The majority of studies on the immunobiology of Trichinella species have centred on the larval muscular phase (L1) with a view to identifying immunodominant antigens located on the surface of the cuticle and in the larval secretions; the nucleus of the parasite-host interaction. These antigens have been classified as eight groups (TSL-1-TSL-8), of which those belonging to the group TSL-1 have been most intensely studied. The principal constituents are glycoproteins, glycan carriers that contain a unusual sugar, the tyvelose (3,6-dideoxy-d-arabinohexose). Studies aimed at improving serodiagnostic techniques to detect trichinellosis indicate that these antigens are ideal candidates. They are capable of inducing a strong humoral response involving the generation of specific antibodies against beta-tyvelose, a sugar that seems to be exclusive to the Trichuroidea. Furthermore, these glycoproteins appear to fulfil an important function in the development and maintenance of the parasite in the muscular niche, and they appear to be fundamental for the invasion of the intestinal epithelium. It has also been demonstrated that specific monoclonal antibodies against tyvelose can mediate a degree of immunoprotection in the rat through the phenomenon known as rapid expulsion.  相似文献

JNPL3 transgenic mice expressing a mutant tau protein, which develop neurofibrillary tangles and progressive motor disturbance, were crossed with Tg2576 transgenic mice expressing mutant beta-amyloid precursor protein (APP), thus modulating the APP-Abeta (beta-amyloid peptide) environment. The resulting double mutant (tau/APP) progeny and the Tg2576 parental strain developed Abeta deposits at the same age; however, relative to JNPL3 mice, the double mutants exhibited neurofibrillary tangle pathology that was substantially enhanced in the limbic system and olfactory cortex. These results indicate that either APP or Abeta influences the formation of neurofibrillary tangles. The interaction between Abeta and tau pathologies in these mice supports the hypothesis that a similar interaction occurs in Alzheimer's disease.  相似文献

The aim of this study is the development and evaluation of a rapid and accurate quantitative PCR (qPCR)‐based protocol for detection of zoonotic pathogen Streptococcus iniae in bacterial cultures and tissues of diseased fish. For this purpose, the lactate permease‐encoding (lldY) gene was selected as a target for the design of S. iniae‐specific primers based on comparative genomic analysis using 45 sequences retrieved from NCBI genome database. Specificity and applicability of these primers were tested using 115 bacterial strains and fish tissues infected with S. iniae. Sensitivity, reproducibility and efficiency of qPCR assay were also determined. The developed qPCR assay showed 100% specificity with pure bacterial cultures or DNA extracted from S. iniae or tissues of fish infected with the bacterium. The method has high sensitivity with a detection limit of 1.12 × 101 amplicon copies per assay (equivalent to 2 × 10–9 ng/µl) using bacterial DNA and of 1.44 × 101 gene copies in tissues of fish infected with S. iniae. In conclusion, this qPCR protocol provides an accurate and sensitive alternative for the identification of S. iniae and its detection on fish tissues that can be implemented as a routine tool in microbiological laboratories.  相似文献

Skeletal muscle dysfunction is a common systemic manifestation in several prevalent diseases. Predictive values are useful tools for the diagnosis and prognosis of diseases. In experimental animals, no reference values of muscle function evaluation have been so far reported. The objective was to obtain predictive values of maximal inspiratory pressure (MIP) and grip strength measurements in healthy rats. In 70 healthy rats, MIP and grip strength were measured in vivo weekly for five consecutive weeks using non‐invasive methodologies. Three ranges of rat body weights (250–299, 300–349 and 350–399 g) and lengths (37.0–41.0, 41.1–42.0 and 42.1–44.0 cm) were established. MIP and grip strength measurements falling within the ranges of weight 350–399 and 300–349 g and length 42.1–44.0 cm were significantly greater than values falling within 250–299 g and 37.0–41.0 cm ranges respectively. Specific weight‐ and length‐percentile distributions for MIP and grip strength measurements were calculated. As significant direct correlations were observed between rat weights and lengths and either MIP or grip strength measurements, regression equations relating all these variables were also determined. Skeletal muscle dysfunction is frequently associated with highly prevalent conditions. The significant predictive equations described for both MIP and grip strength measurements will enable scientists to better estimate the respiratory and peripheral muscle dysfunctions of laboratory animals, especially when conducting follow‐up and/or intervention investigations.  相似文献

Acorn production patterns and the annual evolution of water relations parameters of Quercus ilex ssp. ballota (Desf.) Samp. in a dehesa (an open woodland forest) subject to various soil treatments (ploughing, ploughing + sowing, control) were studied in southwest Spain from June 2006 to December 2008. The effects of soil water holding capacity and effective soil depth on soil and plant water status and acorn production were also investigated. Water parameters showed significant differences between the ploughing treatment and the control, and there were also significant temporal differences. Xylem water potential ranged from −3.4 ± 0.1 to −0.7 ± 0.2 MPa, and cuticular transpiration was 62.4–192.9 mmol H2O kg−1 s−1. Acorn production did not differ significantly according to soil treatment, and showed large intra-specific variability. Individual values ranged from 0 to 1,220 g m−2 (fresh weight). Positive relationships were found between xylem water potential, cuticular transpiration and relative water content measured at midsummer, and acorn production during the three studied years. These results suggest that climatic conditions and soil water availability have a strong influence on plant water status, and therefore on acorn development during summer. The results also reflect the ability of this species to adapt to the Mediterranean climate by preserving water during dry periods, which to a large extent can be attributed to stomatal closure at high relative water content levels, and low cuticular transpiration during these periods.  相似文献

This work describes a primer pair and a high‐throughput SYBR Green I‐based real‐time PCR protocol combined with melting curve analysis for identification and quantification of Vagococcus salmoninarum in bacterial cultures and infected fish tissues. The 16S rRNA gene was selected for the design of the primer pair (SalF and SalR). The sensitivity and specificity of this primer pair were compared with other previously designed for conventional PCR. Although both primer pairs showed 100% specificity using pure bacterial cultures or DNA extracted from bacteria or fish tissues, the primer pairs designed in this study showed the highest sensitivity with a detection limit of 0.034 × 100 amplicon copies per assay (equivalent to 2 × 10?11 ng/µl, Cq value of 30.49 ± 1.71). The developed qPCR protocol allowed the detection of V. salmoninarum in non‐lethal and lethal fish samples with detection levels of 0.17 × 100 gene copies in tissues artificially infected and 0.02 × 100 in tissues of fish experimentally infected with V. salmoninarum. The high sensitivity of the developed method suggests that it could be considered as a useful tool for diagnosis of vagococcosis and the detection of V. salmoninarum in asymptomatic or carrier fish.  相似文献

The effect of supplementing dehydrated seaweed Ulva clathrata (20 g/kg) in the squid fraction of a formulated fresh diet on the reproductive performance of Litopenaeus vannamei broodstock was evaluated in two commercial hatcheries (A and B). The origin of broodstock, use of ablation on females, tank/culture conditions (for maturation, spawning, and larval rearing) were different between hatcheries. Daily mortality was higher (< .01) for females in the control group compared to Ulva‐fed group of hatchery A, where females were ablated. No differences in mortality were found for unablated females in hatchery B, or with males from either hatchery. In hatchery A, broodstock fed Ulva produced more eggs (an increase in +5% compared to control, < .05) and nauplii per female (+18%, < .01). In hatchery B, broodstock fed Ulva produced significantly more eggs (+26%, < .05) and nauplii per day (+33%, < .05). In hatcheries A and B, eggs from broodstock fed Ulva had increased hatching rates (+13 and +7%, respectively, < .001 in both cases). The number of spawns per day had a tendency to be higher in broodstock fed Ulva in both hatcheries, but the difference did not reach statistical significance (+14% for hatchery A and +20% for hatchery B). Dehydrated U. clathrata appears therefore as a functional additive in L. vannamei maturation diets that enhances the reproductive performance at commercial scale, regardless of the management conditions.  相似文献

White clover is considered amongst the most important forage legumes in temperate agricultural systems. It contributes to increased quality of pastures through its ability to fix atmospheric nitrogen, providing high nutritional feed for livestock. White clover is host to the potexvirus White clover mosaic virus (WClMV), which is readily transmitted via animal trampling/grazing and mowing. WClMV infection may strongly impact white clover's performance, affecting its ability to compete with other plants in the pasture. Understanding the extent to which the virus influences white clover's growth and yield may contribute to improve pastoral management by pointing out the importance of stress factors on pasture plant performance and herbage yield. In the current study, the effects of WClMV infection and simulated grazing on white clover's fitness, oxidative damage and the induction of antioxidants were investigated. Two cultivars of white clover, Grasslands Huia and Grasslands Kopu II, were subjected to three harvests. In response to WClMV infection and grazing‐like cutting, white clover experienced oxidative stress as evidenced by the accumulation of lipid peroxides and protein carbonyls. Biomass accumulation, given as fresh weight and dry matter content, was also negatively affected. Both stresses induced an increase in ascorbate and glutathione levels and enhanced the activity of superoxide dismutase, ascorbate peroxidase, glutathione peroxidase, catalase and glutathione reductase. Glutathione S‐transferase activity and the levels of reduced ascorbate and reduced glutathione were not affected by either stress.  相似文献
