Adsorbed phosphate in soils can be chemically extracted; however, this process is both time‐consuming and not cost‐effective if large numbers of samples have to be analysed. Indirect assessment of adsorbed phosphate by pedotransfer functions (PTFs) can help optimize fertilizer strategies. This study aimed to evaluate the spatial variability of adsorbed phosphate (Pads), iron oxides and magnetic susceptibility (MS) in oxisols and to calibrate PTFs to predict Pads. A total of 308 soil samples were collected from Hapludox and Eutrudox soils formed from sandstone in Brazil. The contents of clay (196–607 g/kg), iron oxides (40–165 g/kg), MS (1.2–29 × 10?6 m3/kg) and Pads (327–842 mg/kg) were in the range of typical values for these highly weathered soils. This study showed that the attributes studied were spatially dependent. Geomorphic surfaces enabled understanding of spatial variability and helped to develop a more efficient sampling scheme to calibrate PTFs. Moreover, the adsorbed phosphate in these oxisols could be predicted by a PTF using iron oxides and MS as predictors. The MS attribute enabled the most accurate prediction (concordance coefficient = 0.95, root‐mean‐square error = 46 mg/kg and relative improvement in root‐mean‐square error = ?4.12) of spatial variability through PTF compared to other predictors.  相似文献

Amplification of the 16S rRNA gene from a blood sample obtained from a dog in southeastern Brazil was used to confirm a naturally acquired Ehrlichia (E.) canis infection. Following isolation and culturing of the new bacterial strain called Uberlândia, partial sequences of the dsb and p28 genes were obtained. The dsb partial sequence of the novel strain was 100% similar to dsb gene sequences of E. canis obtained from different geographic areas around the world. Conversely, the p28 partial sequence for the E. canis Uberlândia strain differed at several nucleotides from other sequences available in GenBank. To confirm the antigenic profile of the Uberlândia strain, an indirect immunofluorescence assay against E. canis antigens was performed using dog sera collected from two different areas in Brazil (Uberlândia and São Paulo). The results suggest that both antigens were able to identify animals seropositive for E. canis in Brazil since these Brazilian strains appear to be highly conserved.  相似文献

The dynamics of catecholamine (CA)-synthesis enzymes have been poorly studied in fish. Tyrosine hydroxylase (TH), the rate-limiting enzyme of CA synthesis has been only studied inin vitro conditions. In the present report thein vivo CA synthesis and the CA metabolism were studied in different regions of the forebrain of the rainbow trout. Levels of norepinephrine (NE), dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC) and the rate of accumulation of 3,4-dihydroxyphenylalanine (DOPA) were determined by HPLC following a treatment with hydroxybenzylhydrazine (NSD), a potential inhibitor of DOPA decarboxylase. Kinetics of the accumulation of DOPA and of the decline of DOPAC were in agreement with those found in rat, evidencing that the accumulation of DOPA following NSD can be used in trout to quantify thein vivo enzymatic activity of tyrosine hydroxylase. Experiments using treatment with NSD or with methyl-p-tyrosine reached a same conclusion: the DA neuronal activity in trout is much higher than NE neuronal activity. However, the hypothalamus had high DA levelsvs. lowin vitro andin vivo TH activities and exhibited a low CA turnover.  相似文献

IAA and GA3 had increased the stem thickness by increasing that of cortex, phloem fibers and xylem. Each had increased straw and fiber yield/plant, but the effect was more pronounced with GA3 than IAA. The percentage of long fibers, strength and fineness increased significantly with GA3. Until recently, Toxoplasma gondii was considered clonal with very little genetic variability. Recent studies indicate that T. gondii isolates from Brazil are genetically and biologically different from T. gondii isolates from USA and Europe. In the present study, we retyped 151 free range chicken isolates from Brazil including 117 newly isolated samples from 11 geographically areas (Alagoas, Bahia, Ceará, Maranh?o, Paraná, Pernambuco, Rio de Janeiro, Rio Grande do Norte, S?o Paulo, Sergipe, and Rondonia) and 34 previously reported isolates from the very north (Pará) and the very south (Rio Grande do Sul). Ten PCR-RFLP markers including SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico were used to genotype all isolates. Overall analysis of 151 T. gondii isolates revealed 58 genotypes. Half (29/58) of these genotypes had single isolate and the other half of the genotypes were characterized with two or more isolates. Only 1 of 151 isolates was clonal Type I strain and 5 were clonal Type III strains. Two isolates had mixed infections. Clonal Type II strain was absent. One strain was Type II at all loci, except BTUB. The results confirm high genetic diversity of T. gondii isolates from Brazil.  相似文献

Corynebacterium pseudotuberculosis is the etiologic agent of caseous lymphadenitis, a disease that affects goats and sheep, and can cause severe economic losses. In this study, four different antigenic extracts were obtained from the attenuated strain T1, which was isolated in the state of Bahia (Brazil). Forty-four Canindé breed goats were divided in five groups, each receiving a different antigen solution and saline buffer as a control. The humoral response was monitored through the identification of specific IgG by indirect ELISA and Western Blotting, and the production of IFN-gamma was followed in order to observe the activation of cellular response. After twelve weeks of antigen inoculation, the animals were challenged with 2 x 10(5)CFU of a wild strain, also isolated in Bahia, and necropsy was performed on all animals twelve weeks afterwards. It was observed that the attenuated bacteria gave a protection of 33.3%, in addition to the weak humoral response elicited. Animals inoculated with secreted antigen associated with Freund's incomplete adjuvant and oligodeoxynucleotide containing unmethylated CpG dinucleotides (CpG ODN) showed a strong humoral response, but this inoculation could not prevent the spread of challenge bacteria in the majority of animals. These results demonstrate the immunogenic potential of the attenuated T1 strain in the development of a vaccine against caseous lymphadenitis in goats.  相似文献

The Brazilian Amazon is endemic for malaria and natural infections by Plasmodium spp. have been detected in Neotropical primates. Despite the diversity of primate species in the region, studies on infections by these agents are limited. The aim of the present study was to investigate the frequency of infection by Plasmodium vivax and P. falciparum in free‐born primates that were kept in captivity, in the western Amazon, Brazil. Blood samples were collected from 98 Neotropical primates. Detection of P. vivax and P. falciparum DNA was performed using a semi‐nested PCR, and the amplified products were sequenced. Plasmodium spp. DNA was detected in 6.12% (6/98) of the primates. P. vivax, and P. falciparum DNA was detected in 2.04% (2/98) and 4.08% (4/98) of these mammals, respectively. Sequencing and phylogenetic analysis confirmed the results obtained from the semi‐nested PCR. The presence of infected non‐human primates (NHP) can be auxiliary in the maintenance of P. falciparum and P. vivax and may have implications for the malaria surveillance and control in the Brazilian Amazon. It is necessary to structure an efficient surveillance system for the aetiological agents of malaria that infect NHP and humans to reduce the risk of Plasmodium spp. introduction into new areas, to protect all susceptible species.  相似文献

The biological, serological and genomic diversity of three Citrus tristeza virus (CTV) isolates from various geographical regions was studied: isolate P1 from lemon cv. 'Meyer' in a field near Marrakech (MA) in 1983, and isolates P2 and R1 detected in imported Spanish clementine germplasm by the Moroccan NPPO in 1998 and 2000. P1 induced severe vein clearing on Mexican lime and grapefruit, mild stem pitting on Mexican lime and moderate stem pitting on grapefruit. P2 and R1 only induced mild vein clearing on Mexican lime and caused no stem pitting or other symptoms on indicator plants used as controls. Only isolate P1 reacted with monoclonal antibody MCA-13, whereas all isolates reacted positively with the 3DF1+3CA5 mixture. The Moroccan clones P1–3 and P1–5, and all other severe isolates obtained from GenBank, showed a phenylalanine at amino acid position 124 of their coat protein sequences. This epitope confers MCA13 reactivity. The Spanish clones had tyrosine instead at this position. The deduced amino-acid sequence of coat protein of P1 clones clusters close to severe strains CB3–104 and FL7, respectively from Brazil and USA (Florida) (Group 5), whereas the sequences from P2 and R1 cluster close to typical strains from Portugal 25–120 and USA (Florida) T30 (Group M). The three techniques for distinguishing CTV isolates were clearly correlated.  相似文献

Levels of catecholamines and tyrosine hydroxylase (TH) activity were measured in brain homogenates from female rainbow trout. In triploid fish or in diploid fish in ovarian recrudescence, the patterns of catecholamine content expressed as a function of in vitro TH activity vary in different areas of the brain. Km for the pterin cofactor is lower in the telencephalon than in the hypothalamus. Dopamine (DA) and 2-hydroxyestradiol (20HE2) inhibit TH activity (by competitive and non-competitive interaction respectively). The K1 for DA were different in the telencephalon and the hypothalamus and this could explain the different patterns of catecholamine levels and TH activity for these two structures. 20HE2 inhibits TH activity in vitro; a catechol moiety is required since estradiol (E2) is notinhibitory. However, the exact mechanism of inhibition remains unclear. The rapid effect of 20HE2 cannot explain the previously reported activation of catecholamine synthesis by E2 in vivo.
Reśumé Les taux de catécholamines cérébrales et l'activité in vitro de la tyrosine hydroxylase (TH) ont été déterminés chez la truite arc en ciel femelle. Chez des animaux triploides ou diploides en recrudescence ovarienne, les taux de catécholamines sont corrélés différemment à l'activité TH selon la structure cérébrale considérée. Le télencéphale présente un Km apparent pour la ptérine plus faible que l'hypothalamus. La dopamine (DA) et le 2-hydroxyestradiol (20HE2) inhibent l'activité de la TH selon des mécanismes respectivement compétitifs Les différents K1 pour la DA obtenus au niveau du télencéphale et de l'hypothalamus pourraient expliquer les différentes relations entre taux de catécholamines et activité TH obtenues pour ces deux structures. Le 20HE2 inhibe également l'activité TH in vitro: la présence d'un radical catéchol s'avère nécessaire (l'oestradiol (E2) n'est pas actif par lui-même), mais le mécanisme de l'inhibition reste à déterminer. Les effets rapides du 20HE2 ne peuvent expliquer l'activation de la synthèse catécholaminergique par l'E2 in vivo démontrée antérieurement.
