Changes in gene regulation are thought to have contributed to the evolution of human development. However, in vivo evidence for uniquely human developmental regulatory function has remained elusive. In transgenic mice, a conserved noncoding sequence (HACNS1) that evolved extremely rapidly in humans acted as an enhancer of gene expression that has gained a strong limb expression domain relative to the orthologous elements from chimpanzee and rhesus macaque. This gain of function was consistent across two developmental stages in the mouse and included the presumptive anterior wrist and proximal thumb. In vivo analyses with synthetic enhancers, in which human-specific substitutions were introduced into the chimpanzee enhancer sequence or reverted in the human enhancer to the ancestral state, indicated that 13 substitutions clustered in an 81-base pair module otherwise highly constrained among terrestrial vertebrates were sufficient to confer the human-specific limb expression domain. 相似文献

Vanilla is naturally distributed in Mexico and parts of Central America and the history of origin of cultivated vanilla suggests
that the entire stock outside Mexico may be from a single genetic source. In the present study, RAPD polymorphism was used
to estimate the level of genetic diversity and interrelationships among different collections of Vanilla planifolia Andr., and few related species, including both leafy and leafless types such as V. tahitensis J.W.Moore, V. andamanica, Rolfe, V. pilifera Holtt., and V. aphylla Blume. Studies revealed that there are very limited variation within collections of V. planifolia, indicative of its narrow genetic base, and of the related species we tested, V. tahitensis is nearest to V. planifolia. The species studied are diverse and have a similarity ranging from 1.2 to 57.3 %. Of the sampled taxa, V. andamanica is the most divergent and there is also reasonable variability within its collections, indicating the possibility of natural
seed set. A total of 82 polymorphic bands expressed in the RAPD profiles were used to generate a genetic distance matrix,
which was then used in cluster analysis. Specific groupings were revealed by the cluster analysis whereby the leafless forms
(V. aphylla, V. pilifera and the new species) and V. andamanica formed separate clusters. This is the first report of species interrelationship studies, including both cultivated and wild

The oomycete Phytophthora colocasiae that causes taro leaf blight is the most devastating disease of taro and is widely distributed worldwide. Molecular and phenotypic techniques were employed for assessing and exploiting the genetic variability among four populations of P. colocasiae obtained from a fine spatial scale (multiple leaf blight lesions on single taro leaf). Phenotypic characters such as virulence, morphology and mating type showed no variation. ITS characterization revealed detectable polymorphism among isolates of P. colocasiae. The mean number of haplotypes (H), haplotype diversity (HD), nucleotide diversity (π), and nucleotide substitution rate (θ) among analyzed sequences were 6.75, 1.00, 0.069, and 0.088 respectively. High levels of inter and intra specific variation were detected by random amplified polymorphic DNA (RAPD) assays. Moderate genetic diversity (H?=?0.2651) was observed among populations of P. colocasiae. Analysis of molecular variance (AMOVA) confirmed that most of the genetic variability was confined to within a population (63.54 %). The coefficient of genetic differentiation among populations (G ST ) was 0.2007 and estimates of gene flow (Nm) among populations was 1.991 migrants per generation. Cluster analysis using UPGMA revealed that individuals from the same population failed to cluster in one distinct group. The results of the study reveal considerable genetic diversity among and within populations of P. colocasiae obtained from fine spatial scale. The possible mechanisms and implications of this genetic variation are discussed. 相似文献

A cobalt-mediated decrease in ethylene production reduced the inhibition of nodulation by nitrate in Vigna radiata (mungbean). Nitrate increased the ethylene production in 5-day-old seedlings, while it caused a reduction in the nodulation
status (nodule number and nodule weight) and nodule efficiency (acetylene reduction activity) in mungbean plants. The application
of cobalt chloride inhibited nitrate-affected ethylene production and also decreased the inhibitory effect of nitrate on nodulation.
The effect of cobalt was most marked on nodule number.

The phthalimide fungicide captan has been widely used to control plant pathogenic fungi. A strain of Bacillus circulans utilized the fungicide captan as sole source of carbon and energy. The organism degraded captan by a pathway involving its initial hydrolysis to yield cis-1,2,3,6-tetrahydrophthalimide, a compound without fungicidal activity. The formation of this compound was confirmed by HPLC, IR, NMR, and mass spectral analysis. The results also revealed that cis-1,2,3,6-tetrahydrophthalimide was further degraded to o-phthalic acid by a protocatechuate pathway. These findings indicated that there was a complete mineralization of fungicide captan by B. circulans. 相似文献

The intercellular colonization of rice roots by Azorhizobium caulinodans and other diazotrophic bacteria has been studied using strains marked with the lacZ reporter gene. A. caulinodans were able to enter the roots of rice at emerging lateral roots (lateral root cracks) by crack entry and this was observed by light microscopy. After colonization of lateral roots, bacteria moved into intercellular space within the cortical cell layer of roots. Naringenin at 1×10-5 and 5×10-5 M concentration significantly enhanced root colonization. The role of nodABC and regulatory nodD genes was also studied; lateral root crack (LRC) colonization of rice was shown to be Nod factor and NodD independent. Lateral root crack colonization of rice was also observed with similar frequency following inoculation with Azospirillum brasilense and the colonization by A. brasilense was stimulated by naringenin and other flavonoid molecules. 相似文献

Andrographis paniculata [(Burm. f.) Wall. ex Nees, family Acanthaceae], commonly known as kalmegh is used both in Ayurvedic and Unani system of medicines for a number of ailments related to digestion, hepatoprotection, hypoglycaemic and as anti-bacterial, analgesic, anti-inflammatory, vermicidal and antipyretic. Although kalmegh is extensively used in Indian Systems of Medicine, but it is not yet cultivated on a large-scale. The supply of herb is met from the wild growing populations which is now under constrain due to over-collection. Considering its value as a drug raw material, genetic variability of A. paniculata was augmented from different geographical regions of India by explorations and was conserved at the National Gene Bank, NBPGR, New Delhi. The therapeutic activity of the herb has been attributed to andrographolide, a bicyclic diterpenoid lactone present in the whole herb. Thirty germplasm collections of A. paniculata were evaluated for bioactive compound andrographolide to find out superior genotype for high drug yield. The andrographolide content in the germplasm was quantitatively analyzed by high performance thin-layer chromatography (HPTLC) at 232 nm. Results indicated wide variability among thirty accessions of A. paniculata for andrographolide content which ranged from 0.72 to 2.99 % on dry weight basis. DIVA-GIS analysis of A. paniculata germplasm for andrographolide content showed that diverse accessions of A. paniculata exist in three Indian states of Orissa, Tamil Nadu and Kerala. Promising accessions having high andrographolide content were identified; IC520361 (2.99 %), IC520395 (2.86 %), IC399125 (2.81 %), IC369404 (2.77 %) and IC520394 (2.61 %). Due to the economic importance of the drug, these promising collections can be exploited for large-scale cultivation for their pharmacologically important constituents mainly andrographolide content and also can serve as potential source for breeding and improvement of cultivars for obtaining higher drug yield. 相似文献
