A method was developed for the quantification of alternariol and alternariol monomethyl ether on tangerines with and without symptoms of Alternaria brown spot disease. The method employs solid-phase extraction for cleanup, followed by high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS) for detection. This method was validated on flavedo (exocarp or epicarp, exterior yellow peel) and on albedo tissue (mesocarp, interior white peel). An excellent linearity over a range of 0.50-20.0 mg/kg was achieved, with r2 >or= 0.997. The limits of detection (LOD) and quantification (LOQ) were fewer than 0.13 and 0.50 microg/kg, respectively. Cobalamin (vitamin B12) is an essential cofactor in a variety of enzymatic reactions and most prokaryotes contain transport systems to import vitamin B12. A gene coding for a periplasmic cobalamin-binding protein of Photobacterium damselae subsp. piscicida was identified by in silico analysis of sequences from a genomic library. The open reading frame was composed of 834 bp encoding a protein of 277 amino acids. The protein showed 61% identity with the vitamin B12-binding protein precursor of P. profundum , 53% identity with the corresponding protein of Vibrio parahaemolyticus and 43% identity with the periplasmic binding protein BtuF of Escherichia coli. The expression of the native protein was investigated in P. damselae subsp. piscicida , but BtuF was weakly expressed under normal conditions. To characterize the BtuF of P. damselae subsp. piscicida , the recombinant protein was expressed with a C-terminal His6-tag and purified; the molecular weight was estimated to be approximately 30 kDa. The protein does not contain any free thiol group, consistent with the view that the two cysteine residues are involved in a disulphide bond. Coastal erosion is a widespread phenomenon on sandy coasts throughout the Mediterranean region; along the Thyrrenian coast of Tuscany (Italy), stone pine (Pinus pinea L.) stands originally planted for the protection of agricultural crops further inland are often damaged. In the present study, a pairwise comparison of stands at different distance from the sea at eroded and control sites highlighted the effects of coastal erosion alone on pine growth and function. Dendroecological analyses made it possible to determine the temporal dynamics of the phenomenon since 1930 and the interactions with climate, whilst additional structural (LAI, sapwood area) and functional (carbon isotope discrimination) measurements were used to discriminate between stress mechanisms. Salty winds, exacerbated by the removal of dunal vegetation, were found to be the most likely cause of the observed growth decline. The presence and, in more recent times, the reduction of surfactants in sea water played an important synergistic effect. The intrusion of salty water in the water table, on the contrary, played a marginal role at the site. Finally, stressed trees were more sensitive to the inter-annual variability in precipitation; at all sites, growth was stimulated by June, November and December precipitation in the current and two preceding years.  相似文献

A combined serological and PCR method for the detection of Listeria monocytogenes infection in symptomatic and asymptomatic ovine flocks was evaluated. Seventy-eight milk samples and 157 serum samples were analysed using a L. monocytogenes PCR detection kit and an anti-listeriolysin O IgG immunoassay kit. The combined use of these commercial kits allowed a rapid and effective detection of L. monocytogenes infection in both the early stage, before seroconversion, and in a later phase, even after antibiotic therapy.  相似文献

Context

Forest ecosystem carbon uptake is heavily affected by increasing drought in the Mediterranean region.

Aims

The objectives of this study were to assess the capacity of a modified 3-PG model to capture temporal variation in gross primary productivity (GPP), and ecosystem net carbon uptake (NEE) in two Mediterranean forest types.

Methods

The model was upgraded from a monthly (3-PG) to a daily time step (3-PGday), and a soil water balance routine was included to better represent soil water availability. The model was evaluated against seasonal GPP and NEE dynamics from eddy covariance measurements.

Results

Simulated and measured soil water content values were congruent throughout the study period for both forest types. 3-PGday effectively described the following: GPP and NEE seasonal patterns; the transition of forest ecosystems from carbon sink to carbon source; however, the model overestimated diurnal ecosystem respiration values and failed to predict ecosystem respiration peaks.

Conclusions

The model served as a rather effective tool to represent seasonal variation in gross primary productivity, and ecosystem net carbon uptake under Mediterranean drought-prone conditions. However, its semi-empirical nature and the simplicity inherent in the original model formulation are obstacles preventing the model working well for short-term daily predictions.  相似文献

Microbial biomass (MB) produced by different industries is thought to be a beneficial supplement in fish feed due to high contents of antioxidants and pigments. However, little is known about their impact on fish health. In this experiment, 960 tilapia (26.84 ± 1.03 g) were fed one of eight experimental diets—a control diet with no MB (C), a control diet with vitamin E (VE) and six diets with three types of MB at two concentrations (0.25% and 0.5%): Rubrivivax gelatinosus (RG25 and RG50), Spirulina platensis (SP25 and SP50) and Saccharomyces cerevisiae (SC25 and SC50). Adding MB to diets decreased plasma total oxidant status, malonaldehyde and leucocyte respiratory burst; increased the total antioxidant status; and did not affect the blood biochemical parameters. In flesh, the use of the MB lowered the thiobarbituric acid‐reactive substances and increased redness (except for SC) and carotenoid deposition (except SC25). So, it was concluded that the use of the MB provided an antioxidant effect in tilapia blood plasma, decreased lipid oxidation and increased pigmentation and carotenoid deposition in the fish flesh, without imparting a negative impact on the animals’ health.  相似文献

The aim of this study was to evaluate the effects of different carotenoids sources in tilapia fish diets on the animals’ performance and fillets characteristics. Nine hundred and sixty tilapias, Oreochromis niloticus, averaging 15 g were distributed into 24 tanks to receive one of the six treatments (four repetitions) for 80 days: basal diet with no pigment (control group), basal diet with 350 mg/kg astaxanthin 10 % and basal diets added of four different concentrations of Rubrivivax gelatinosus biomass (175, 350, 700 and 1400 mg/kg). Variables analyzed included feed consumption, weight gain, feed conversion and specific growth rate for the animals and pH, proximate composition, carotenoids, polyunsaturated fatty acids and color for the fillets. Productive parameters did not differ statistically. Moisture content was lower on the fillets of treatments with pigments. The protein contents on the fillets of diets supplemented with the bacterial biomass were higher than in control group, while pH, minerals and lipids did not vary among treatments. Lightness and yellowness did not differ among the treatments, but redness and carotenoids contents were higher for all the groups that received the pigments than for the control group. The ratio of polyunsaturated fatty acids n-6/n-3 was improved with the dietary biomass. So, it was concluded that the use of the pigmenting ingredients did not alter productive parameters but increased redness and carotenoids contents in the fillets. Moreover, the use of R. gelatinosus biomass also increased the protein contents and improved the fatty acids profile in the fillets.  相似文献

Methane emission by Nellore cattle subjected to different nutritional plans   总被引:1,自引:1,他引:0  
The objective was to evaluate the emission of enteric methane by Nellore cattle subjected to different nutritional plans, as well as the intake and digestibility of nutrients from the diets supplied. Forty-seven animals in a confinement system (feedlot) were fed a corn silage-based diet for 35 days. Afterwards, these animals were evaluated in a grazing system during the rainy periods, in Urochloa brizantha cv Marandu paddocks, for 44 days. Chromic oxide was used as external marker to estimate the fecal production of animals. Samples of feces, ingredients, and pasture were collected and sampled for subsequent chemical analyses. The SF6 tracer gas technique was adopted to quantify the methane gas emitted by the animals. The experimental design was completely randomized, using procedure GLM of software SAS (9.2), including the fixed effects of sex and nutritional plan and the linear effect of the co-variable weights. Means were analyzed by Tukey’s test at 5 % probability. The intakes of total dry matter and organic matter were greater for the animals subjected to the feedlot diet (P?P?P?4/day (104.01 g) by the animals fed the feedlot diet (P?4/CEB) and methane emission per dry matter consumed were lower in relation to the grazing treatment (3.75 vs 4.23 % and 11.67 vs 15.71 g/kg). The better quality of the diet in the feedlot promoted lower energy losses as methane.  相似文献

We analyzed the response to drought of 420 individuals from eight half-sib families from a small and isolated population of silver fir (Abies alba Mill.): 105 of them were kept in well-watered conditions as control while the remaining 315 were exposed to drought for 27 days. A model describing stomatal behavior derived from Monteith and developed in beech by Leonardi et al. was fitted to experimental transpiration data obtained simply from the difference between two daily pot weighings. The estimated parameters were maximum stomatal conductance, maximum transpiration in well-watered conditions and sensitivity to soil water deficit. The model worked well: convergence for all but four individuals and concordance between experimental and fitted data were good (R(2)=0.86). Inter-individual variability for all three estimated parameters was high and two of them (maximum stomatal conductance and sensitivity to soil water deficit) were significantly different among families, suggesting genetic control. Our results validate the simplified method used to evaluate individual stomatal parameters. We also show that in the small and isolated population of our study substantial adaptive variability remains, a crucial prerequisite to endure environmental conditions determined by climatic change foreseen for the next decades.  相似文献
