Evaluation of pentoxifylline and Basal Medium Eagle supplemented to diluent on cryopreserved goat spermatozoa

This study investigated the effect of pentoxifylline (PTX) and Basal Medium Eagle (BME) on frozen–thawed goat spermatozoa. Immediately after initial examination of ejaculated semen, samples were pooled and reexamined for quality. Then, samples were divided into eight equal aliquots and diluted with a basic tris-extender containing PTX (3, 6, 9 mM) and BME (5 mM) to reach a final concentration of 25 × 10⁹ and frozen. After 24 hr, the samples were individually thawed at 37°C for 30 s and evaluated for different characteristics. Obtained post-thaw results from Computer-Assisted Sperm Analysis indicate using of 3 and 6 mM PTX led significantly to an improvement in total motility, progressive motility and velocity characteristics of spermatozoa, except the beat/cross frequency (BCF) which indicated statistically no differences (p > .05) among control and treatments. Diluents prepared with BME (5 mM) and PTX alone (3 and 6 mM) improved significantly the membrane integrity–functionality, acrosome integrity and also hyaluronidase activity. Regarding recovery rate, the results showed significantly (p < .05) higher values for diluents containing 3 and 6 mM PTX compared to other groups. Malondialdehyde concentration exhibited also a significant difference (p < .05) in diluents supplemented with 5 mM BME, 3, 6 and 9 mM PTX, and mixture of 3 mM PTX and 5 mM BME which illustrate a similarity for active mitochondria, apoptotic-like and dead spermatozoa. Finally, the ratio of sperm chromatin dispersion stained spermatozoa presented significant differences (p < .05) among treatments in which the diluents added PTX alone demonstrated significantly lower values than control and extenders containing the mixtures of BME and PTX. In conclusion, the observation in this study indicates using of 3 and 6 mM PTX and BME alone may improve significantly (p < .05) the quality of cryopreserved goat spermatozoa.

Epidemiology and antibacterial susceptibility patterns of bloodstream infections, 2001-2004: an experience with BACTEC 9240 in Southern Iran

This study was conducted to determine the prevalence of bacteria recovered from bloodstream samples by Bactec 9240 at our hospital wards and to evaluate their antibacterial susceptibility patterns. During January 2001 through December 2004, 9407 referred blood samples in Bactec bottles from admitted patients at three main wards, neonates, pediatrics and adults at Nemazee Hospital, affiliated to Shiraz University of Medical Sciences in Shiraz were processed. Positive cultures were purified and identified according to standard methods. Sensitivity of bacteria to different antibiotics was determined by Kirby-Bauer disk diffusion method. Staphylococcus aureus 132(25%), Escherichia coli 64(12.1%) and Pseudomonas aeruginosa 52(9.8%) were the most pathogenic bacteria which were recovered from the blood samples. Pathogenic microorganisms were isolated from blood samples of 305 (57.8%) at pediatrics, from 181 (34.2%) at adults and from 42 (8%) at neonates wards. The highest antibiotics activities against gram positive isolates observed for vancomycin (98.4%), chloramphenicol (86.4%) and ciprofloxacin (77.4%), while in gram negative bacteria imipenem (96.1%), ciprofloxacin (83%) and amikacin (77.9%), were effective antibiotics. Frequency of isolated bacteria at pediatrics compared to adults and neonates wards were approximately two and seven folds high, respectively which indicates special attention should be paid to pediatrics patients both in prevention and treatment aspects. Vancomycin and imipenim are the effective antibiotics and could cover majority of gram positive and negative bacteria. Therefore, combined administrations of these antibiotics seems mandatory for empirical therapy.     

Soil physical properties degrade further on skid trails in the year following operations

Short-term recovery of soil physical properties on skid trails was investigated upon cessation of skidding operations and 1 year later. Bulk density and porosity were assessed at three levels of traffic intensity and two slope gradients. Compared to undisturbed areas, bulk density increased, total porosity and macroporosity decreased, and microporosity increased in the compacted areas immediately after skidding. In all cases, changes were significantly greater when traffic intensity was higher and when slopes were steeper. Surface soil compaction did not show any recovery over the 1-year period, illustrating the persistent effects of compaction on soil structure. In fact, surficial compaction further increased and macroporosity further decreased after 1 year of recovery, compared to immediately after skidding. While these changes may reflect inherent small-scale site variability or differences in soil moisture content at times of sampling, direct negative physical impacts on newly exposed soil (such as by raindrops), in addition to the loss of organic matter after canopy removal and skidding, likely delayed any physical recovery, particularly on heavily trafficked trails on steeper slopes. These results raise serious concerns about long-term resilience to traffic on forest soils and indicate the necessity to properly retire skid trails to avoid further soil degradation as trails age.     

Evaluating land degradation for assessment of land vulnerability to desert conditions in the Sokoto area,Nigeria

There are no standard indicators to evaluate or predict desertification. This paper describes an attempt to assess vulnerability to desert conditions in part of the Sokoto district in northwestern Nigeria, using remote sensing coupled with other ancillary data (erosion, sealing, crusting, compaction, cover change, organic matter monitoring, salinity and aridification) within a geographic information system environment.     

Mnemonic function of the dorsolateral prefrontal cortex in conflict-induced behavioral adjustment

Our cognitive abilities in performing tasks are influenced by experienced competition/conflict between behavioral choices. To determine the role of the anterior cingulate cortex (ACC) and dorsolateral prefrontal cortex (DLPFC) in the conflict detection-resolution process, we conducted complementary lesion and single-cell recording studies in monkeys that were resolving a conflict between two rules. We observed conflict-induced behavioral adjustment that persisted after lesions within the ACC but disappeared after lesions within the DLPFC. In the DLPFC, activity was modulated in some cells by the current conflict level and in other cells by the conflict experienced in the previous trial. These results show that the DLPFC, but not the ACC, is essential for the conflict-induced behavioral adjustment and suggest that encoding and maintenance of information about experienced conflict is mediated by the DLPFC.     

Role of selenium in mitigation of cadmium toxicity in pepper grown in hydroponic condition

Selenium (Se) is an essential element for humans but is not considered as essential for plants. However, its beneficial role in improving plant growth and stress tolerances is well established. In order to study the role of Se in cadmium (Cd) toxicity in pepper (Capsicum frutescens cv. Suryankhi Cluster), this experiment was carried out in greenhouse conditions. Treatments comprised Cd [0, 0.25, and 0.5 mM cadmium chloride (CdCl₂)] and Se [0, 3, and 7 µM sodium selenite (Na₂SeO₃)] with three replications. The result showed that Cd decreased chlorophyll a, chlorophyll b, and carotenoids, whereas Se supplementation diminished Cd toxicity on photosynthetic pigment. Selenium at 7 µM significantly increased the leaf area in the plants grown at 0.25 mM Cd. The application of Se at 3 µM with 0.25 mM Cd and Se at 3 µM and Se at 7 µM with 0.5 mM Cd increased the activity of catalase (CAT). Selenium at 7 µM decreased the proline content of pepper leaves exposed to Cd at 0.5 mM (30%). Selenium significantly enhanced the antioxidant activity of leaves, which was diminished by Cd toxicity. In general, Se has a beneficial effect on plant growth and is an antioxidant enzyme of pepper cv. Suryankhi Cluster under Cd stress and non-stress conditions.     

Studying the physiological and yield responses of sunflower inbred lines to full and limited irrigation

In order to study the physiological and yield responses of sunflower inbred lines to full and limited irrigation, an experiment was conducted in the growing season of 2014–2015 in the research field of the Agricultural Research Station in Khoi, Iran. Water withholding was imposed in the flowering stages from R4 (initial flowering) to R6 (full flowering). The results showed that the main effect of irrigation and inbred lines and the interaction effect between irrigation and inbred lines in terms of the proline and protein contents, the catalase, peroxidase, and polyphenol oxidase activities, and seed yield in all inbred lines, and relative water content(RWC) were significant at 1% level. Water withholding in the flowering stage increased the proline content and the catalase, peroxidase, and polyphenol oxidase activities, whereas limited irrigation decreased the protein content and seed yield in all inbred lines and RWC. The lowest protein content and the highest catalase, peroxidase, and polyphenol oxidase activities were observed in BGK 39 under limited irrigation condition, while BGK 37 revealed the highest proline content in such circumstances. The highest seed yield was seen in BGK 1 and BGK 375 in full irrigation condition. Limited irrigation increased the proline content by 49.51%, compared to that of full irrigation condition.     

Effect of selective cutting type on the chainsaw productivity in Caspian Forests

Time and motion studies were conducted to evaluate the impact of selective cutting type on Chainsaw productivity in Caspian forests,the selective cutting performed as single and group selection cutting in the region.The selective cutting was performed on a 42-ha tract with an average slope of 30%.The results indicate that felling time for per tree is most affected by diameter at breast height,the distance among harvested trees in single-tree selection method and diameter at breast height in group selection method.The production rate in single and group selection cutting were 21.2 m 3 h-1 and 28.4 m 3 h-1 for one person,respectively.Considering the gross and net production rate in single and group selection cutting,the unit cost was 1.11 USD m-3 and 0.88 USD m-3,respectively.The results indicate that group selection cutting can be more profitable than single-tree selection method.
