It has long been recognised that steroids can have both organisational and activational effects on the reproductive neuroendocrine axis of many species, including the sheep. Specifically, if the ovine foetus is exposed to testosterone during a relatively short 'window' of in utero development (from approximately day 30-90 of a 147 day pregnancy) the neural mechanisms regulating gonadotrophin releasing hormone (GnRH) secretion become organised in a male-specific manner. In post-natal life the consequences of foetal androgen exposure are sexually differentiated responses of the GnRH neuronal network to activation by factors such as photoperiod and ovarian steroid hormones. Studies in the gonadectomized lamb have demonstrated that elevated concentrations of oestrogen (E) are unable to trigger a preovulatory-like GnRH surge in the male and the androgenized ewe lamb. Further, these animals have markedly reduced sensitivity to the inhibitory actions of progesterone on tonic GnRH release compared with normal ewes. The reasons for these abnormal steroid feedback mechanisms may reside in sexually dimorphic inputs to the GnRH neurone, including those from oestrogen-receptive neurones in the arcuate nucleus that synthetize the neuropeptide, neurokinin B (NKB). The consequences of in utero androgen exposure are reflected in a progressive and dramatic impairment of fertility in the ovary-intact ewe.  相似文献

Tissue culture cells from several mammalian species, including three primate lines, were transfected with recombinant vectors carrying Escherichia coli xanthine-guanine phosphoribosyltransferase or Tn5 aminoglycoside phosphotransferase dominant selectable markers. Human HeLa and SV40-transformed xeroderma pigmentosum cells exhibited stable transformation frequencies of at least 10(-3) (0.1 percent). CV-1, an African green monkey kidney cell line, could be stably transformed with the exceptionally high frequency of 6 X 10(-2) (6 percent).  相似文献

Members of the genus Acanthamoeba are usually free-living amebae and are found in a variety of ecological niches including soil, fresh and brackish water, dust in air, filters of heating, ventilating, and air conditioning units, swimming pools and hot tubs, etc. Occasionally, they are also known to cause central nervous system infections in humans and other animals. We isolated into culture an amoeba from the liver tissue of a keel-billed toucan and identified it as Acanthamoeba sp. based on culture characteristics and immunofluorescent analysis. Further, we characterized the cultured amoeba and also the amoeba in the liver tissue as Acanthamoeba, genotype T4, by sequencing a diagnostic region of the nuclear small subunit ribosomal RNA gene.  相似文献

The bioavailability and pharmacokinetic disposition of tiamulin in broiler chicken were investigated after administration through the crop, drinking water, and feed at 40 mg/kg body weight. Residues of tiamulin in tissues of broiler chicken were also assessed. Plasma and tissue concentrations of tiamulin were analyzed by reverse‐phase high‐performance liquid chromatography (HPLC) method. Plasma concentration–time data were described by the non‐compartmental model for all three routes, and pharmacokinetic parameters were calculated. There were no significant differences (p > 0.05) in pharmacokinetic parameters and mean plasma concentrations of tiamulin between three routes tested (crop, water, and feed), indicating equal efficacy. Tiamulin residues in edible tissues (muscles, skin, and fat) were lower than the advocated maximum residue limit (MRL of 0.1 µg/g and that of liver was 1 µg/g) on the 3rd day. No traces were found on the 5th day after drug administration. This indicated that the withdrawal period (less than 5 days) is very short, which makes it safer. This study shows that tiamulin can be used with equal efficacy through all routes of administration in broiler chicken (crop, water, and feed).  相似文献

Oxisols cover ≈ 23% of the land surface in the tropics and are utilized extensively for agricultural purposes in the tropical countries. Under the variable input types of agricultural systems practiced locally, some of these soils still appear to have problems in terms of proper soil classification and subsequently hinder attempts to implement sustainable agro‐management protocols. The definition for Oxisols in Soil Survey Staff (1999) indicates that additional input is still required to refine the definition in order to resolve some of the outstanding classification problems. Therefore, the objective of this study is to examine the properties of some Oxisols and closely related soils in order to evaluate the classification of these soils. Soils from Brazil, several countries in Africa, and Malaysia were used in this study. Field observations provided the first indication that some of the presently classified kandi‐Alfisols and kandi‐Ultisols were closer to Oxisols in terms of their properties. Water‐retention differences and apparent CEC of the subsurface horizons also supported this idea. The types of extractable Fe oxides and external specific surface areas of the clay fractions showed that many kandic horizons have surface properties that are similar to the oxic horizons. Micromorphology indicated that the genetic transition from the argillic to the oxic involves a diminishing expression of the argillic. Properties, such as CEC, become dominant. The kandic horizon is therefore inferred as a transition to the oxic horizon. It is proposed that the Oxisols be keyed out based only on the presence of an oxic horizon and an iso–soil temperature regime. The presence of a kandic horizon will be reflected at lower levels in Oxisols. The Oxisols will now be exclusive to the intertropical belt with an iso–soil temperature regime. The geographic extend of the Oxisols would increase and that of kandi‐Alfisols and Ultisols would decrease. A few kandi‐Alfisols and Ultisols in the intertropical area will have low CEC which would fail the weatherable mineral contents. The kandic subgroups of some Alfisols and Ultisols will be transitional between the low (< 16 cmolc [kg clay]–1)‐ and high (> 24 cmolc [kg clay]–1)‐activity clay soils. The proposed changes to classification will contribute to a better differentiation of the landscape units in the field. Testing of the proposed classification on some Malaysian soils showed that the new definition for Oxisols provides a better basis for the classification of the local soils and the development of meaningful soil‐management groups for plantations.  相似文献

A sensitive spectrophotometric assay has been developed for flavonoids based on their binding with molybdenum, antimony, or bismuth. Acetylation of the hydroxyl group of flavonoids abolished metal binding, thus suggesting a direct role of the hydroxyl groups. From a comparison of several related flavonoids differing in the position of hydroxyl substitutions, the hydroxyl group at position 3 was found to be an important requirement for the formation of a yellow complex. This flavonoid metal complex showed that a specific and significant bathochromic shift in the visible spectrum of the native flavonoid and the corresponding lambda(max) value was used for the colorimetric assays with different metal salts. The molybdenum complex was found to yield higher absorbance compared to antimony and bismuth complexes of various flavonoids. The present method offers a sensitive assay in the 5-25 nM range for these flavonoids and gave comparable results with HPLC quantitative determination.  相似文献
