A new rot caused by a binucleate Rhizoctonia sp. affecting the tuberous root cortex of the domesticated yacon ( Smallanthus sonchifolius ) has been observed in Brazil. Isolates of a binucleate Rhizoctonia sp. were collected from roots with rot symptoms and characterized by the number of nuclei per cell, hyphal anastomosis, RAPD molecular markers, ITS-5·8S rDNA sequence and pathogenicity tests. All isolates had a mean of 1·9–2·2 nuclei per cell and anastomosed with the binucleate Rhizoctonia sp. AG G-tester strain. RAPD analysis was carried out between 11 isolates recovered from yacon and 11 AG (A, Ba, Bb, Bo, C, D, F, G, O, P, Q) standard testers of binucleate Rhizoctonia sp. Genetic similarities of 94·8–100% were observed among isolates of the binucleate Rhizoctonia sp. from yacon and all isolates were genetically more closely related to the AG G tester than other strains according to upgma analysis using RAPD markers. Homologies of complete ITS nucleotide sequences were 100% between binucleate isolates of Rhizoctonia sp. from yacon and the AG G tester. BACKGROUND: Dengue fever is a severe public health problem for several countries. In order to find effective larvicides to aid control programs, the structure‐activity relationships of eugenol derivatives against Aedes aegypti (Diptera: Culicidae) larvae were evaluated. Additionally, the composition and larvicidal activity of Syzygium aromaticum essential oil was assessed. RESULTS: Four compounds representing 99.05% of S. aromaticum essential oil have been identified. The essential oil was active against Ae. aegypti larvae (LC50 = 62.3 and 77.0 ppm, field‐collected and Rockefeller larvae respectively). The larvicidal activity of eugenol, the major compound of the essential oil, was further evaluated (LC50 = 93.3 and 71.9 ppm, field‐collected and Rockefeller larvae respectively). The larvicidal activity and structure‐activity relationships of synthetic derivatives of eugenol were also assessed. The larvicidal activity of the derivatives varied between 62.3 and 1614.9 ppm. Oxidation of eugenol allylic bond to a primary alcohol and removal of the phenolic proton resulted in decreased potency. However, oxidation of the same double bond in 1‐benzoate‐2‐methoxy‐4‐(2‐propen‐1‐yl)‐phenol resulted in increased potency. CONCLUSION: Structural characteristics were identified that may contribute to the understanding of the larvicidal activity of phenylpropanoids. The Azospirillum brasilense genomic region containing the glnBA genes was previously cloned in a broad-host range plasmid named pAB441. Previous results from our laboratory have shown that A. brasilense spontaneous mutants resistant to ethylenediamine carrying this plasmid had no nitrogenase activity. The 19 kb insert of this plasmid was fully sequenced and no other gene potentially involved with nitrogen fixation was identified, nor any gene potentially involved in the regulation of nif genes expression. Apart from glnBA, this region contains several genes involved with different functions: assisting protein folding and degradation, DNA organization, electron transport and energy conversion and conserved hypothetical proteins of unknown function. Since previous data on the literature showed that mutation of clpX gene of A. brasilense resulted in higher nitrogenase activity, we hypothesize that higher expression of the clpX gene from the plasmid pAB441 may be involved with Nif? phenotype of the A. brasilense mutant strains. 相似文献

Rhizoctonia solani AG-1 IA causes leaf blight on soybean and rice. Despite the fact that R. solani AG-1 IA is a major pathogen affecting soybean and rice in Brazil and elsewhere in the world, little information is available on its genetic diversity and evolution. This study was an attempt to reveal the origin, and the patterns of movement and amplification of epidemiologically significant genotypes of R. solani AG-1 IA from soybean and rice in Brazil. For inferring intraspecific evolution of R. solani AG-1 IA sampled from soybean and rice, networks of ITS-5.8S rDNA sequencing haplotypes were built using the statistical parsimony algorithm from Clement et al. (2000) Molecular Ecology 9: 1657–1660. Higher haplotype diversity (Nei M 1987, Molecular Evolutionary Genetics Columbia University Press, New york: 512p.) was observed for the Brazilian soybean sample of R. solani AG-1 IA (0.827) in comparison with the rest of the world sample (0.431). Within the south-central American clade (3-2), four haplotypes of R. solani AG-1 IA from Mato Grosso, one from Tocantins, one from Maranhão, and one from Cuba occupied the tips of the network, indicating recent origin. The putative ancestral haplotypes had probably originated either from Mato Grosso or Maranhão States. While 16 distinct haplotypes were found in a sample of 32 soybean isolates of the pathogen, the entire rice sample (n=20) was represented by a single haplotype (haplotype 5), with a worldwide distribution. The results from nested-cladistic analysis indicated restricted gene flow with isolation by distance (or restricted dispersal by distance in nonsexual species) for the south-central American clade (3-2), mainly composed by soybean haplotypes. 相似文献

The aim of the present study was to investigate the presence of white spot syndrome virus (WSSV) in the Paraíba River, Brazil. Eight sampling sites were established on the bank of the river near water intake areas for the farming of Litopenaeus vannamei. Ten specimens of the shrimp Palaemon pandaliformis and the gastropods Pomacea lineata and Melanoides tuberculatus were collected at each site. Eighty‐one gill fragments from P. pandaliformis, 40 whole individuals of M. tuberculatus and 26 muscle fragments from P. lineata were collected. All samples were stored in microcentrifuge tubes with 95% ethanol (1:10; v:v). Tests were performed at the Potiporã Molecular Analysis Laboratory (state of Rio Grande do Norte, Brazil) for the detection of WSSV using Loop Mediated Isothermal Amplification with the aid of the LAMP WSSV kit (Concepto Azul, Ecuador). Twenty‐nine per cent of P. pandaliformis, 48% of M. tuberculatus and 8% of P. lineata tested positive. The findings demonstrate that WSSV is present in wild crustaceans and mollusks, which may serve as vectors and/or reservoirs of the virus, thereby posing a potential risk to local shrimp farming. The chemical composition, the fatty acids profile, and conjugated linoleic acids content in Longissimus muscle (LM) of steers have been determined. For such, 18 steers (6, Nellore, NEL) and their Simmental (6, NSI), and Santa Gertrudes (6, NSG) crossbreds finished in pasture system with Brachiaria brizantha cv. marandu for about 3 months with approximate weight at slaughter of 480 kg at average age approximate of 25 months. The lipid content increased in the following order influenced by genetic groups: Nellore, F1 Nellore × Simmental and F1 Nellore × Santa Gertrudes crossbreds. The lipid content increased while moisture, ash and protein contents decreased. The content of saturated fatty acids (SFA) was affected by genetic groups. The conjugated linoleic acids contents (CLA) in fat were similar in the genetic groups, but the quantity of CLA concentrations in muscle lipids of steers with larger total lipid was higher. Rhizoctonia solani causes pre- and post-emergence damping-off, root and hypocotyl rot and foliar blight in soybean. Foliar blight has resulted in yield losses of 31–60% in north and northeast Brazil. The aim of this study was to characterize isolates of R. solani associated with soybean in Brazil. Among 73 Rhizoctonia isolates examined, six were binucleate and 67 were multinucleate. The multinucleate iso1ates were characterized according to hyphal anastomosis reaction, mycelial growth rate, thiamine requirement, sclerotia production, and RAPD molecular markers. Four isolates that caused hypocotyl rot belonged to AG-4 and using RAPD analysis they grouped together with the HGI subgroup. Another isolate that caused root and hypocotyl rots was thiamine auxotrophic, grew at 35°C, and belonged to AG-2-2 IIIB. All 62 isolates that caused foliar blight belonged to AG-1 IA. RAPD analysis of R. solani AG-1 IA soybean isolates showed high genetic similarity to a tester strain of AG-1 IA, confirming their classification. The teleomorph of R. solani, Thanatephorus cucumeris was produced in vitro by one AG-1 IA isolate from soybean. The AG-4 and AG-2-2 IIIB isolates caused damping-off and root and hypocotyl rots of soybean seedlings cv. FT-Cristalina, under greenhouse conditions. The AG-2-2 IIIB isolate caused large lesions on the cortex tissue, that was distinct from the symptoms caused by AG-4 isolates. The AG-1 IA isolates caused foliar blight in adult soybean plants cv. In assessing a fish stock, indices based on catch per unit effort (CPUE) are frequently used. Estimates of three indices of catch per unit effort were compared here (CPUE1, CPUE2 and CPUE3), considering the fitting of two models: (i) a bivariate geostatistical model for catch and effort; (ii) a bivariate model where catch and effort were considered spatially independent. For comparing the estimates of the three indices after the fitting of the two models, catch and effort data were simulated in different scenarios. The simulation study showed that, in general, the estimates of CPUE1 expressed by the ratio of the means of catch and effort, present better results for different scenarios and that the estimates from (i) are better than (ii), mainly when there is a correlation between catch and effort and an additional spatial correlation. 相似文献
