Mammalian tooth crowns have precise functional requirements but cannot be substantially remodeled after eruption. In developing teeth, epithelial signaling centers, the enamel knots, form at future cusp positions and are the first signs of cusp patterns that distinguish species. We report that ectodin, a secreted bone morphogenetic protein (BMP) inhibitor, is expressed as a "negative" image of mouse enamel knots. Furthermore, we show that ectodin-deficient mice have enlarged enamel knots, highly altered cusp patterns, and extra teeth. Unlike in normal teeth, excess BMP accelerates patterning in ectodin-deficient teeth. We propose that ectodin is critical for robust spatial delineation of enamel knots and cusps.  相似文献

The aim of this study was to assess the diversity of thermotolerant Campylobacter spp. isolated from turkey flocks at six rearing farms 1-2 weeks prior to slaughter (360 faecal swab samples) and from 11 different stages at the slaughterhouse (636 caecal, environmental, neck skin and meat samples). A total of 121 Campylobacter isolates were identified to species level using a multiplex PCR assay and were typed by pulsed-field gel electrophoresis (PFGE) and flaA-short variable region (SVR) sequencing. All Campylobacter isolates were identified as Campylobacter jejuni. PFGE analysis with KpnI restriction enzyme resulted in 11 PFGE types (I-XI) and flaA SVR typing yielded in nine flaA-SVR alleles. The Campylobacter-positive turkey flocks A, C and E were colonized by a limited number of Campylobacter clones at the farm and slaughter. The present study confirms the traceability of flock-specific strains (PFGE types I, V and IX; flaA types 21, 36 and 161) from the farm along the entire processing line to meat cuts. It seems that stress factors such as high temperature of the defeathering water (54-56 °C), drying of the carcass skin during air chilling (24 h at 2 °C), and oxygen in the air could not eliminate Campylobacter completely. Campylobacter-negative flocks became contaminated during processing by the same subtypes of Campylobacter introduced into the slaughter house by preceeding positive flocks even if they were slaughtered on subsequent days. Proper and efficient cleaning and disinfection of slaughter and processing premises are needed to avoid cross-contamination, especially in countries with a low prevalence of Campylobacter spp. The majority of flaA SVR alleles displayed a distinct association with a specific PFGE type. However, a linear relationship for all strains among both typing methods could not be established. To specify genetic relatedness of strains, a combination of different genotyping methods, is needed.  相似文献

Background

Campylobacter is the most common cause of bacterial enteritis worldwide. Handling and eating of contaminated poultry meat has considered as one of the risk factors for human campylobacteriosis.Campylobacter contamination can occur at all stages of a poultry production cycle. The objective of this study was to determine the occurrence of Campylobacter during a complete turkey production cycle which lasts for 1,5 years of time. For detection of Campylobacter, a conventional culture method was compared with a PCR method. Campylobacter isolates from different types of samples have been identified to the species level by a multiplex PCR assay.

Methods

Samples (N = 456) were regularly collected from one turkey parent flock, the hatchery, six different commercial turkey farms and from 11 different stages at the slaughterhouse. For the detection of Campylobacter, a conventional culture and a PCR method were used. Campylobacter isolates (n = 143) were identified to species level by a multiplex PCR assay.

Results

No Campylobacter were detected in either the samples from the turkey parent flock or from hatchery samples using the culture method. PCR detected Campylobacter DNA in five faecal samples and one fluff and eggshell sample. Six flocks out of 12 commercial turkey flocks where found negative at the farm level but only two were negative at the slaughterhouse.

Conclusion

During the brooding period Campylobacter might have contact with the birds without spreading of the contamination within the flock. Contamination of working surfaces and equipment during slaughter of a Campylobacter positive turkey flock can persist and lead to possible contamination of negative flocks even after the end of the day''s cleaning and desinfection. Reduction of contamination at farm by a high level of biosecurity control and hygiene may be one of the most efficient ways to reduce the amount of contaminated poultry meat in Finland. Due to the low numbers of Campylobacter in the Finnish turkey production chain, enrichment PCR seems to be the optimal detection method here.  相似文献

Instead of sole nutrient starvation to boost algal lipid production, we addressed nutrient limitation at two different seasons (autumn and spring) during outdoor cultivation in flat panel photobioreactors. Lipid accumulation, biomass and lipid productivity and changes in fatty acid composition of Nannochloropsis oculata were investigated under nitrogen (N) limitation (nitrate:phosphate N:P 5, N:P 2.5 molar ratio). N. oculata was able to maintain a high biomass productivity under N-limitation compared to N-sufficiency (N:P 20) at both seasons, which in spring resulted in nearly double lipid productivity under N-limited conditions (0.21 g L−1 day−1) compared to N-sufficiency (0.11 g L−1 day−1). Saturated and monounsaturated fatty acids increased from 76% to nearly 90% of total fatty acids in N-limited cultures. Higher biomass and lipid productivity in spring could, partly, be explained by higher irradiance, partly by greater harvesting rate (~30%). Our results indicate the potential for the production of algal high value products (i.e., polyunsaturated fatty acids) during both N-sufficiency and N-limitation. To meet the sustainability challenges of algal biomass production, we propose a dual-system process: Closed photobioreactors producing biomass for high value products and inoculum for larger raceway ponds recycling waste/exhaust streams to produce bulk chemicals for fuel, feed and industrial material.  相似文献

In hydropeaking rivers, flow regulation typically follows demands in electricity consumption. One hundred and twenty one adult salmon were tagged with radio transmitters to study their spawning run migration patterns in hydropeaking conditions. The fish were released in small groups into the rivers Kemijoki and Iijoki, in Finland. Typically, salmon made subsequent up‐ and downstream movements ascending upstream to the tailrace of the power station and then descending again downstream to lower water velocities. The rate of these migration attempts was higher in mid‐summer and at relatively high flows. On an hourly basis, the pattern of migration attempts followed a trend of increasing attempts at dawn and higher discharges and a decreasing trend towards dusk and low flows. It was concluded that day length and discharge were the most important environmental factors, while temperature had less effect on the pattern of migration attempts.  相似文献

A common observation is that egg size increases with maternal size within populations. Several hypotheses have been proposed to explain this phenomenon. Most recently, it was suggested that the optimal egg size depends on female size due to correlations between (i) breeding time and egg size selection and (ii) female size and breeding time, and as such, the positive egg size–maternal size relationship represents an adaptive strategy. Here, we test the second of these two premises in two salmonid species, Atlantic salmon (Salmo salar) and coho salmon (Oncorhynchus kisutch) for which we have detailed breeding and egg size data. The body size of spawning females decreased throughout the season in coho salmon, but not in Atlantic salmon. Furthermore, we did not find a direct correlation between egg size and breeding time, which would also be expected if this premise was correct. Accordingly, breeding date does not appear to be as important as other factors in shaping among female differences in egg size.  相似文献

Impacts of forestry have been described in many studies, with a strong focus on changes in run-off water quality. Biological responses to forest drainage and the efficiency of commonly used water protection methods remain largely unknown, however. We used a six-year replicated before-after-control-impact (BACI) field experiment to assess the effects of forest drainage improvement works (ditch cleaning; digging of supplementary ditches) on water chemistry, sedimentation, brown trout egg mortality and macroinvertebrate community composition in six headwater streams in Northern Finland. Standard water protection methods (ditch break combined with sedimentation pit) were used to reduce fine sediment (<1.0 mm) loading into the streams. Six artificial spawning sites were constructed in each stream, three downstream (treatments) and three upstream (controls) of the drainage areas. Sediment loading did not increase after forest drainage, and no changes were detected in water quality. Brown trout egg mortality rate increased after drainage works, but more so in the upstream controls than downstream treatment sections, and only in smaller (watershed <6.25 km2) streams. All macroinvertebrate metrics (e.g. total macroinvertebrate diversity and density) decreased significantly through time. However, these responses were observed in both control and treatment sections, and could not have been caused by the drainage works directly but were rather related to inter-annual variability in precipitation and discharges. Our results thus highlight the importance of placing results from local monitoring studies in a regional context.  相似文献
