Background: Selective phosphodiesterase (PDE3) inhibitors improve cardiac contractility and may use in congestive heart failure. However, their proarrhythmic potential is the most important side effect. Methods: In this research, we evaluated the potential cardiotonic activity of six new synthesized selective PDE3 inhibitors (6-hydroxy-4-methylquinolin-2(1H)-one derivatives) using the spontaneously beating atria model. In each experiment, atrium of reserpine-treated rat was isolated and the contractile and chronotropic effects of a synthesized compound were assessed. The 3-isobutyl-1-methylxanthine, a non-selective PDE inhibitor, was used for comparison. Results: The results showed that, among new compounds, the best pharmacological profile was obtained with the compound 6-[4-(4-methylpiperazine-1-yl)-4-oxobutoxy]-4-methylquinolin-2(1H)-one, C6, which displayed selectivity for increasing the force of contraction (168 +/- 5% change over the control) rather than the frequency rate (138 +/- 5% change over the control) at 300 muM. However, C6 at concentrations of 10 and 100 muM produced left and upward shift in the positive inotropic concentration-response curve of isoprenaline. The -log EC50 of isoprenaline was 8.843 +/- 0.171 in the absence, 9.448 +/- 0.138 and 9.456 +/- 0.107 in the presence of 10, 100 muM of C6, respectively (P<0.001, n = 6). Also, amrinone, a selective PDE3 inhibitor, shifted the isoprenaline concentration-response curve to the left and upward. The concentration of 10 and 100 muM amrinone decreased -log EC50 of isoprenaline to 9.527 +/- 0.287 and 9.423 +/- 0.243, respectively (P<0.001, n = 6). Moreover, the positive chronotropic effect of isoprenaline was not affected by amrinone or C6. Conclusion: This study provides functional evidence for the positive inotropic effect of C6. Considering the augmentation of isoprenaline positive inotropic concentration-response with C6 and amrinone, we conclude that C6 produces its effect via potentiation of cAMP-dependent signaling system and possibly by inhibition of PDE3 activity.  相似文献

Bio-drainage can be considered as an important part of sustainable irrigation water management. Bio-drainage has potential for managing shallow water conditions in arid and semiarid areas especially when traditional subsurface drains are not available. Bio-drainage theory does not go back too far. The relationship between soil characteristics, water management regimes, and climatic conditions is not yet well defined. This study attempted to use a mathematical model (SAHYSMOD) to evaluate factors affecting design and operation of a bio-drainage system and study its sensitivity to different variables. The study showed that the major constraint of bio-drainage is salt accumulation in tree plantation strips in arid and semiarid regions. Maximum soil water salinity which can be controlled by bio-drainage is around 3 dS m?1 in rather medium run and sustainability may only be achieved where a salt removal mechanism is considered. The study also showed that the effectiveness of the system is higher where the neighboring strips are narrower. It also showed that bio-drainage is very sensitive to the amount of applied water. While the barrier depth does not have an important effect on water table draw down, it does have a great influence on lowering the salinization rate of tree plantation strips. The application of bio-drainage could be economically controversial since in humid areas water is sufficient for agricultural crops, allocating parts of the expensive land to mostly non-fruit trees may not be feasible, while in arid and semiarid regions there is usually enough cheap land to grow trees.  相似文献

The aim of this study was to examine the effects of various levels of supplemental calcium iodate (CI) on productive performance, egg quality, blood indices and iodine (I) accumulation in the eggs in commercial laying hens. A total of 240 White Leghorn layers (Hy‐line W36) were divided through a completely randomized design into six treatments with five replicates and eight hens per each at 32 weeks of age. This experiment lasted for 12 weeks. Concentrations of I in the mash diets were 0.74, 3.13, 5.57, 8.11, 10.65 and 12.94 mg I/kg of feed in treatments 1–6 respectively. The added doses of CI were included 0.0 (control), 2.5, 5.0, 7.5, 10.0 and 12.5 mg/kg of diet for treatments 1–6 respectively. There were no significant differences in productive performance among the treatments. The highest eggshell strength was observed in group fed diet containing 3.13 mg I/kg (= .014). The highest percentage of calcium and lowest percentage of phosphorus in eggshell were observed in group fed diet containing 12.94 mg I/kg (= .0001). Feeding hens with diet containing 12.94 mg I/kg increased serum triiodothyronine‐to‐thyroxine ratio (= .0001). Serum alanine aminotransferase activity in hens fed diet containing 12.94 mg I/kg was significantly more than control (= .041). Blood Serum triglycerides in hens fed diet containing 8.11 mg I/kg were significantly higher than control (= .0001). Edible fraction of the eggs of birds fed diet containing 12.94 mg I/kg was enriched by I almost 3 times more than those fed diet containing 0.74 mg I/kg. The results suggested that egg production, egg mass, feed intake and feed conversion ratio were not significantly affected by dietary I levels. Iodine accumulation in the eggs were increased by increasing dietary I levels and the level of 10 mg/kg CI could supply I enrichment of the eggs.  相似文献

The objective of this study was to investigate the possibility of modifying the growth trajectory in Raeini Cashmere goat breed. In total, 13,193 records on live body weight collected from 4788 Raeini Cashmere goats were used. According to Akanke’s information criterion (AIC), the sing-trait random regression model included fourth-order Legendre polynomial for direct and maternal genetic effect; maternal and individual permanent environmental effect was the best model for estimating (co)variance components. The matrices of eigenvectors for (co)variances between random regression coefficients of direct additive genetic were used to calculate eigenfunctions, and different eigenvector indices were also constructed. The obtained results showed that the first eigenvalue explained 79.90% of total genetic variance. Therefore, changing the body weights applying the first eigenfunction will be obtained rapidly. Selection based on the first eigenvector will cause favorable positive genetic gains for all body weight considered from birth to 12 months of age. For modifying the growth trajectory in Raeini Cashmere goat, the selection should be based on the second eigenfunction. The second eigenvalue accounted for 14.41% of total genetic variance for body weights that is low in comparison with genetic variance explained by the first eigenvalue. The complex patterns of genetic change in growth trajectory observed under the third and fourth eigenfunction and low amount of genetic variance explained by the third and fourth eigenvalues.  相似文献

Density and resin content are two factors that have a signifi- cant effect on the production cost of wood composite. However, particle size affects resin content and density, which suggests that the interaction of these three factors can be manipulated to reduce the board density and resin content of particleboard without adversely influencing its mechani- cal properties. Some mathematical functional forms based on resin con- tent, board density and slenderness ratio were regressed and an appropri- ate form was chosen. According to analysis of the results using SHA- ZAM 9 software, the exponential function best fit the experimental data. Finally, "indifference curves" of mechanical properties were illustrated and analyzed. The results indicated that negative effects of density or resin content reduction on mechanical properties could be compensated for by controlling particles’ slenderness ratio. Interestingly, increases in slenderness ratio compensated for the negative effects of decreases in resin content or board density on module of rupture (MOR) and module of elasticity (MOE). Moreover, this "compensation ratio" intensified as resin content or density decreased and/or as the MOR or MOE increased. On the other hand, reduction in slenderness ratio indicated a comple- mentary effect on reducing internal bond (IB) strength, a result of de- creases in resin content or density. Moreover, this "complementary ratio" was intensified as resin content or density decreased and/or as IB strength increased.  相似文献

Evidence suggests that the expected increase in frequency and magnitude of extreme weather events during climate change will alter plant productivity. Therefore, extreme weather events might also be capable of changing C sequestration and allocation. Here, experimental grassland communities of two species compositions, differing in their diversity, were exposed either to a simulated single drought or to a heavy‐rainfall event. The magnitude of these manipulations imitated the local 100‐year weather extreme according to extreme‐value statistics. Effects on Net Ecosystem CO2 Exchange (NEE in µmol m–2 s–1) as well as aboveground biomass production and leaf‐area index (LAI) were recorded from prior to the manipulations until two months after the manipulations ended. Initial light utilization efficiency and maximum NEE increased after the drought. No change in the respiration was detected and maximum uptake capacity (GPPmax) was 15% higher for the drought‐manipulated plots compared to controls, which indicates an enhanced CO2 uptake into the systems. The level of diversity was also found to alter the light‐response curves, increasing respiration and maximum NEE to a higher degree than drought in the more diverse compared to the less diverse community. This resulted in an increase of GPPmax by 55%. No significant interactions between species composition and weather manipulations were detected. Interestingly, aboveground biomass production was not significantly affected by weather manipulations, even though LAI increased due to drought. This increase was caused by a decrease in the ratio between reproductive and vegetative growth. The heavy‐rainfall manipulation resulted in no significant effects. Our data suggest that C sequestration can be enhanced by a single weather event. However the importance, long‐term duration, and thresholds or turning points of such effects need to be investigated further as intensification of weather extremes is currently emerging as one of the most important facets of climate change.  相似文献

This study was done to evaluate the effects of different dietary digestible lysine (dig Lys) levels on growth performance, blood metabolites, carcass and breast yield, and breast meat quality of broilers 23–38 days of age. Three hundred 23-day-old Cobb-500 male broiler chickens were allocated to a completely randomized design with five treatments (finisher diet containing 0.88%, 0.94%, 1.00%, 1.06% and 1.12% dig Lys) and six replicates of 10 birds each. Feed intake (FI) was not affected by different dietary dig Lys levels. Weight gain (WG) linearly increased and feed conversion ratio (FCR) linearly decreased with an increasing dietary dig Lys levels. With increasing dietary dig Lys levels, carcass and breast yield and breast meat hue angle (h*) linearly increased, but abdominal fat decreased quadratically, whereas breast meat lightness (L*) and ether extract composition linearly decreased. Dietary dig Lys levels did not show any significant effect on serum metabolites, breast meat cooking loss and water holding capacity. Based on the linear broken-line regression models, the weight gain, feed conversion ratio and breast yield were optimized when dietary dig Lys levels were 0.95%, 1.01% and 1.02% respectively. It is concluded that Lys requirements vary according to what productive parameter is taken for optimization. A minimum of 1.02% dig Lys concentration in the finisher diet is suggested to optimize breast yield, feed efficiency and performance in broiler chickens.  相似文献

In this research, impact of climate change on wheat yield was simulated using SWAP in Myandoab, Iran. Field data of wheat in 2004–2005 and 2005–2006 periods were used for calibration and validation of the SWAP, respectively. The HadCM3 outputs, for A1B, A2 and B1 scenarios and 2046–2065 periods, were used in SWAP and wheat yield in mentioned period was compared with base period. Three irrigation levels, based on meeting 100, 70 and 50% of the crop ET demand, were used to study the climate change effects on wheat yield in full and deficit irrigation conditions. The results showed that temperature and reference evapotranspiration will increase in 2055s. Temperature increase cause to decrease crop growing period and crop evapotranspiration. The results showed that the negative impact of temperature increase has dominated to the positive impact of CO2 concentration and the yield is reduced. For full irrigation situation, the reduction will be 24.1, 22.4 and 20.8% in A1B, A2 and B1, respectively. Reduction in yield can be related to the shorter maturity period of the crop. The results showed that under deficit irrigation, the yield is also decreased but this decrease rate of yield is higher in full irrigation.  相似文献

  1. The estimation of sulphur amino acid requirement is a vital key to providing appropriate nutrition in poultry. The estimation of amino acid requirement depends on what production parameter is taken into consideration for optimisation.

  2. A complete randomised block design was performed with 5 treatments and 6 replicates of 8 Hy-line layers (W-36) each from 32 to 44 weeks of age. The blocks were made to have a replicate of each treatment. The dietary treatments were consisted of 5 concentrations of digestible sulphur amino acid (DSAA) at 5.1, 5.6, 6.1, 6.6 and 7.1 (g/kg).

  3. Egg production, egg mass, egg weight and feed conversion ratio (FCR) were significantly affected by an increase in DSAA intake. However, feed intake, egg component yield, Haugh unit, specific gravity, eggshell thickness, egg protein and dry matter (DM) were not altered by DSAA intake.

  4. A significant increment in plasma high-density lipoprotein was concomitant with a reduction in low-density lipoprotein when DSAA intake was increased. However, triglyceride, cholesterol, uric acid and total protein in plasma were not affected by DSAA intake.

  5. The DSAA requirements estimated by the linear broken-line model to optimise egg production, egg mass, egg weight and FCR were 678, 673, 641 and 656 mg/bird.d in the whole experimental period, respectively.

  6. The DSAA requirement estimated by the quadratic broken-line model to optimise egg production, egg mass, egg weight and FCR were 4.71%, 7.87%, 8.73% and 7.62% higher than those estimated by linear broken-line fit model in the whole experimental period, respectively.
