The objective of this study was to characterize follicular dynamics in pre-pubertal, pubertal and post-pubertal periods, as well as the effect of high-energy intake on follicular development and age at puberty in heifers. Thirty-one Nelore (Bos indicus) heifers, 6 months old, were randomly assigned to receive two different diets: one of low (GI) and other of high dietary energy intake (GII). Animals were evaluated in relation to body weight gain by being weighed every 21 days. Heifers were evaluated every other day by real-time linear ultrasonography to characterize ovarian structures development from weaning to post-pubertal period. Blood samples were collected to determine plasmatic concentrations of progesterone by RIA method. The ovulation was determined when progesterone concentrations were >1 ng/mL in three consecutive samples, and by ultrasound images of corpus luteum; and oestrous behaviour in some animals. Age at puberty differed among heifers of GII (17.00 +/- 0.46 months) compared with heifers of GI (19.87 +/- 0.47 months; p < or = 0.05). Maximum size of the dominant follicles at pre-pubertal period was greater in GII heifers than in GI (10.52 +/- 0.33 and 9.76 +/- 0.15 mm, respectively; p < or = 0.05). As heifers approached first ovulation time, size of dominant follicle increased (11.75 +/- 0.37 mm for GI and 12.52 +/- 0.91 mm for GII; p < or = 0.05). Body weight at puberty was not different in both groups (302.33 +/- 27.31 kg for GI and 326.19 +/- 27.78 kg for GII heifers; p > 0.05). We conclude that animals receiving high dietary energy intake attained the puberty earlier and the development of follicles were different than in low dietary energy intake.  相似文献

Twenty-one genetically lean, obese or contemporary slaughter weight castrated male pigs (6 mo old; seven of each genotype) were assigned to individual tether stalls and fed either a control diet (low fiber) or a diet containing 80% alfalfa meal (high fiber) at 1.50% of initial body weight for 71 d (1.75% for d 1 to 4). Apparent dry matter digestibility of the diets was estimated by determining acid insoluble ash in fecal samples. Fecal cellulolytic bacteria and total viable bacteria were enumerated at d 0, 14, 35, 49 and 70. Fecal inocula were used to determine 48-h in vitro digestibility of alfalfa meal fractions on the same days. Digesta rate of passage was determined by feeding a pulse dose of chromium-mordanted alfalfa fiber to the pigs fed the high-fiber diet. In vivo digestibility of both diets was less for the obese pigs than for the lean or contemporary genotypes. In vitro digestibility of alfalfa fiber fractions was not different between the genotypes fed either diet. When the high-fiber diet was fed, in vitro digestibility increased for all genotypes from d 0 to d 14, but not thereafter. The numbers of cellulolytic bacteria for all three genotypes were greater when pigs were fed the high-fiber diet (23.0 X 10(8), 51.6 X 10(8), 37.2 X 10(8) per gram fecal dry weight; obese, lean and contemporary, respectively) compared to the low-fiber diet (3.0 X 10(8), 3.2 X 10(8), 3.4 X 10(8), respectively).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Twenty-one genetically lean, obese or contemporary barrows (6 mo old; seven of each genotype) were assigned to individual tether stalls and fed a control diet (low-fiber) or a diet containing 80% alfalfa meal (high-fiber) at 1.50% of initial body weight for 71 d (1.75% for d 1 to 4). Backfat thickness was recorded ultrasonically at 2-wk intervals, and body weight was recorded at the beginning and end of the 10-wk experiment. Pigs were slaughtered after a 24-h fast, and carcass weight, length and backfat thickness and cross-sectional area of the longissimus muscle were measured. Weights of cecum, heart, liver and kidney and of full and empty stomach and colon and empty small intestine were recorded. Volume and weight of colon and cecum contents were determined. Restriction of digestible energy reduced weight gain to zero or below in pigs fed alfalfa meal compared with 220 g daily in pigs fed the low-fiber diet. Restriction of energy reduced backfat in all three genotypes. Liver, kidney and empty segments of the gastrointestinal tract as a percentage of body weight were increased by high fiber. Obese pigs had smaller longissimus muscle area, more backfat and smaller liver, heart, empty stomach and colon than lean or contemporary pigs, but there were no diet X genotype interactions for any of these traits. Obese pigs consistently had smaller digesta volumes and dry matter weights than the other genotypes. The increased relative organ weights and the associated disproportionate contribution of these organs to body energy expenditure have important implications for effects on basal metabolic rate.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Four Duroc x White composite crossbred (21.8 +/- 1.0 kg BW) and four 12-wk-old Meishan purebred (20.7 +/- 1.6 kg BW) growing barrows were used to determine the relative breed differences in metabolic and microbial responses to a high-fiber diet. The pigs were trained to consume 700 g of a diet containing 35% (as-fed basis) dehydrated alfalfa meal once daily. The pigs' daily intakes of DM, N, GE, NDF, and ADF were 610 g, 16.6 g, 2.64 Mcal, 150 g, and 88 g, respectively. On d 12 after surgical catheterization of the portal vein, ileal vein, and carotid artery, a 3-d total urine and feces collection was conducted. On d 24 after surgery, each pig was placed in an open-circuit calorimeter, and its catheters were connected to a system for simultaneous measurements of oxygen consumption by portal-drained viscera and by whole body, and the net portal absorption of VFA after a 24-h fasting and during a 5-h postprandial period. The VFA measured included acetic, propionic, isobutyric, butyric, isovaleric, and valeric acids. A second 3-d total urine and feces collection was conducted on d 30 after surgery. There were no differences (P = 0.13) between the first and second collections in apparent total-tract digestibility coefficients for nutrients and N retention of pigs. Compared with Duroc x White composite crossbred pigs, Meishan pigs had lower (P = 0.05) apparent digestibility coefficients for DM, N, NDF, hemicellulose, and N retention, but their portal-drained viscera used a greater (P = 0.05) fraction of whole-body oxygen consumption. No differences (P = 0.12) were found between Duroc x White composite crossbred and Meishan pigs in total viable bacteria and cellulolytic bacteria from fecal samples, in vitro digestibility of alfalfa NDF fractions by fecal inocula, whole-body oxygen consumption, net portal absorption of VFA, total energy of absorbed VFA, and the potential of absorbed VFA for meeting the energy needs for whole-body heat production. These results indicate that, in contrast to previous beliefs, the ability of Meishan growing pigs to utilize a high-fiber diet is not superior to that of Duroc x White composite crossbred growing pigs.  相似文献

Conservation of nitrogen in cattle feedlot waste with urease inhibitors.   总被引:3,自引:0,他引:3  
Feedlot cattle normally retain less than 20% of their dietary nitrogen intake. Sixty to 80% of the nitrogen excreted is normally lost through volatilization of ammonia, which is primarily generated from urea. This loss of ammonia nitrogen pollutes the environment and creates an unfavorable ratio of nitrogen to phosphorous (N:P) in the waste for crop growth. Two urease inhibitors, cyclohexylphosphoric triamide (CHPT) and N-(n-butyl) thiophosphoric triamide (NBPT) were evaluated for their ability to reduce the rate of urea hydrolysis in beef cattle feedlot pens. Initially, a total of six pens were used, two pens per treatment, with approximately 70 cattle per pen, and a single topical application of CHPT or NBPT at 20 mg/kg of manure. Essentially no urea was found in untreated pens. However, with CHPT treatment, 2 g of urea/kg of dry manure accumulated by d 4, and all gradually disappeared by d 11; NBPT conserved 3 and 3.5 g of urea/kg by d 4 and 9, respectively, and it had disappeared by d 14 (treatment [trt] x day, P = .003). A second study involved application of NBPT weekly for 6 wk. This caused urea to accumulate to a peak concentration of 17 g/kg of manure by d 30 (trt x day2, P = .001). Once the treatment was stopped the urea concentration began to decrease. When the NBPT was applied weekly, the concentration of ammonia in the waste was less for the treated pens (trt x day, P = .01), the total nitrogen was greater (trt x day, P = .04), pH tended to be lower (trt x day, P = .10), and the total volatile acids were not different (trt x day, P = .51) from untreated pens. We concluded that urease inhibitors could be used to control ammonia emissions from animal wastes, prevent environmental damage, and produce a more balanced (N:P) fertilizer from manure.  相似文献

Expression of Kit ligand (KL) and insulin‐like growth factor binding protein (IGFBP3) genes was studied at different in vivo and corresponding in vitro stages of development of the ovarian follicles in sheep. The expression of both KL and IGFBP3 was significantly higher in the primordial follicles relative to any other stage of development. Compared to the other stages, the KL expression in the cumulus cells from in vivo grown large antral follicles and that of IGFBP3 in COCs’ isolated from large antral follicles matured in vitro for 24 hr were significantly higher. In the oocytes from in vivo grown ovarian follicles, the expression of KL was the same at all the stages of development. Insulin‐like growth factor binding protein 3 expression was also the same in the oocytes at all the stages of the development except for a significantly lower expression in those from antral follicles. The expression of KL in the cumulus cells decreased significantly in the in vitro grown early antral follicles but did not change further as the development progressed. The expression of IGFBP3 in the cumulus cells from in vitro grown ovarian follicles appeared to increase as the development progressed although the increase was not significant between any two consecutive stages of development. In the oocytes in in vitro grown ovarian follicles, the expression levels of KL and IGFBP3 genes did not change with development. It is concluded that (i) KL and IGFBP3 genes follow specific patterns of expression during ovarian folliculogenesis and (ii) in vitro culture of preantral follicles compromises the development potential through alterations in the stage‐specific patterns of expression of these and other developmentally important genes.  相似文献

Corn ethanol production removes starch and concentrates the remaining nutrients, including CP and minerals. When wet distillers grains with solubles (WDGS) are fed to cattle in place of corn, CP and minerals often exceed dietary needs. This may increase N emission, P run-off, and odor production. These variables are evaluated in this study. Crossbred steers (n = 160; 434 +/- 8 kg) were assigned in a completely randomized block design to 9 x 9 m pens with concrete floor (10 animals/pen; 4 pens/treatment). Steers were fed a finishing diet that contained 0, 20, 40, or 60% WDGS on a DM basis, and provided 13.3, 15.5, 20.6, or 24.9% CP, respectively. Two kilograms of manure slurry (14 to 23% DM) were collected from each pen monthly (Aug. 20, Sep. 24, and Oct. 22). Samples were analyzed immediately for odorants, DM, pH, NH(3), total alcohol, l-lactate, and concentrations of generic Escherichia coli. After incubation of the samples at 22 degrees C for 2, 4, 7, 10, 15, 21, and 28 d, samples were analyzed for methane production in addition to the above characteristics. Before incubation, NH(3), H(2)S, indole, phenol, isovalerate, isobutyrate, and acetate increased (P < 0.01) with increasing amounts of WDGS in the diet. Other odorants, including skatole, caproate, valerate, butyrate, and propionate, were greater (P < 0.01) in manure slurries from cattle fed 20 or 40% WDGS, compared to 0% WDGS. The l-lactate was greater (P < 0.01) in slurries from cattle fed 0% WDGS (447 mu mol/g of DM) compared with the other treatment slurries (14 to 15 mu mol/g of DM). After incubation, l-lactate contributed to lowered slurry pH (6.3, 7.1, 7.6, and 8.2, respectively, for 0, 20, 40, and 60% WDGS), which inhibited microbial fermentation, E. coli persistence, and methane production. Because of the favorable, more neutral pH in the 40 and 60% WDGS slurries, many of the odorant compounds were rapidly converted to methane during a 28-d static incubation. Escherichia coli O157:H7 inoculated into subsamples of the manure slurries exhibited behavior similar to that of naturally present generic E. coli, surviving in greater numbers longer (P < 0.05) in 20 and 40% WDGS slurries than in 0% WDGS. These data indicate feeding WDGS can increase odorants in manure slurries and extend the persistence of E. coli.  相似文献

Livestock odors are closely correlated to airborne concentrations of volatile organic compounds (VOC), which are a complex mixture of carbon-, sulfur-, and nitrogen-containing compounds produced primarily during the incomplete anaerobic fermentation of animal manure by microorganisms. Volatile fatty acids, alcohols, and aromatic ring compounds comprise a substantial fraction of VOC, yet very little is known about their biochemical origin and environmental factors controlling their production. The anaerobic production of fermentation products and consumption of substrates (CP, starch, and nonstarch carbohydrate) were analyzed in slurries of fresh (< 24 h) and aged (> 1 d) cattle manure over several weeks. Ethanol, acetate, propionate, butyrate, lactate, and H2 were the major products of fermentation. Aged cattle manure produced twice the concentration of VFA during incubation produced by the fresh manure (P < 0.001). Aromatic compounds (phenols, indoles, and benzoates) remained unchanged in both manures. Production of VFA from fresh manure was inhibited when the pH fell below 4.5. It is likely that the presence of calcareous soil, which has a high buffering capacity, and lactate-consuming microorganisms minimized acidification in the aged manure slurries. Low starch content limited VFA production in the aged manure. Starch was the likely biochemical source for fermentation products in both manures based on the strong negative correlations between fermentation product and starch content (r = -0.944 and -0.773) and ratio of fermentation products produced to starch consumed (r = 0.64 and 0.72) for fresh and aged manure, respectively. Nonstarch carbohydrate served an indeterminate role in the production of fermentation products. Nonstarch carbohydrate decreased by 4.7 and 23.4 g/L in the fresh and aged manure, respectively, whereas the starch content decreased by 18.6 and 22.4 g/L in the fresh and aged manure, respectively. The concentration of CP did not change, which suggests a balance between protein consumption and new bacterial biomass production. We conclude that the types of substrates in cattle manure and the feedlot soils where they are deposited are significant factors in the production of odors.  相似文献

Studies have demonstrated the importance of mitochondria to sperm functionality, as the main source of ATP for cellular homoeostasis and motility. However, the role of mitochondria on sperm metabolism is still controversial. Studies indicate that, for some species, glycolysis may be the main mechanism for sperm energy production. For ram sperm, such pathway is not clear. Thus, we evaluated ram sperm in response to mitochondrial uncoupling and glycolysis inhibition aiming to assess the importance of each pathway for sperm functionality. Statistical analysis was performed by the SAS System for Windows, using the General Linear Model Procedure. Data were tested for residue normality and variance homogeneity. A p < .05 was considered significant. Groups treated with the mitochondrial uncoupler Carbonyl cyanide 3 chlorophenylhydrazone (CCCP) showed a decrease in the percentage of cells with low mitochondrial activity and high mitochondrial membrane potential. We also observed that the highest CCCP concentration promotes a decrease in sperm susceptibility to lipid peroxidation. Regardless the lack of effect of CCCP on total motility, this substance induced significant alterations on sperm kinetics. Besides the interference of CCCP on spermatic movement patterns, it was also possible to observe such an effect in samples treated with the inhibitor of glycolysis (2‐deoxy‐d ‐glucose, DOG). Furthermore, treatment with DOG also led to a dose‐dependent increase in sperm susceptibility to lipid peroxidation. Based on our results, we suggest that the glycolysis appears to be as important as oxidative phosphorylation for ovine sperm kinetics as this mechanism is capable of maintaining full motility when most of the cells have a low mitochondrial membrane potential. Furthermore, we found that changes in the glycolytic pathway trough glycolysis inhibition are likely involved in mitochondrial dysfunction and sperm oxidative unbalance.  相似文献

An accurate method for estimation of lignin concentration is important for prediction of the digestible energy content of livestock feeds. The accuracy of lignin concentration estimates based on the Klason lignin and acid detergent lignin methods was compared. Ten diverse forage samples were analyzed for protein, carbohydrates, lipids, organic acids, ash, lignin (by both methods), and gross energy. The accuracy of the two lignin concentration estimates was examined by comparing the measured forage gross energy to a gross energy value calculated from the compositional analysis. Use of the acid detergent lignin estimate in this gross energy calculation accounted for 68-84% of the forage gross energy compared to 85-97% of the gross energy using Klason lignin. These results indicate that while Klason lignin estimates are substantially higher than acid detergent lignin estimates, Klason lignin is the more accurate lignin method and does not overestimate lignin because gross energy recoveries were less than 100%.  相似文献
