Extraction of a copper contaminated soil material by the percolation of an amino acid containing residue hydrolysate. 2. Time course of amino acid elution and input/output balance of amino acids During 16 days an amino acid containing blood meal hydrolysate (amino acid concentration: 188 mMol·L?1) was percolated through a column packed soil material (soil content per column: 4.1 kg dry weight, four parallels). The copper contaminated material (soil type: Typic Udifluvent, soil texture: sandy loam, loamy sand) was sampled from an area formerly used for cultivation of hop (Humulus lupus). Besides the investigation of the copper liberation the experiments aimed to determine the elution dynamic and input/output balance of amino acids (time span for amino acids balance 14 days). In total 11.7 L of hydrolysate, containing 2.2 Mol of amino acids, were introduced into each column. The mean amino acid output with the column effluent was 1.13 Mol. This corresponds to an elution degree of 51.2%, related to the sum of applicated amino acids, and to a mean substance specific elution degree of 48.4% reflecting the elution of 15 compounds. The substance specific elution ranged from 9.6% (serine) to 75.5% (valine). The highest concentrations of serine and threonine were determined in the effluents after two days, whereas the histidine concentration was highest at the last sampling. The differences in the percolation properties of the amino acids are discussed in terms of important retention and elimination processes (biodegradation, ad-/desorption, intercalation).  相似文献

The role of Pasteurella multocida serogroup F in inducing disease in rabbits was investigated in this study. Three groups of 12 Pasteurella-free rabbits each were intranasally (i.n.), subcutaneously (s.c.), and perorally (p.o.) challenged, respectively. Six rabbits of each group were immunosuppressed using dexamethasone. Eight rabbits (four of them immunosuppressed) inoculated i.n. showed symptoms of respiratory distress resulting in respiratory failure and died or were euthanized in the terminal stage of the disease 3-6 days post-infection (p.i.). The main pathological findings were fibrinopurulent pleuropneumonia (immunocompetent rabbits) or diffuse haemorrhagic pneumonia (immunosuppressed rabbits). Septicemic syndrome ending with shock occurred in 11 rabbits (6 of them immunosuppressed) inoculated s.c., which died or were euthanized in the terminal stage of the disease 2-3 days p.i. The most significant pathological findings were extensive cutaneous and subcutaneous lesions. All of the p.o. inoculated rabbits survived the challenge showing no clinical signs of the disease and no macroscopic lesions. The observations in this study indicate that in addition to serogroups A and D of P. multocida, serogroup F also can be highly pathogenic for rabbits and therefore might be a cause of considerable economic loss in commercial rabbit production.  相似文献

Browsing of overabundant free-living herbivores often limits the natural as well as artificial regeneration of forest in Central-European Mountains. The aim of the study was to assess the efficiency of the extensive reductions of herbivore populations for protection of forest regeneration in preferred areas. We analysed the relationship between the intensity of shoot browsing and relative density of herbivores in viewpoint of natural regeneration of secondary mountain spruce stands with low proportion of broadleaved trees in Králicky Sněžník Reserve (Czech Republic). The distribution of seasonally migrating herbivores was established by counting faecal pellet groups (standing crop method) in growing season. The intensity of browsing of spruce, beech and rowan was assessed using the proportion of browsed shoots on individual tree sapling. Density of red deer was re-counted from the number of pellet groups and it was 15–56 individual/km2. The intensity of spruce browsing was low in the whole area in all seasons. In general, the intensity of rowan and beech shoot browsing was high in the whole area. There were no correlations between deer density and browsing intensity of rowan (browsing was severe in all plots). We conclude that the natural regeneration of attractive trees and shrubs is nearly impossible in habitats where proportion of these food items is too small, even when the density of herbivores is low. Reduction of herbivores density is useful for protecting spruce and other tree species with low preference by herbivores. Forest managers should take into account the high palatability of deciduous tree species and distribution of herbivores on localities. Preference of localities is often based on grasses in the food supply. Regeneration of highly palatable tree species requires both low density of herbivores and sufficient protection of saplings.  相似文献

The difference in the frequency of the occurrence of reflexes/reactions in 355 cows (average weight 505.5 ± 7.9 kg) and 262 bulls (average weight 735.6 ± 8.4 kg) following stunning with a Matador SS 3,000 B trigger‐activated captive bolt gun (Termet) was determined. The stun shot more than 2 cm from the ideal position was found in 79.6% of animals. Vocalization, corneal reflex, rhythmic breathing, blinking, eyeball rotation and the absence of tongue protrusion occurred more frequently in bulls (p < .05). Spontaneous limb movements and nystagmus occurred more frequently in cows (p < .05). No difference between bulls and cows was determined in the occurrence of a response to painful stimuli or attempts to regain normal posture. No dependency was determined between the frequency of the occurrence of a reflex/reaction and the distance of the stun shot from the ideal point on the skull for any of the monitored signs. This study shows that the occurrence of reflexes/reactions following the stunning of cattle with a captive bolt is not only dependent on the position of the shot if placed within a 9 cm radius from the ideal point on the cattle skull.  相似文献

Fifty accessions of 25 Lactuca species,L. serriola ×L. sativa and Mycelismuralis were analyzed for chromosome number and relative DNA amountvariation. In the majority of Lactuca species studiedchromosome counts, as earlier reported (n = 8, 9, 17), were verified; however,for L. dregeana andL. homblei (probablyL. schweinfurthii orL. longespicata) the chromosome number(n = 9) was determined for the first time. Relative nuclear DNA content,estimated by using flow cytometry (DAPI staining), showed that 2C DNA contentranged from 2.02 pg in L.capensis to 17.96 pg inL. canadensis. Statistical and clusteranalysis of data based on relative nuclear DNA contents correspond fairly wellwith recently accepted taxonomic classification of the genusLactuca. However, the position of certain species as wellas clarification of taxonomic determinations of someLactuca accessions needs further examination.  相似文献

Three new sulfur-containing compounds were identified in Allium L. species belonging to the subgenus Melanocrommyum as the first examples of sulfur-containing pyrrole derivatives in nature. Some of these species are traditionally used in Southwest and Central Asia as vegetables and herbal drugs. A hypothetical biogenetic scheme is proposed in which L-(+)- S-(3-pyrrolyl)cysteine sulfoxide is enzymically degraded. The resulting 2-lactyl-3'-pyrrolyl sulfoxide is condensed readily to the red pigment 3,3'-dithio-2,2'-dipyrrole. All compounds are chemically unstable, rendering the analysis extremely difficult. Correlation NMR in combination with diffusion NMR allowed the identification of these low molecular weight compounds. For the first time, the compounds involved in the coloring process of Allium plant material have been identified from native plant material.  相似文献

Afew puzzles relating to a small fraction of my endeavors in the 1950s are summarized herein, with answers to a few questions of the Editor-in-Chief, to suggest that the rules of variability in time complement the rules of genetics as a biological variability in space. I advocate to replace truisms such as a relative constancy or homeostasis, that have served bioscience very well for very long. They were never intended, however, to lower a curtain of ignorance over everyday physiology. In raising these curtains, we unveil a range of dynamics, resolvable in the data collection and as-one-goes analysis by computers built into smaller and smaller devices, for a continued self-surveillance of the normal and for an individualized detection of the abnormal. The current medical art based on spotchecks interpreted by reference to a time-unqualified normal range can become a science of time series with tests relating to the individual in inferential statistical terms. This is already doable for the case of blood pressure, but eventually should become possible for many other variables interpreted today only based on the quicksand of clinical trials on groups. These ignore individual differences and hence the individual's needs. Chronomics (mapping time structures) with the major aim of quantifying normalcy by dynamic reference values for detecting earliest risk elevation, also yields the dividend of allowing molecular biology to focus on the normal as well as on the grossly abnormal.  相似文献

We tested the hypotheses that freezing-induced embolism is related to conduit diameter, and that conifers and angiosperms with conduits of equivalent diameter will exhibit similar losses of hydraulic conductivity in response to freezing. We surveyed the freeze-thaw response of conifers with a broad range of tracheid diameters by subjecting wood segments (root, stem and trunk wood) to a freeze-thaw cycle at -0.5 MPa in a centrifuge. Embolism increased as mean tracheid diameter exceeded 30 microm. Tracheids with a critical diameter greater than 43 microm were calculated to embolize in response to freezing and thawing at a xylem pressure of -0.5 MPa. To confirm that freezing-induced embolism is a function of conduit air content, we air-saturated stems of Abies lasiocarpa (Hook.) Nutt. (mean conduit diameter 13.7 +/- 0.7 microm) by pressurizing them 1 to 60 times above atmospheric pressure, prior to freezing and thawing. The air saturation method simulated the effect of increased tracheid size because the degree of super-saturation is proportional to a tracheid volume holding an equivalent amount of dissolved air at ambient pressure. Embolism increased when the dissolved air content was equivalent to a mean tracheid diameter of 30 microm at ambient air pressure. Our centrifuge and air-saturation data show that conifers are as vulnerable to freeze-thaw embolism as angiosperms with equal conduit diameter. We suggest that the hydraulic conductivity of conifer wood is maximized by increasing tracheid diameters in locations where freezing is rare. Conversely, the narrowing of tracheid diameters protects against freezing-induced embolism in cold climates.  相似文献
