Phytoplankton blooms characterize temperate ocean margin zones in spring. We investigated the bacterioplankton response to a diatom bloom in the North Sea and observed a dynamic succession of populations at genus-level resolution. Taxonomically distinct expressions of carbohydrate-active enzymes (transporters; in particular, TonB-dependent transporters) and phosphate acquisition strategies were found, indicating that distinct populations of Bacteroidetes, Gammaproteobacteria, and Alphaproteobacteria are specialized for successive decomposition of algal-derived organic matter. Our results suggest that algal substrate availability provided a series of ecological niches in which specialized populations could bloom. This reveals how planktonic species, despite their seemingly homogeneous habitat, can evade extinction by direct competition.  相似文献

Egg yolk (EY, control) is an essential ingredient of diluents for boar semen cryopreservation. Pasteurized egg yolk (PEY) reduces hygienic risks in processing and is easier to standardize. The aim of this study was to evaluate the in vitro effect of PEY (treatment) on frozen-thawed boar semen. In a split-sample approach (n = 13 boars), it could be shown that there is neither an influence (p > .05) on post-thawing motility (PTM: 5, 30 and 120 min) nor on morphologically intact sperm, percentage of acrosome defects and membrane fluidity using a PEY extender compared to the control. Mitochondrial activity (p = .043), membrane integrity (p = .015) and PTM 300 min (p = .023) were slightly affected in the treatment group. Overall, sperm quality was at a high level in both experimental groups. Further studies are needed to determine the impact of PEY on the fertilizing capacity of boar ejaculates.  相似文献

Over the last years, several outbreaks of virulent systemic feline calicivirus (VS-FCV) infection have been described in the USA and several European countries. The paper describes two outbreaks of VS-FCV infection in cats in Germany. Data concerning clinical, laboratory, and histopathological features ofVS-FCV infection were collected from two outbreaks affecting 55 and 4 cats, respectively. Presence of feline calicivirus was confirmed by PCR followed by sequencing of the PCR-products. Clinical signs were variable, including severe upper respiratory tract infection, dyspnoea, oral and footpad ulceration, facial oedema, enteritis, pneumonia, bleeding disorder, high fever, and icterus. Both outbreaks were characterized by a high mortality rate.The present report describes the first documented outbreaks of VS-FCV infection in cats in Germany. Clinical and histopathological features are comparable to outbreaks described in the USA and Europe. However, phylogenetic analysis of the virus genome suggests that virus strains involved in these outbreaks were different from each other and from virulent strains isolated before, confirming the known genetic variability of FCV.  相似文献

By the end of the twentieth century, the forest cover over most of Europe had stabilized or was increasing after many decades of decline. Persistence and change in forest cover are driven by complex human–environmental interactions and feedback loops operating on different temporal and spatial scales. A promising method to detect these complex interactions between driving forces is a causal analysis based on historical documents. In the first step of this study, forest cover was reconstructed based on historical and contemporary maps of the Canton of Zurich at seven different points in time between 1664 and 2000. Secondly, causal chains of drivers were constructed based on historical document analysis, in order to investigate whether forest cover was stable or whether any compensating mechanisms in place were reducing the net changes. While the overall net forest cover remained considerably persistent in the Canton of Zurich throughout the 336-year study period, major gross forest cover losses and gains were detected during certain periods. Major deforestation events occurred during times of crisis, e.g., at times of economic or political crisis. In contrast, the strong persistence of net forest cover was mainly a consequence of the Forest Police Law, but can also be attributed to intensive land use. The law prohibits deforestation in general, and intensive land use is preventing the kind of natural reforestation occurring in other regions, e.g., the Swiss Alps. These empirical findings shed light on the relevance of a high degree of political and economic stability in terms of maintaining landscape persistence. These insights into the driving forces of forest cover change and persistence can contribute to protecting and managing valuable landscapes in a rapidly changing world.  相似文献

Bacterial biocontrol agents (BCAs) open up the possibility of controlling plant pathogens in an environmentally friendly way. Although they are naturally occurring microbes, some of them can cause diseases in humans. For successful registration it is necessary to test potentially adverse effects on the human health of at-risk candidates. Existing pathogenicity assays are cost-intensive, time-consuming and furthermore they are often inappropriate for facultative pathogens. We developed a new, fast and inexpensive bioassay on the basis of the nematode Caenorhabditis elegans, which is a well-accepted model organism to study bacterial pathogenicity. A selection of eight strains from clinical and environmental origin as well as potential and commercial BCAs from the genera Bacillus, Pseudomonas, Serratia and Stenotrophomonas were screened for their potential to kill the nematode in an in vitro agar plate assay. Furthermore, the motility and reproductive behaviour of nematodes exposed to strains were tested in comparison with those fed by the human pathogen Pseudomonas aeruginosa QC14-3-8 (positive control) and the negative control Escherichia coli OP50. Commercial as well as potential biocontrol strains did not display any adverse effects in all tests. In contrast, the C. elegans assay showed slight effects for clinical and environmental Stenotrophomonas strains. Results showed that the nematode C. elegans provides a model system to indicate the pathogenic potential of BCAs in a very early stage of product development.  相似文献

Background

Ridge and furrow (RIFU) systems and associated soils are a widespread legacy of medieval agriculture, are archives of historical land use, and might affect recent ecosystems. Open questions about RIFU formation and potential legacy effects still exist, especially related to physical soil properties.

Aims

Our aims were (1) to characterize the soil properties of RIFU soils and (2) to compare the drought sensitivity and the growth resistance in extremely dry years of trees growing on ridges and furrows, respectively.

Methods

We studied soil physical (bulk density, saturated soil hydraulic conductivity, and texture) and chemical (soil pH, soil organic matter, and nitrogen content) properties and the climate sensitivity of tree growth on RIFU systems for three study sites in Prignitz, Germany.

Results

RIFU systems showed a high spatial heterogeneity of soil stratigraphy due to ridge construction and increased accumulation of soil moisture and organic matter in furrows due to post-abandonment pedogenesis. Slight spatial differences in soil physical properties were found, with increased air capacity in ridge soils and higher available water contents in furrow soils. No differences in drought sensitivity were observed for trees growing on ridges and furrows, except for a wet site, where trees in furrows showed a higher sensitivity. Resistance in dry years tended to be similar or increase from furrows to ridges.

Conclusions

The results reflect a spatial differentiation of stratigraphy and post-abandonment pedogenesis on abandoned RIFU systems and suggest an adaption to different moisture conditions through RIFU construction. Differences in drought sensitivity of tree growth with relative land surface could only be detected for one of the three sites, where trees were found to be less drought sensitive on ridges.  相似文献

A 13-year-old Golden Retriever was referred for evaluation of generalized corneal edema, dyscoria, and suspected glaucoma affecting the right eye. The dog exhibited no signs of systemic illness, but had a history of a melanoma of the nail bed of the first digit of the right hind limb, excised 1 year previously. Ophthalmologic examination revealed diffuse corneal edema, mild anterior uveitis, glaucoma and a mass within the right iris. Enucleation was performed and histopathologic evaluation of the globe revealed the iris to be infiltrated with an anaplastic melanoma, suspected to represent a metastasis of the original nail bed tumor.  相似文献

Infectious hematopoietic necrosis virus (IHNV) is a significant pathogen of young salmonid fishes worldwide but particularly within the historical range of the Pacific Northwest and California. In the Sacramento and San Joaquin River drainages of California, IHNV outbreaks in juvenile Chinook salmon Oncorhynchus tshawytscha have been observed regularly at large production hatcheries, including Coleman National Fish Hatchery (established in 1941) and Feather River State Fish Hatchery (FRH; established in 1967), since facility operations began. Recent severe epidemics at the FRH in 1998 and 2000-2002 prompted investigations into the characteristics and potential sources of virus at this facility. Both phylogenetic analyses of a variable portion of the glycoprotein gene and serologic comparisons based on neutralization with three polyclonal rabbit sera were used to characterize 82 IHNV isolates from the Feather River watershed between 1969 and 2004. All isolates examined were in the L genogroup and belonged to one of three serologic groups typical of IHNV from California. The IHNV isolates from the Feather River area demonstrated a maximum nucleotide sequence divergence of 4.0%, and new isolates appeared to emerge from previous isolates rather than by the introduction of more diverse subgroups from exogenous sources. The earliest isolates examined from the watershed formed the subgroup LI, which disappeared coincidently with a temporal shift to new genetic and serologic types of the larger subgroup LII. Experimental challenges demonstrated no significant differences in the virulence for juvenile Chinook salmon and rainbow trout O. mykiss from selected isolates representing the principal types of IHNV found historically and from recent epidemics at FRH. While most isolates were equally virulent for both host species, one isolate was found to be more virulent for Chinook salmon than for rainbow trout.  相似文献

Monocytes are important mediators of inflammatory processes and are in the focus of immunological studies. While the preparation of human monocytes is widely established, little is published on the isolation of porcine monocytes for experimental studies. The aim of this study is to establish a cost efficient method of preparing and culturing porcine monocytes of considerable purity and reasonable yield. In our method, we combined and modified different protocols of human monocyte preparation. The blood of a single pig is harvested and treated with EDTA to prevent coagulation. Peripheral blood mononuclear cells are obtained by a density gradient centrifugation using a Bicoll gradient and monocytes are harvested by culturing on serum-treated culture flasks, rinsing and tapping. A high yield is obtained by constant cooling of flasks and tubes. The purity of the culture is evaluated by the expression of CD14, using flow cytometry. Using this method, we reached a purity of 92.6 % (+/? 3.06 %). With this procedure, we established a reliable method to prepare and cultivate porcine monocytes which can be performed cost effectively and does not require special equipment.  相似文献
