We observed 15 goats for 9 days after subcutaneous infection with 10(3) TCID(50) with isolates of peste-des-petits ruminants virus from Africa and India and five concurrent, uninfected control goats. Typical clinical signs of the infection were present in all 15 infected goats by day 8 and in most by day 6 and some signs were present by day 4. However, 6 out of 15 goats already have detectable virus shedding by day 3 and four more were shedding by day 4 and every goat had virus shedding for at least 1 day before the recognition of clinical signs. This experiment indicates that incubatory carriers therefore might play a role in the transmission of PPRV among small ruminants. 相似文献

The objective of this study was to determine whether a prophylactic regimen of intramuscular polysulfated glycosaminoglycans (PSGAG), administered to growing Thoroughbred foals, would significantly reduce the incidence or severity of osteochondritis dissecans (OCD) or traumatic joint lesions that had been found in untreated foals (historical controls) from the same farm during the preceding 3 years. The number of PSGAG-treated foals with clinically significant joint lesions was 44.1% lower than the number of foals with OCD/joint injuries that had been found in the untreated historical controls. Surgical intervention of the treated foals declined 87.7% from the incidence on the farm during the preceding 3 years. Although the incidence of OCD lesions of the hock, stifle, and fetlocks in treated foals was not significantly different from that found in the matched controls, the lesions in the PSGAG-treated foals resolved without surgery. These results suggest that PSGAG has the potential to improve the clinical course of osteochondrosis (OC) and OCD. Additional studies are underway to determine whether a shorter or more targeted prophylactic regimen will provide a similar benefit. 相似文献

Port Orford cedar (Chamaecyparis lawsoniana) is endemic to northern California and southwestern Oregon and is considered a foundation species that plays critical roles in riparian areas and on nutrient‐poor soils. Since 1952, a non‐native, pathogenic oomycete (Phytophthora lateralis) has been spreading throughout the range of the cedar. Most spread occurs by vehicles carrying infested soil along gravel roads primarily used for timber harvest. In a previous study conducted in 1998 and 1999, Port Orford cedar and P. lateralis were censused in a 37‐km2 study area and dendrochronology was used to reconstruct the history of pathogen invasion. That work, which represents the only detailed analysis of spread rates for P. lateralis, showed that the first successful invasion into the study area took place in 1977 and that 43% of the susceptible host sites (stream crossings) were infested by 1999. In the work presented here, all sites that were uninfested in 1999 were re‐censused in 2012, extending the historical reconstruction of P. lateralis spread to 35 years. Two new infestations were initiated between 1999 and 2012, suggesting that the rate of spread of P. lateralis has slowed greatly. Between 1980 and 1989, the average number of new site infestations was 1.8 infestations per year, while between 1990 and 1999 the average was 0.4 infestations per year and between 2000 and 2009 the average was 0.2 infestations per year. Several potential explanations for the reduced number of new infestations are discussed. 相似文献

Two different protocols for in vitro regeneration of cassava using zygotic embryos and nodal axillary meristems have been developed. In both cases, buds were regenerated directly from excised explants without an intervening callus phase after a two-step culture procedure. In cotyledonary explants derived from zygotic embryos, prolific shoot formation occurred within 2—3 weeks on MS medium supplemented with 0.5—5 mg/1 BAP alone or in combination with 0.1 mg/1 NAA. Nodal explants with axillary meristems derived from aseptically grown seedlings or stem cuttings were used to initiate a round compact bulb-like structure on MS medium containing 10 mg/1 BAP. These latter structures, when cultured on MS medium supplemented with 0.1 mg/1 NAA, 1 mg/1 BAP and 0.1 mg/1 GA3, produced multiple shoots. Somatic embryos isolated at the globular/torpedo stage from zygotic embryo explants were also capable of multiple shoot production on medium with 1 mg/1 BAP. Rooting of regenerated shoots exceeded 95 % in phytohormone-free MS medium. No change in their ploidy levels was observed. Parklands are mixtures of trees and shrubs that farmers select for certain functions. In the Sahel, parklands are cultivated
together with staple food crops, such as millet and sorghum. Parkland trees are sources of foods, including fruits, fats,
oils, leafy vegetables, nuts and condiments that complement food crops in the local diet. Despite their socio-economic and
ecological role, many studies have shown that the parklands are degrading very rapidly. Therefore, there is a need to undertake
restoration actions that are based on a clear understanding of the livelihood context, such as the wealth status of the farmers,
in which these agroecosystems are evolving. Thus, we conducted a wealth survey with rural communities in two different farming
systems of Burkina Faso that are the cereal-based system in the “Plateau Central” and the cotton-based system in the “Boucle
du Mouhoun”. A total of twelve villages were sampled, six villages for each system. The Participatory Analysis of Poverty
and Livelihood Dynamics (PAPoLD) method was used by rural farmers to rank farmer households of their communities according
to their wealth status. The results revealed that 70% of households in the Plateau Central and 56% in the “Boucle du Mouhoun”
managed to escape poverty, and became wealthy. However, 2% of households of the villages in the Plateau Central and 6% in
“Boucle du Mouhoun” fell into poverty in the same period whereas 9 and 12% remained poor in these zones, respectively. The
main causes associated with households getting out of poverty are subsidies for cotton, external revenues from relatives working
in towns and gardening activities. Death of spouse, illness, high number of people in the household and indebtedness are the
main causes that lead into poverty. The implications of these findings in designing appropriate policies and management options
for a sustainable management of agroforestry parklands under different land use systems were finally discussed. 相似文献

Trypsin inhibitors are pathogenesis-related (PR) proteins, which play an important role in the plant defense mechanism against insects and pathogens. Peanut trypsin inhibitors are low molecular mass seed storage proteins. Like peanut allergens, they are stable to acid and heat, resistant to digestion, and can have a negative impact on human health. In peanut, five Bowman-Birk trypsin inhibitors (BBTI) have been isolated and amino acid sequences published. However, to date, no peanut BBTI sequence is available at both the cDNA and the genomic levels. The objectives of this investigation were (i) to synthesize degenerate oligonucleotides based on conserved regions of published amino acid sequences of BBTI, BII, and BIII; (ii) to isolate, sequence, and analyze at least one positive peanut trypsin inhibitor cDNA clone using the synthesized (32)P-labeled oligonucleotides as probes; and (iii) to determine its trypsin inhibitory activity. Thirty-two degenerate oligonucleotides DNA primers of 24 nucleotides each were synthesized based on the published amino acid sequences of peanut BBTI, and two were selected as probes to screen a peanut Lambda gt 11 cDNA library. Three putative positive clones were isolated, purified, and subcloned, and one was sequenced. Sequence analysis revealed a partial cDNA clone of 643 bp with a start codon. This clone shares 93 and 96% nucleotide sequence homology with peanut allergens Ara h 3 and Ara h 4 cDNA clones, respectively. A trypsin inhibitor assay revealed that peanut allergen Ara h 3 has a trypsin inhibitory activity of 11 238 TIA/mg protein.
