A previous RFLP analysis showed that the Aegilops ventricosa chromosome 6MV which compensates for the absence of 6D in 6MV (6D) wheat substitution lines was a 2/6 translocated chromosome, either 2S–6S.6L or 2S–6L.6S. The distal part of its long arm consists of a translocated segment belonging to homoeologous group 2. Chromosome 6MV carries a gene(s) for resistance to cereal cyst nematode. In order to define the part of 6MV (2S or 6S or 6L) involved in this resistance, addition lines with a 6MV deleted either for its short arm or for the distal part of its long arm were evaluated. It was shown that the gene(s) is carried by the group 2 translocated segment. The hypothesis that the gene(s) could be allelic to Cre2, another gene conferring resistance to the nematode introduced into the wheat complement from Ae. ventricosa is discussed.  相似文献

Previous studies showed that the intermediate level of resistance in bread wheat line ‘VPM1’ to pathotype Ha12 of the cereal cyst nematode could be conferred by an Aegilops ventricosa‐derived gene, CreX, in chromosome arm 2AS, which also carries the rust resistance genes Yrl7, Lr37 and Sr38. Near isogenic lines (NILs) differing for the presence and absence of the Ae. ventricosa‐derived linked genes Yrl7/Lr37/Sr38 were tested with cereal cyst nematode. Lines carrying Yr17 produced significantly fewer nematode cysts than the controls. An infested soil experiment produced better differentiation among resistant and susceptible genotypes. Susceptibility of ‘Trident’ indicated that linkage between CreX and Yr17 is incomplete. Microsatellite markers did not differentiate between ‘Trident’ and CreX‐carrying genotypes. However, Xgwm636 (104) was associated with the presence of Yr17 in all six genetic backgrounds. Since none of the reported cereal cyst nematode resistance genes is located in chromosome 2AS, CreX was designated as Cre5.  相似文献

Vector efficiency of 44 clonal lines (clones) of Sitobion avenae belonging to 31 different genotypes (distinct patterns for five microsatellite loci) originating from Western France was evaluated by transmitting the isolate PAV4 of BYDV-PAV to barley seedlings. Variation in transmission rates from 3.7% to 92.5% was observed, with significant effects of the aphid clone, of the plant species on which clones were collected, and of the reproductive mode of the clones. When genotypes are considered instead of clones, a continuum in transmission rates was observed. A subset of S. avenae clones was tested for transmission of one (10 clones) and 13 (4 clones) other BYDV-PAV isolates, and a clear clone effect modulated by an isolate effect was observed. Crosses were made between clones with different vectoring phenotypes and their F1 progeny were tested for PAV4 transmission. The narrow sense heritability of the PAV transmission character was rather high in the F1 families (h2=0.5) and the segregation analyses suggested an oligo/polygenic determinism of this character. The possibility of generating new transmission variants by sexual reproduction and its consequences on transmission mechanism studies and on BYD epidemics are discussed.  相似文献

ABSTRACT Two types of genetic resistance to Leptosphaeria maculans usually are distinguished in Brassica napus: qualitative, total resistance expressed at the seedling stage and quantitative, partial resistance expressed at the adult plant stage. The latter is under the control of many genetic factors that have been mapped through quantitative trait loci (QTL) studies using 'Darmor' resistance. The former usually is ascribed to race-specific resistance controlled by single resistance to L. maculans (Rlm) genes. Three B. napus-originating specific Rlm genes (Rlm1, Rlm2, and Rlm4) previously were characterized. Here, we report on the genetic identification of two novel resistance genes, Rlm3 and Rlm7, corresponding to the avirulence genes AvrLm3 and AvrLm7. The identification of a novel L. maculans- B. napus specific interaction allowed the detection of another putative new specific resistance gene, Rlm9. The resistance genes were mapped in two genomic regions on LG10 and LG16 linkage groups. A cluster of five resistance genes (Rlm1, Rlm3, Rlm4, Rlm7, and Rlm9) was strongly suggested on LG10. The relation between all these specific resistance genes and their potential role in adult-plant field resistance is discussed. These two Rlm-carrying regions do not correspond to major QTL for Darmor quantitative resistance.  相似文献

A cross-sectional study was conducted in Hawassa town and its surroundings from October 2007 to May 2008 to estimate the prevalence of bovine tuberculosis (BTB) based on comparative interadermal tuberculin test (CIDT) and abattoir survey. Accordingly, 39 herds comprising 413 cattle were subjected to CIDT, and the herd and individual animal prevalence were 48.7% (19/39) and 11.6% (48/413), respectively. One of the 16 milk samples collected from tuberculin-positive cows was culture positive. The prevalence significantly differed among the age group (P = 0.001) and management system (P = 0.001). Thus, age group over four (OR = 7.9) and animal with poor management system (OR = 4.1) had a higher odds for tuberculin reactivity compared to those with age group under four and cattle with good management system, respectively. Of the total 1,023 cattle subjected to postmortem examination, 11 (1.1%) were found to be positive for gross tuberculous lesions. Larger proportion (50%) of TB lesion was recorded in the respiratory pathway followed by digestive system (28.6%) and prescapular lymph nodes (21.4%). Of 14 tissue specimens collected from the gross lesions, four (28.6%) were positive for histopathological TB lesions. In conclusion, this study revealed the importance of BTB in the study area in particular and the region in general.  相似文献

Higher harvest index, widely reported as a main trait supporting major improvements in rice yield potential, also supports the higher performance of hybrids over inbreds. Although higher sink size is generally claimed for being the driver of higher grain filling in hybrid rice, it is relevant to question whether efficient sink regulation, that is timing and magnitude of the successive sink activities over crop growth from early stage, plays a role in supporting higher harvest index and is a key feature ensuring higher performance of hybrid rice. The partitioning coefficient (PC), as the increase in dry matter of one single organ over that of shoot, was calculated over short and successive periods to quantify the dynamics of dry matter partitioning. Four hybrid and four inbred genotypes of similar crop duration were grown under the same cropping management on the IRRI farm during a wet and a dry seasons. While PC to blade and sheath was similar for both plant types in both seasons at the early stage, PC to culm of hybrids was higher than that of inbreds, and PC to blade was lower, in the late vegetative and early reproductive phases. During the late reproductive phase, PC to panicle of hybrids was higher than that of inbreds, and PC to culm was lower whereas culm elongation was similar for both plant types. During grain filling, PC to culm was significantly more negative with hybrids which indicated stronger remobilization with hybrids. Specific culm length before anthesis, as low as 40 cm g−1, accounted for storage ability, and at maturity, as high as 130 cm g−1, for remobilization ability. Sink strength index, as a better indicator of dry matter partitioning efficiency than harvest index, and unfilled grain size, as an indicator of assimilate wastage, were suggested as key indices to account for the better sink regulation in hybrid rice and to improve screening protocols for increasing yield potential and tolerance to lodging.  相似文献

Shigella, the leading cause of bacillary dysentery, uses a type III secretion system (TTSS) to inject proteins into human cells, leading to bacterial invasion and a vigorous inflammatory response. The bacterium is protected against the response by the O antigen of lipopolysaccharide (LPS) on its surface. We show that bacteriophage-encoded glucosylation of Shigella O antigen, the basis of different serotypes, shortens the LPS molecule by around half. This enhances TTSS function without compromising the protective properties of the LPS. Thus, LPS glucosylation promotes bacterial invasion and evasion of innate immunity, which may have contributed to the emergence of serotype diversity in Shigella.  相似文献

Brassica napus-B. nigra addition lines were previously created using the variety ‘Darmor’ as the oilseed rape genetic background. Two isozyme loci and 46 RAPD markers were added on five different B. nigra chromosomes. The oilseed rape variety used was highly susceptible to blackleg at the cotyledon stage and only the addition of chromosome 4 gave the same level of blackleg resistance as B. nigra. This resistance was efficient whatever the isolates used. A significant effect on the development of stem canker under field conditions was observed only for the line carrying chromosome 4 which was more resistant than the susceptible control. The potential effects of two other chromosomes have to be confirmed. F1 hybrids obtained by crosses between two highly susceptible lines and the monosomic addition line carrying chromosome 4 were examined under field conditions. No effect of the oilseed rape genetic background on the expression of resistance was detected. The introduction of this resistance and mapping of the gene(s) into oilseed rape varieties are discussed.  相似文献

In order to establish whether commercial hybrid seed production between apetalous and apetalous rapeseed lines could be developed through insect cross-pollination, observations were made to compare the foraging behaviour of honeybees on both flower types with regard to their attractiveness. Number of flowers/m2 and nectar volumes, considered as important attractive factors, did not differ between the two lines but nectar sugar composition was slightly different. Honeybee density was significantly higher on the apetalous line when their foraging activity was rather high. Honeybees showed a considerable constancy to each flower type. Passages between both types were not very frequent. Nectar foraging behaviour, observed under cages, differed with the floral morphology. On the apetalous flowers, honeybees visited the two inner nectaries by crawling over the anthers and the stigma, while on the apetalous ones they often inserted their tongues between the sepals, thus having less contact with the reproductive organs. From these results, the honeybee efficiency in cross-pollination between petalous and apetalous lines appears to be reduced.  相似文献
