppm were detected in leachates from cattle manure heaps (n = 54).  相似文献

We investigated microbial biomass, fungal biomass and microbial community structure at three altitudes (1000, 2000 and 3000 m) and in two soil layers [L/F layer (Layer I) and underlying H/Ah layer (Layer II)] of tropical mountain rain forests in southern Ecuador. Basal respiration, microbial biomass and concentration of ergosterol generally declined from Layer I to Layer II and peaked at 2000 m. Compared to temperate forest ecosystems microbial biomass and ergosterol concentrations were generally low. Patterns in phospholipid fatty acids indicated that the composition of microbial communities markedly changed from Layer I to Layer II. These differences between layers decreased with increasing altitude. The concentration of the arbuscular mycorrhizal fungal marker PLFA 16:1ω5c decreased with altitude in Layer I but increased in Layer II. The fungal-to-bacterial ratio increased with altitude and was higher in Layer I than in Layer II. Presumably, low microbial biomass in soils of tropical forest ecosystems is due to high temperature associated with high respiration but also low litter quality, with the latter declining with altitude. These conclusions are supported by the fact that at higher altitude the microbial community changed from a bacterial-dominated to a fungal-dominated system. CCA showed that microbial biomass correlated closely with density of a number of putatively bacterial feeding testate amoeba species including Corythion dubium Taranek, 1871, Euglypha cristata Leidy, 1879, Trigonopyxis arcula Penard, 1912, Tracheleuglypha dentata Deflandre, 1928 and Trinema lineare Penard, 1890. Ergosterol concentrations, but not the PLFA 18:2ω6c, strongly correlated with the putatively fungal feeding species Phryganella acropodia (Hertwig, Lesser, 1874) Hopkinson, 1909. Generally, parallel to microbial biomass and ergosterol concentrations the density of testate amoebae peaked at 2000 m. However, compared to microbial parameters changes in testate amoebae communities between two layers were less pronounced. The data suggest that density and community structure of testate amoebae are driven by the availability of food resources (bacteria and fungi) which at high altitude decrease with increasing moisture and decreasing pH.  相似文献

Eighty-four olive accessions obtained from the National Conservatory of Boughrara-Sfax (Tunisia), previously evaluated for morphological traits, were analysed with 47 random amplified polymorphic DNA (RAPD) markers. They were compared with other olive genotypes originated from Eastern or Western Mediterranean. The highest and lowest similarities between genotypes, estimated by simple matching algorithm, were 0.98 and 0.40, respectively. A dendrogram based on Ward's method and a factorial correspondence analysis (FCA) showed that most of Tunisian accessions are closely related to olive genotypes originating from the Eastern Mediterranean and some are clustering with genotypes originated from the Western Mediterranean. These findings suggested multiple and complex origin of Tunisian olive. A comparative study between a previous morphological analysis and current RAPD assay was carried out and discussed.  相似文献

Tunisia, other countries in northwestern Africa, and the Iberian Peninsula, represent a center of diversity of Daucus. The genus traditionally has included about 20–25 species worldwide, but a recent molecular study redefined and expanded Daucus to include representatives from nine other genera. By this classification, Daucus now contains about 40 species, with some of them having winged fruits in addition to its traditionally recognized spiny fruits. The taxonomy of Daucus in Tunisia has recently been studied with morphological data, concluding that D. carota subsp. capillifolius is a subspecies that co-occurs and crosses with subsp. carota. The present study extends these findings with additional data from Genotyping-by-Sequencing (GBS) from 33 Tunisian accessions of D. carota (93 individuals with two to three replicates per accession), including the first collections of subsp. gummifer from Tunisia. Placed in the context of additional GBS data, D. carota subsp. gummifer has separate origins from other collections of subsp. carota in Tunisia and/or immediately adjacent areas to the north in Italy or its surrounding islands. Our results add support to the utility of large SNP datasets for species-level phylogenetic studies in Daucus.  相似文献

Most of the studies on organic acids and sugars in citrus were performed during fruit maturation, and less is known before this stage of development. The aim of our study was to investigate acids and sugars in lemon, lime, and orange from fruit-set toward development. We chose to compare organic acid and sugar accumulation among acidic and acidless varieties within three species. We estimated the acidity by titrimetry and quantified the concentrations of seven organic acids and three sugars by reverse HPLC. During the first 50 days of development, quinic acid was the major organic acid whatever the variety. Afterward, citric acid predominated in acidic varieties, while in acidless, malic acid exceeded it. Fructose substituted citric acid in acidless and could be synthesized either from citric acid or directly from glucose. Our results provided the first complete report on sugar and organic acid accumulation during the early stages of fruit development in several citrus varieties.  相似文献
