ABSTRACT The relative contribution of migration of Rhizoctonia solani anastomosis group 3 (AG-3) on infested potato seed tubers originating from production areas in Canada, Maine, and Wisconsin (source population) to the genetic diversity and structure of populations of R. solani AG-3 in North Carolina (NC) soil (recipient population) was examined. The frequency of alleles detected by multilocus polymerase chain reaction-restriction fragment length polymorphisms, heterozygosity at individual loci, and gametic phase disequilibrium between all pairs of loci were determined for subpopulations of R. solani AG-3 from eight sources of potato seed tubers and from five soils in NC. Analysis of molecular variation revealed little variation between seed source and NC recipient soil populations or between subpopulations within each region. Analysis of population data with a Bayesian-based statistical method previously developed for detecting migration in human populations suggested that six multilocus genotypes from the NC soil population had a statistically significant probability of being migrants from the northern source population. The one-way (unidirectional) migration of genotypes of R. solani AG-3 into NC on infested potato seed tubers from Canada, Maine, and Wisconsin provides a plausible explanation for the lack of genetic subdivision (differentiation) between populations of the pathogen in NC soils or between the northern source and the NC recipient soil populations.  相似文献

Mutational inactivation of the retinoblastoma susceptibility (RB) gene has been proposed as a crucial step in the formation of retinoblastoma and other types of human cancer. This hypothesis was tested by introducing, via retroviral-mediated gene transfer, a cloned RB gene into retinoblastoma or osteosarcoma cells that had inactivated endogenous RB genes. Expression of the exogenous RB gene affected cell morphology, growth rate, soft agar colony formation, and tumorigenicity in nude mice. This demonstration of suppression of the neoplastic phenotype by a single gene provides direct evidence for an essential role of the RB gene in tumorigenesis.  相似文献

Transgenic peanut lines expressing oxalate oxidase, a novel enzyme to peanut, were evaluated for resistance to Sclerotinia blight in naturally infested fields over a 5-year period. Area under the disease progress curve (AUDPC) for transgenic lines in single rows planted with seed from single-plant selections averaged 78, 83, and 90% lower than nontransgenic parents in 2004, 2005, and 2006, respectively. In addition, AUDPC in 14 transgenic lines planted with bulked seed in two-row plots averaged 81% lower compared with nontransgenic parents in 2005 and 86% lower in 16 transgenic lines in 2006. Six transgenic lines yielded 488 to 1,260 kg/ha greater than nontransgenic parents in 2005, and 10 lines yielded 537 to 2,490 kg/ha greater in 2006. Fluazinam (0.58 kg a.i./ha) fungicide sprays in 2008 and 2009 reduced AUDPC in transgenic and nontransgenic lines but AUDPC was lowest in transgenic lines. Without fluazinam, yields of transgenic lines averaged 1,133 to 1,578 kg/ha greater than nontransgenic lines in 2008 and 1,670 to 2,755 kg/ha greater in 2009. These results demonstrated that the insertion of barley oxalate oxidase in peanut conveyed a high level of resistance to Sclerotinia blight, and negated the need for costly fungicide sprays.  相似文献

The extent to which terrestrial ecosystems can sequester carbon to mitigate climate change is a matter of debate. The stimulation of arbuscular mycorrhizal fungi (AMF) by elevated atmospheric carbon dioxide (CO(2)) has been assumed to be a major mechanism facilitating soil carbon sequestration by increasing carbon inputs to soil and by protecting organic carbon from decomposition via aggregation. We present evidence from four independent microcosm and field experiments demonstrating that CO(2) enhancement of AMF results in considerable soil carbon losses. Our findings challenge the assumption that AMF protect against degradation of organic carbon in soil and raise questions about the current prediction of terrestrial ecosystem carbon balance under future climate-change scenarios.  相似文献

Plant diversity can have a profound impact on disease dynamics, with important applications for enhancing sustainability. Disease is often reduced by intercropping, but variability can be high. This study investigated integration of several management approaches to stabilize this variability for early leaf spot (ELS) and late leaf spot (LLS) of groundnut, over seven seasons in three phases. In phase 1, monocrops and alternating row and strip intercrops with maize were artificially inoculated with ELS in an area with little groundnut production. Reductions in AUDPC of 37–73% in strip treatments compared to monocrops prompted testing of the efficacy of intercropping in intensive production areas for phases 2 and 3. Additional treatments included cotton strip intercrops, and integration of intercropping with reduced fungicide treatments and partial resistance to leaf spots. In phase 2, the use of cotton strip intercrops lowered natural ELS epidemics by 25–41% (AUDPC) through delayed disease onset, but maize had inconsistent effects. Intercropping was not effective against LLS, which dominated in phase 3. Reduced fungicide regimes and partial resistance lowered disease, and in one case interacted with intercropping to enhance disease suppression. Groundnut yields generally were inversely proportional to disease levels and not significantly reduced by intercropping. Separate studies to determine maize impacts on ELS infection implicated disruption of dispersal as the mechanism of disease reduction. This work demonstrates that intercropping may be most effective where low levels of ELS are present, using strip patterns with cotton, and combined with other tools such as resistance and reduced fungicide application.  相似文献

ABSTRACT Amendment of peat-based potting media with Al(2)(SO(4))(3) suppresses damping-off of Vinca (Catharanthus roseus) caused by Phytophthora parasitica. The species of aluminum (Al) responsible for disease suppression have not been identified. The objective of this study was to determine the effects of amount and pH of Al(2)(SO(4))(3) amendment solutions on survival of P. parasitica. In separate experiments, peat was amended with Al(2)(SO(4))(3) solutions adjusted to pH 4 or 6 at either 0.0158 or 0.0079 g of Al per gram of peat. Amended peat was placed in Büchner funnels maintained at -2.5 kPa matric potential. Peat was infested with P. parasitica by placing zero, two, or five colonized Vinca leaf disks in each funnel, and 15 Vinca seeds were placed in each funnel. After 24 h, the matric potential was brought to 0 kPa to induce zoospore release and returned to -2.5 kPa after 24 h. Pathogen populations and stand counts were assessed after 2-week incubation. Al amendment solutions at both pH 4 and 6 reduced pathogen populations at 0.0158 g of Al per gram of peat. Solutions at pH 4 reduced pathogen populations by more than 90% at both inoculum levels; amendment solutions at pH 6 reduced populations by 95% at the low inoculum level and 65% at the high inoculum level. The prevalence of Al(OH)(2)(+) in peat amended with Al(2)(SO(4))(3) solution at pH 6 suggests that ions other than Al(3+) may be responsible for pathogen suppression. Based on the difference in chemical conditions of Al-amended peat and suppressive mineral soils, the mechanism of Al-mediated suppression of plant pathogens is speculated to be different in the two systems. Peat containing Al-peat complexes was chemically suppressive to P. parasitica and may confer Al-mediated suppression of plant pathogens with a nonphytotoxic form of Al.  相似文献
