Objectives of this study were to compare fatty acid (FA) composition of ruminal bacterial (B) and protozoal (P) cells, and to investigate effect of protozoa on FA profile in the rumen of cattle. Three cows were used to prepare ruminal B and P cells. Four faunated and three defaunated cattle (half‐siblings) were used to study effect of protozoa on ruminal FA profile. Proportions of C16:0 and C18:0 in total fatty acids in B cells were 20.7% and 37.4%, whereas those in P cells were 33.4% and 11.6%, respectively. Proportions of trans‐vaccenic acid (VA) and cis‐9, trans‐11 conjugated linoleic acid (CLA) in B cells were 3.9% and 1.0%, and those in P cells were 5.5% and 1.6%, respectively, being higher in P cells. Proportions of C18:1, C18:2 and C18:3 in P cells were two to three times higher than in B cells. Proportions of unsaturated fatty acids, VA and CLA in B cells of faunated cattle were higher than those of defaunated. VA and CLA in the ruminal fluid of faunated were also 1.6 to 2.5 times higher than those of defaunated. This tendency was similar for cell‐free fraction of ruminal fluid. These results indicate that protozoa contribute greatly in VA and CLA production in the rumen.  相似文献

The efficiency of using monosomic alien addition lines (MAALs) to introgress agronomical traits of interest carried by wild diploid Gossypium species into the main cultivated cotton species G. hirsutum depends on the opportunities of confronting the alien chromosome with the recipient background genome at each generation and on the occurrence of translocations and homoeologous recombinations. The selfed-progeny of five MAALs of G. australe in G. hirsutum was screened with SSR markers to determine the transmission frequency of the alien chromosome and monitor its integrity. Three MAALs revealed a transmission frequency significantly lower than the expected ratio and one MAAL presented an exclusive transmission of the additional chromosome. In these four MAAL the alien chromosome was transmitted almost unaltered. With the fifth MAAL the alien chromosome was normally transmitted but was altered in half of the plants carrying it. In one MAAL, normally carrying brown fiber, the emergence of some plants carrying white and brown fiber revealed the somatic elimination of the additional chromosome. The loss of this chromosome seems to be triggered by its deletion.  相似文献

In May 2017 and 2018, Planococcus vovae (Nasonov) (Hemiptera: Pseudococcidae) was collected in Hammam Sousse (Tunisia) on the ornamental plant Cupressus macrocarpa (Cupressaceae). The infestation was observed on needles which showed symptoms of desiccation and chlorosis. This is the first record of P. vovae in Tunisia and in North Africa.  相似文献

Hypocryphalus scabricollis (Eichhoff) (Coleoptera, Curculionidae, Scolytinae) is a new pest on fig trees (Ficus carica L.). The severe damage observed on this host in Tunisia is described and illustrated. This paper describes the morphological identification of the species and gives information on the biology of the pest and initial recommendations for its control. Many research studies have shown the importance of seaweed extract (SWE) in alleviating stress damage to plants. In this study, we examined the effect of liquid SWEs made from brown seaweed, Fucus spiralis, on germination, growth, antioxidant enzymes and some biochemical attributes of durum wheat under different levels of NaCl concentrations. Application of SWE at different concentrations (5, 10, 25, and 50%) significantly enhanced seed germination and growth parameters under salt stress, especially with 25% of seaweed liquid extract. Results show that the activity of antioxidant enzymes increased with increasing the algal extract concentration to 50%. Therefore, algal treatment is proved to be an effective technique to improve the growth of wheat seedlings under salt stress conditions. This study provides important information on the identification and utilization of seaweed resources for agriculture and it is the first study to report on the uses of this macroalgae as biostimulants in agriculture.

The catabolism of arginine (Arg) by mixed rumen bacteria (B), mixed rumen protozoa (P), and their mixture (BP) was quantitatively investigated in an in vitro system in order to confirm the metabolic pathway of Arg and provide basic information for enzymatic and molecular studies as well as an understanding of the quantitative distribution of metabolites. Rumen microbial suspensions (B, P, and BP) collected from fistulated goats were anaerobically incubated with or without 1 mmol/L Arg at 39°C for 12 h. Arg and other related compounds such as citrulline (Cit), ornithine (Orn), proline (Pro) and 5‐aminovaleric acid (5AV) in both supernatant and hydrolyzates of B, P, and BP suspensions were analyzed by HPLC. The metabolic pathways of Arg in mixed rumen bacteria and mixed rumen protozoa were considered to be as follows: rumen bacteria, Arg → Cit → Orn → Pro → 5AV → VFAs + NH3; rumen protozoa, Arg → Cit → Orn → Pro → 5AV. The disappearance of Arg (1 mmol/L) was approximately 52.9 and 88.2% in B, 33.9 and 55.6% in P, and 52.8 and 85.2% in BP during 6 and 12 h incubations, respectively. When expressed in units of ‘per gram (g) of microbial nitrogen (MN)’, the net degradation rate of Arg in BP (50.3 µmol/g MN/h) was approximately 46% lower than that of B during a 12 h incubation period. The presence of protozoa tended to inhibit the production of Orn from Cit and the production of 5AV from Pro which were thought to be rate‐limiting steps of Arg metabolism in rumen microorganisms. As a result, protozoa appeared to have a saving effect on Arg metabolism, that is, protozoa protected Arg from wasteful exhaustion in the rumen.  相似文献

Background: Vitellogenin(VTG) is a precursor of egg yolk proteins synthesized within the liver of oviparous vertebrates. Betaine is an important methyl donor that is reported to improve egg production of laying hens with an unclear mechanism. In the present study, we fed betaine-supplemented diet(0.5%) to laying hens for 4 wk and investigated its effect on VTGII expression in the liver.Results: Betaine did not affect chicken weight, but significantly(P 0.05) increased egg laying rate accompanied with a significant(P 0.05) increase in hepatic concentration and plasma level of VTGI. Plasma estrogen level did not change,but the hepatic expression of estrogen receptor α(ERα) mR NA was significantly(P 0.05) up-regulated. Betaine did not affect the protein content of ERα, but significantly(P 0.05) increased hepatic expression of glucocorticoid receptor(GR)at both mR NA and protein levels. Also, ERα/GR interaction tended to be enhanced in the liver nuclear lysates of betainesupplemented hens as determined by co-immunoprecipitation. Furthermore, dietary betaine supplementation significantly increased(P 0.05) the hepatic expression of methyl-transfer enzymes, such as BHMT, GNMT, and DNMT1,which was associated with higher SAM/SAH ratio and hypomethylation of GR promoter regions.Conclusions: Betaine activates hepatic VTGII expression in association with modified DNA methylation of GR gene promoter, GR expression and ERα/GR interaction. Activation of hepatic VTGII expression may contribute, at least partly, to improved egg production in betaine-supplemented hens.  相似文献

Rust caused by Uromyces appendiculatus (Pers., Pers.) Unger is one of the major foliar diseases of common bean (Phaseolus vulgaris) in Uganda. The use of host resistance remains the best option in managing this disease. The objective of this study was to identify sources of broad-spectrum rust resistance in common bean germplasm including landraces, commercial cultivars and introduced genotypes using a combination of phenotypic and genotypic screening with 22 simple sequence repeat (SSR) markers located on chromosome Pv04. A total of 138 genotypes were field screened from 2014 and 2015 using an alpha lattice design. The variance and correlation of disease incidence, area under the disease progression curve (AUDPC) and total grain yield were computed using GenStat. The polymorphism information content of the genotypes was determined, and the association of the markers and the disease resistance traits were analyzed using PowerMarker and TASSEL respectively. Resistance of each genotype was compared to the presence and absence of amplified markers. There were highly significant differences (P < 0.001) among the genotypes for disease incidence, AUDPC and total grain yield and a strong correlation (P < 0.001) between disease incidence and AUDPC in both years. The SSR markers, BARC_PV_SSR04725, bean_ssr_0778 and bean_ssr_2892 were associated (P ≤ 0.05) with rust resistance. Fifteen 15 genotypes which included the landraces Nabufumbo, and Kapchorwa white, and the commercial cultivar NABE 2 were identified as new sources of rust resistance that would be useful in future bean breeding programmes in Uganda.  相似文献
