Organic production of one of the most popular botanical supplements, Echinacea, continues to expand in the U.S. Echinacea seeds typically show a high degree of dormancy that can be broken by ethephon or gibberelic acid (GA), but these methods are currently disallowed in organic production. In order to determine the efficacy of non-chemical seed treatments, we evaluated the effect of varying seed source and supplying light, with and without cold-moist stratification, on seed germination of the three most important medicinal species of Echinacea, E. angustifolia DC, E. purpurea (L) Moench, and E. pallida (Nutt.) Nutt. Treatments included cold-moist stratification under 24 h light, 24 h dark, and 16/8 h light/dark to break seed dormancy. We found that germination was greater in the E. purpurea and E. pallida seeds from a commercial organic seed source compared to a public germplasm source. When seeds were not cold-moist stratified, 16-24 h light increased germination in E. angustifolia only. Echinacea angustifolia, E. purpurea, and E. pallida seeds that were cold-moist stratified under 16-24 h of light for 4 wk had a significantly greater percentage and rate of germination compared to seeds germinated in the dark. Therefore, cold-moist stratification under light conditions is recommended as a method to break seed dormancy and increase germination rates in organic production of Echinacea. 相似文献

The aim was to evaluate the effect of different concentrations of caffeine added in activating solution over sperm motility in fresh and thawed semen of pacu, Piaractus mesopotamicus, and curimba, Prochilodus lineatus. The activating solutions were prepared with sodium bicarbonate solution of 0.76% (NaHCO3) and caffeine was added at concentrations of 2.5, 5.0, 10.0, and 20.0 mM. As control, a solution of NaHCO3 0.76 without caffeine was used. Eight males of pacu and 20 males of curimba were used. Aliquots of 200 μL of semen were diluted in 800 μL extender solution (DMSO 10% and BTS 5%), placed in 0.5 mL straws and cryopreserved for 7 d in a liquid nitrogen tank. There was a linear increase in sperm motility for fresh semen of pacu, and for curimba fresh and thawed semen (P < 0.05), due to the increase in the concentration of caffeine. There was a quadratic response for duration of motility for thawed semen of pacu and for fresh semen of curimba (P < 0.05), respectively. These results indicate that addition of caffeine in the activator solution can improve sperm motility parameters, however, is dependent on the species and concentration used. 相似文献

A 4-mo-old northern red-shouldered macaw (Diopsittaca nobilis) was admitted to the veterinary hospital of the Arruda Câmara Zoo, in the State of Paraiba, Brazil, for investigation of an orbital mass. Given rapid progression and lack of response to treatment, the bird was euthanized, and an autopsy was performed. Histologically, the mass consisted of a retrobulbar invasive tumor characterized by tubular and rosette-like structures, with interspersed heteroplastic tissues, such as aggregates of neuroglial cells and islands of hyaline cartilage. The tumor was immunopositive for pancytokeratin, GFAP, NSE, and S100. These findings were compatible with an ocular teratoid medulloepithelioma, a neoplasm best described in humans but also reported rarely in young cockatiels and African Grey parrots. 相似文献

During the last few decades, land use changes have largely affected the global warming process through emissions of CO2. However, C sequestration in terrestrial ecosystems could contribute to the decrease of atmospheric CO2 rates. Although Mediterranean areas show a high potential for C sequestration, only a few studies have been carried out in these systems. In this study, we propose a methodology to assess the impact of land use and land cover change dynamics on soil organic C stocks at different depths. Soil C sequestration rates are provided for different land cover changes and soil types in Andalusia (southern Spain). Our research is based on the analysis of detailed soil databases containing data from 1357 soil profiles, the Soil Map of Andalusia and the Land Use and Land Cover Map of Andalusia. Land use and land cover changes between 1956 and 2007 implied soil organic C losses in all soil groups, resulting in a total loss of 16·8 Tg (approximately 0·33 Tg y−1). Afforestation increased soil organic C mostly in the topsoil, and forest contributed to sequestration of 8·62 Mg ha−1 of soil organic C (25·4 per cent). Deforestation processes implied important C losses, particularly in Cambisols, Luvisols and Vertisols. The information generated in this study will be a useful basis for designing management strategies for stabilizing the increasing atmospheric CO2 concentrations by preservation of C stocks and C sequestration. We investigated the effects of partial root-zone drying (PRD) applied at different periods on leaf water relations, vegetative development, fruit yield, must and wine quality in wine grapes (Vitis vinifera L. cv. Monastrell) during a 3-year field experiment in order to determine the importance of the timing of PRD application on physiological and agronomical vine response under semiarid conditions. Two irrigation treatments were applied: conventional drip irrigation (CI) and PRD. Both treatments received the same annual water quantity. Each year the PRD treatment was applied at different periods of the growth cycle. In 1999 PRD was applied from veraison to harvest (end July–early September); in 2000 from fruit set to harvest (mid June–early September); and in 2001 PRD from budburst to harvest (mid April–early September). Leaf water relations and gas exchange during the experimental period were not significantly affected by PRD treatment. In 1999 and 2000 there was no significant treatment effect on vegetative development, yield or fruit quality. However, in 2001 (when PRD was applied from budburst to harvest), reproductive and vegetative development was clearly altered in PRD vines. Fruit set percentage and vegetative development (shoot length, pruning weight and primary and lateral leaf area) were significantly increased in PRD vines compared to CI. This resulted in both higher yield (kg per vine) (43%) and water use efficiency (40%) compared to CI vines. Berry number per cluster and cluster weight were also significantly increased in PRD vines. Notwithstanding higher yield in PRD vines and a similar berry size, the must and wine quality was not significantly altered, indicating a higher synthesis and accumulation of photoassimilates and metabolites in the berries of PRD vines. We conclude that there was an positive effect on vegetative and reproductive growth when long-term PRD was applied from the beginning of growing season (budburst), suggesting that early onset of PRD is desirable to intensify PRD response under these semiarid conditions. Nevertheless from these results we need to further investigate the long- and short-term effects of PRD, with moderate water amounts, on vegetative and reproductive development such as flowering and fruit set processes in wine grapes. 相似文献

This study evaluated the application of stable isotopes of carbon as an alternative and more accurate method to determine gastrointestinal transit time (GTT) in fish by comparing it to the inert marker method. The stable isotope method detects alterations of the normal carbon flow in a biological system by analyzing naturally occurring isotopes of carbon, contrary to studies based on conventional techniques that apply external markers to the diet to determine GTT through visual observation of the color change in feces. Therefore, 320 pacu, Piaractus mesopotamicus juveniles were reared in 32 tanks under two different temperatures (25 and 29 C). The pacu juveniles received two different diets, one based on ingredients derived from C3 photosynthetic cycle plants and the other based on C4 plant ingredients, both containing titanium oxide (TiO2) as a marker. After 40 d, the isotopic signature of the diets was changed, and the marker was replaced by chromic oxide (Cr2O3). In the isotopic technique, the feces were analyzed to determine the exchange in the isotopic ratio of carbon δ13C. Both methods found that GTT was faster (nearly 6 h) in fish at 29 C when using the C4/C3 feeding strategy and slower in fish at 25 C using the C3/C4 strategy (15 h by inert marker and 18 h by the isotopic method). In conclusion, GTT determination in pacu juveniles using the stable isotope technique exhibits the same accuracy obtained with the inert marker method at temperatures suitable (nearly 29 C) for the metabolism of these animals. 相似文献

Mineral fertilizers, organic amendments, and pesticides are inputs commonly used in conventional farming practices. The aim of this study was to evaluate the effects of single or combined applications of spent grape marc-vermicompost, urea, and/or diuron on soil-enzyme activities and the persistence of this herbicide in soils with low organic carbon content. The application of vermicompost enhanced dehydrogenase (DHase) enzyme activity over time but altered soil urease activity to a very limited extent. The reduction in diuron concentrations and the increase in DHase activity indicated that the soil microorganisms were capable of degrading the ureic herbicide. Treatment with vermicompost and diuron had a stimulatory effect on soil microbial activity. On the whole, the application of diuron and urea to the vermicompost-amended soil raised DHase and urease activity to maximum levels (>3 μg INTF g?1 h?1 and >47 μg NH4+ g?1 h?1, respectively). The application of urea to the unamended and vermicompost-amended soil decreased diuron persistence from 18.8 and 33 d to 12.5 and 15 d, respectively. Our findings show that although vermicompost additions reduce diuron availability, this boosts diuron degradation when combined with urea. These additions, under different soil management conditions, minimize the bioavailability and persistence of diuron and consequently the risk of leaching and seepage into aquifers. Compared with untreated soils, these types of treated soils could also improve agricultural sustainability and the quality of the environment. 相似文献
