Real-time PCR was used to detect and quantify Verticillium dahliae and to assess the susceptibility of four Capsicum annuum cultivars (Luesia, Padrón, SCM331 and PI201234) and the Capsicum chinense cv. C118 to this pathogen. The symptoms which developed after infection included stunting and yellowing, and were more acute in the cv. SCM331, which also suffered defoliation in later stages of the disease and in C118, which suffered severe stunting. Quantification of the pathogen DNA in roots 23 and 34 days post-inoculation (dpi) revealed that there were significantly higher amounts of Verticillium dahliae DNA in C118 than in the other cultivars, followed by SCM331, Padrón and PI201234. The lowest amounts of fungal DNA in roots were found in Luesia. In hypocotyls, the highest amounts of fungal DNA were found in SCM331, while Luesia, Padrón and PI201234 had much lower amounts, and C118 had intermediate levels. When a compatible versus an incompatible system was studied, using the near-isogenic tomato lines LA3030 (susceptible) and LA3038 (resistant to V. dahliae), we were able to detect fungal DNA in both lines. As expected, the fungus/plant DNA ratio was lower in LA3038 than in LA3030 and it decreased with time in LA3038. The amount of Verticillium dahliae DNA in the roots of LA3030 remained constant between days 23 and 34 post-inoculation, but increased 10-fold in collars. Finally, when real-time PCR was applied as a diagnostic method to samples from pepper plants, soil and water collected from farms in northwest Spain, we were able to detect V. dahliae DNA in these samples even when symptoms of the disease were not evident.  相似文献

Elemental sulphur levels, sulphur localization in stems, and levels of sulphate, glutathione and cysteine were studied in pepper ( Capsicum annuum ) cvs Yolo Wonder (higher resistance) and Luesia (lower resistance) after inoculation with Verticillium dahliae , the cause of vascular wilt. Accumulation of elemental sulphur (S0) was first detected 10 days after inoculation in Yolo Wonder (mean S0 level 7·3  µ g g−1 DW), and 15 days after inoculation in Luesia (mean S0 level 3·3  µ g g−1 DW). The maximum level was reached 21 days after inoculation in Yolo Wonder (14·1  µ g g−1 DW). In control plants, elemental sulphur was not detected. SEM-EDX (scanning electron microscopy-energy dispersive X-ray microanalysis) indicated that the sulphur was not restricted to a specific location, but was dispersed throughout the vascular tissue. Sulphate levels showed a decline at the end of the experiment in inoculated plants, possibly related to the increase in sulphur levels seen in the two cultivars. The differences in sulphate levels between the two cultivars may be due to faster sulphate breakdown in cv. Yolo Wonder.  相似文献

A preliminary field study was conducted on root mass of young hybrid polar trees (Populus deltoides x P. nigra) using the electrical capacitance method. The objective of this study was to develop a simple calibration relationship that would enable root mass (fresh or dry) to be estimated for young hybrid poplar trees grown in mineral soils using simple, non-intrusive measurements of electrical capacitance. For first-year potted saplings (n = 33), strong linear and positive correlations were found between both root dry mass (adjusted r2 = 0.895, P<0.001) and root fresh mass (adjusted r2 = 0.866, P<0.001) and root electrical capacitance for the particular DN hybrid poplar clone studied. When augmented with data from third-year DN saplings grown in a more harsh landfill environment, a somewhat less strong curvilinear relationship was indicated with root dry mass (adjusted r2 = 0.780, P<0.001, n = 50). This rapid field method may be capable of providing adequate estimates of root mass for young trees in a variety of applications, such as afforestation, agroforestry or phytoremediation studies. Theoretical concepts and relationships were used to develop a deterministic pig growth model. The model predicts, in a continuous form, growth and body composition of boars, barrows, and gilts according to genotype, diet, and management conditions. The model is aggregated at the whole-animal level with three main elements of body composition; total body DNA, total body protein mass (PT), and total body mass of lipids, with PT determining the secondary elements of ash and moisture. The primary factors regulating growth were associated with cellular hyperplasia and hypertrophy in agreement with the basic concepts described by Baldwin and Black (1979). Differential equations representing DNA accretion and protein synthesis and degradation were adapted from Oltjen et al. (1985). Normal pig protein growth was characterized from published data. Body PT was used to reflect several metabolic activities related to animal size and age, as in some prior models. Dietary energy and protein were used in sequence until requirements are satisfied, first for maintenance, then for protein growth, and finally for fat deposition. A comparison between experimental and simulated results illustrates that the model may simulate growth and body composition of young pigs adequately.  相似文献

We studied in growing pigs the effects of exposure to dietary chlortetracycline on bone mineral density (BMD) and bone color. Pigs were randomly allocated to a drug-free diet (n = 48) or a diet fortified with 800 ppm of chlortetracycline, starting either at 28- or 84-d of age, and for either a 28- or 56-d duration (n = 16 pigs/group). The lumbar vertebral discoloration and BMD of randomly chosen pigs were evaluated at 28-d intervals up to 168-d of age. The odds of bone discoloration increased with dosing duration and age at treatment onset, and decreased with the withdrawal time and age at treatment onset interaction (p ? 0.001). The measured trabecular BMD linearly increased with age and squared treatment duration (p ? 0.005). Therefore, TC-induced bone discoloration is reversible, and may be prevented with proper dosing regimen design. Moreover, TC induces a persistent increase on BMD that could be detected with quantitative computed tomography.  相似文献

In Southern Italy the paucity of water is a very important problem and due to the fundamental importance of the water for the processing tomato, in this study the influence of irrigation on processing tomato quality was investigated in the years 2002 and 2003. A hybrid processing tomato (Lycopersicon esculentum L. Mill cv. Ability) was treated with different water regimes obtained combining amount of water and irrigation interval. The effects of the trials were evaluated taking into account the physical and chemical characteristics of the fruits, as well as the content in antioxidants moieties. Furthermore, the relationship among all the quality parameters and the seasonal amount of irrigation water was evaluated.The results, comparing different water regimes obtained combining amount of water and irrigation interval, gave useful indications on the possibility to improve nutritional tomato quality by reducing irrigation water during tomato cropping. In particular, relatively to the Mediterranean areas of tomato cultivation and to the texture of the soil considered in this study, the best compromise between quality and quantity of the processing tomato fruit was achieved both with the extension of the irrigation interval (L40 or L60) and with the limitation of irrigation volume for the second part of the tomato crop cycle (thesis 100-50). Extending the irrigation interval and limiting irrigation volume for the second part of the tomato crop cycle, appeared to be the best management strategy to optimize the yield and nutritional quality of processing tomato.  相似文献

Ninety-six pigs from Large White (LW), Synthetic Genex 3000 (SG) and Meishan-derived dam line (M) genetic lines, each with a mean live body weight (BW) of 20.7 ± 4.2 kg, were used to investigate the relationship that exist between production performance and meat quality parameters for these three genotypes. The animals were assigned to pens in groups of eight and slaughtered at 109.6 ± 3.78 kg BW. At the end of the trial, the LW pigs had the highest (P < 0.001) average daily feed intake and average daily gain and protein deposition rates, the M pigs showed (P < 0.001) the worst feed conversion, the highest daily backfat gain and the lowest residual energy intake, and the SG pigs had the lowest (P < 0.001) daily fat deposition rate. The longissimus muscle of the M pigs had higher a (P < 0.01) and b (P < 0.05) values than the LW pigs, with the SG pigs in between. The M loins also had the highest (P < 0.01) shear force value, which may be explained by the lower (P < 0.01) soluble collagen content. In spite of their lowest fat deposition rate, the highest (P < 0.001) intramuscular fat content was measured in the SG loins, and the highest (P < 0.01) protein content was found in the M loins. Eight canonical correlations were obtained between performance and meat quality data, with the first three correlation coefficients of 0.87, 0.66 and 0.64 being significant. Performance and meat quality data were related to a certain extent. Pigs with a higher average daily feed intake also had a higher average daily gain, an average protein deposition rate, residual energy intake and gain-to-feed ratio, and lower meat dry matter, intramuscular fat, a values and pigment content. Body water content seems to be higher in fast-growing pigs. Furthermore, fast-growing pigs also have lower intramuscular fat, a values and pigment content. However, there is some indication that the magnitude of these correlations can be breed-dependent. The differences among the studied genotypes are much higher in terms of growth performance than in terms of meat quality traits.  相似文献

Ten 56-d-old, 15-kg barrows were surgically fitted with a postvalvular T-cecum cannula at the ileo-cecal junction to evaluate the effect of microbial phytase on apparent and true ileal AA digestibility and N utilization. A semipurified cornstarch- and soybean meal-based diet was formulated to contain 3.4 Mcal of DE/kg, 17.0% CP, 0.8% Ca, and 0.6% P but had a low phytate-P concentration (0.13%; all on an as-fed basis). Chromic oxide and dysprosium chloride were used as indigestible markers. The basal diet was supplemented with 0 or 1,000 phytase units/kg of microbial phytase. Postprandial plasma urea N and alpha-amino N concentrations, excretion of Ca, P, and N in feces and urine, and ileal AA digestibilities were determined 3 times at 4-wk intervals beginning at 70 d of age. The homoarginine (HA) method was used to determine endogenous AA flow by replacing 50% of the basal protein with guanidinated protein. Microbial phytase had no effect on apparent ileal digestibility (AID) or on true ileal digestibilities of N and most AA but did increase AID for arginine (P = 0.006) and methionine (P = 0.037). However, in HA diets, phytase increased the AID of CP (P = 0.01) and several AA. Addition of microbial phytase had no effect on the postprandial alpha-amino N concentrations in plasma but increased overall plasma urea N concentrations (P = 0.035). Barrows fed phytase-supplemented diets had decreased P in feces (P = 0.003) and greater P in urine (P = 0.001) but comparable total P excretion compared with barrows fed no phytase-supplemented diets. In conclusion, the addition of phytase to a semi-purified soybean meal-based diet did not affect the AID of several AA. In addition, differences between the basal and HA diets in N digestibilities indicated that that guanidination may limit the use of the HA method in determining endogenous protein losses.  相似文献

The Cracidae family of neotropical birds is regarded as one of the most severely threatened in the world. They traditionally have been extensively hunted, and, thus, ex situ efforts for their conservation are recommended and involve the optimization of their care in captivity. Nutrition is a fundamental aspect of husbandry, which influences survival and reproduction in captivity. In this study, a total of 29 animals, including 3 species (Penelope obscura, Penelope superciliaris, and Aburria jacutinga), were subjected to monthly physical examination and blood sampling before and after dietary conversion from the traditional diet of broiler feed, fruits, and vegetables to a nutritionally balanced commercial diet specifically designed for wild Galliformes. The diet change produced differences in several parameters tested, including an increase (P < 0.05) in hemoglobin concentration for all species. Increases (P < 0.05) in erythrocyte count, packed cell volume, and body weight were observed in P. obscura, with a concomitant decrease in the standard deviation for such parameters that show improved uniformity. Globulins and lipase also were reduced (P < 0.05) in P. obscura. Although leukocyte count was lowered and eosinophils were increased in all 3 species after dietary conversion, only these 2 changes were significant (P < 0.05) in P. superciliaris. A. jacutinga had higher (P < 0.05) blood glucose concentrations than the other species, but diet had no effect on this parameter. Blood uric acid concentrations were higher (P < 0.05) after conversion to the commercial diet in P superciliaris. The provision of a commercial extruded diet as a single food source was beneficial, which led to a general improvement in clinical aspects and group uniformity in these 3 species of Cracidae.  相似文献

Phosphorus (P) digestibility represents complex digestive and metabolic phenomena which are embodied in a unique coefficient. A dynamic mathematical model, based on mechanistic concepts is developed to improve pig feed formulation systems. In this first step, it simulates the fate of different forms of dietary P in the stomach. The model also accounts for dietary calcium (Ca) content, gastric transit time and pH. It simulates consistently the inorganic P released at the exit end of the stomach. However, this model has to be challenged by external data.  相似文献
