The endotoxic activity of Fusobacterium necrophorum bov 5 was investigated. The supernatant (S) fluid and cell wall (CW) preparation, obtained after differential centrifugation of the ruptured cell mass, were lethal for mice. The toxicity of the S fluid was stable during prolonged storage, treatment with formalin, and heating for 15 minutes at 80, 100, and 121 C, but was destroyed by alkaline hydrolysis with 0.25 N NaOH. The toxic factor was found in a high molecular weight (MW) fraction after gel filtration. The properties exhibited by the toxic S fluid resembled those of endotoxic lipopolysaccharide (LPS). Extracted and partially purified LPS (endotoxin) from F necrophorum bov 5 demonstrated a mouse median lethal dose (mouse LD50) of 16.8 mg/kg of body weight. The toxic LPS material, a high molecular weight moiety as estimated by gel filtration, was resistant to ribonuclease (RNase), deoxyribonuclease (DNase), and pronase treatment. A positive Shwartzman reaction (median skin lesion dose (SLD50) equal to 3.32 mug/kg of body weight) and biphasic fever response (minimal dose required to produce a fever index of 40 sq cm which falls on the linear portion of dose-response curve (FL40) equal to 0.41 mug/kg of body weight) further indicated the toxin was endotoxin in nature. The LPS from F necrophorum bov 5 was less toxic than Salmonella typhimurium LPS; but had considerable toxicity for experimental animals. The toxic activity of the partially purified F necrophorum bov 5 endotoxin was separated into 2 fraction regions by diethylaminoethyl (DEAE)-cellulose chromatography. The data provide evidence for the production of a potent endotoxin, possibly composed of more than one toxic component, which may be released upon cell disruption.  相似文献

OBJECTIVE: To evaluate 3 methods for measuring urine bile acids (UBA) and compare their diagnostic performance with that of the serum bile acids (SBA) test and other routine screening tests in dogs with hepatic disorders. DESIGN: Prospective study. ANIMALS: 15 healthy dogs, 102 dogs with hepatic disorders, and 9 dogs with clinical signs of hepatic disorders that were found to have nonhepatic disorders. PROCEDURES: Blood and urine samples were collected from sick dogs and healthy dogs for serum biochemical analyses, and determination of concentrations of SBA and UBA. Urine samples were obtained from 15 healthy dogs to establish an upper cutoff value for UBA concentrations. The UBA were measured by use of a quantitative-linked enzymatic colorimetric method. Three analytical modifications were evaluated; 1 quantified only urine sulfated bile acids (USBA), 1 only urine nonsulfated bile acids (UNSBA), and 1 quantified both (USBA plus UNSBA). The UBA values were standardized with the urine creatinine concentration. RESULTS: The UNSBA-to-creatinine ratio and USBA plus UNSBA-to-creatinine ratio tests had the best diagnostic performance of the UBA tests; each had a substantially higher specificity, slightly higher positive predictive value, slightly lower negative predictive value, and lower sensitivity than the SBA test. These UBA-to-creatinine values were positively correlated with SBA values. The USBA-to-creatinine ratio had poor sensitivity, indicating a low rate of bile acid sulfation in dogs. CONCLUSIONS AND CLINICAL RELEVANCE: The UBA can be measured in dogs with sufficient repeatability and accuracy for clinical application. The UNSBA-to-creatinine ratio and USBA plus UNSBA-to-creatinine ratio identified dogs with hepatic disorders nearly as well as the SBA test.  相似文献

Immunity in Taeniids is predominantly antibody mediated and thus many serological immuno-determinants will have potential in both protection and diagnosis. The antigenicity of six peptides derived from four potentially protective molecules cloned from a Taenia saginata oncospheres cDNA library have been evaluated as targets for the specific diagnosis of bovine cysticercosis. The six peptides consist of: two peptides (HP6-2 and HP6-3) derived from the sequence of the 18 kDa surface/secreted oncospheral adhesion antigen identified by McAb-HP6, two peptides (Ts45W-1 and Ts45W-5) derived from the sequence of the T. saginata homologue of the T. ovis 45W protective gene family, one peptide (TS45S-10) derived from a T. saginata sequence with significant similarity to the T. ovis 45S protective antigen, and one peptide (TEG-1) derived from the sequence of the T. saginata homologue of Echinococcus spp. main surface protein. Longitudinal studies indicate that T. saginata infected cattle respond to all six peptides by 3-4 weeks post-infection and that the antibody levels remain high for at least 12 weeks post-infection. As protection against Taeniid parasites is predominantly antibody mediated, some of these six peptides may be of value as immuno-prophylactic tools and hence also in assays to determine resistance to infection with the parasite. For diagnosis, on the other hand, only three peptides (HP6-2, TEG-1 and Ts45S-10) performed with the necessary sensitivity and specificity to determine exposure to infection with T. saginata, and now merit an exhaustive evaluation prior to employment as routine diagnostic tools.  相似文献

Access systems are a necessary element of resource production in bottomland hardwood sites. However, road building may have a detrimental effect on hydrologic function of the site. This report describes initial results of a study designed to examine the effect of different road surfacing treatments on water quality.

Four surfacing treatments installed on two test roads included native soil, native soil with vegetative stabilization, 6 cm of gravel, and 15 cm of gravel over geotextile. During the first flooding season periodic sampling measured floodwater suspended sediments and location of erosion and sediment deposition within the road prism. Initial results suggest that sediment movement was confined to the road right-of-way, with no statistically significant sedimentation effects detected beyond the clearing limits of the road. As a system of land use which entails the deliberate association of trees with herbaceous field crops in time, shifting cultivation is one of the most ancient, widespread and, until recently, ecologically stable forms of agroforestry. However, under pressure of population and competing uses for land and labour, traditional swidden systems have been observed historically to undergo more or less predictable processes of intensification. Since shifting cultivation is an indigenous form of agroforestry, scientific agroforestry is not, strictly speaking, an alternative to shifting cultivation, but rather a systematic approach to the recombination of its basic elements into more intensive, sustainable and politically viable forms of land use, whenever pressures signal the need for change in traditional swidden systems.Different agroforestry options open up from different stages of intensification in swidden systems. A review of evolutionary typologies of shifting cultivation gives rise to a framework for the identification of agroforestry interventions and development pathways appropriate to specific systems. technological proposals are limited to a short list of the most promising agroforestry interventions in main sequence swidden systems. These include integral taungya, economically and biologically enriched fallows, variations on the alley cropping theme, and various tree crop alternatives to annual cropping systems. Examples and quantitative data are cited to substantiate the main hypotheses behind the proposals.  相似文献

Abstract

In this study, water content in black spruce (Picea mariana Mill.) and aspen (Populus tremuloides Michx.) sapwood samples was investigated with time-domain magnetic resonance (MR). Time-domain MR measurements easily distinguish water in different environments in wood according to the spin-spin relaxation time and provide quantitative information on water content. The MR techniques employed can distinguish and quantify the individual signal components. Both black spruce and aspen have two signal components at moisture contents above the fiber saturation point. These two signal components correspond to motionally restricted water, often referred to as bound water, and unrestricted, or free water. Bound water content is constant above 40% moisture content. No signal from free water was detected at or below 20% moisture content in either species. We also demonstrate the use of a recently developed portable unilateral magnet that can be employed as a powerful tool in the study and measurement of water content in wood.  相似文献

Many new in vitro methods have been developed to evaluate antioxidant activity. Unfortunately, these in vitro methods often correlate poorly with the ability of compounds to inhibit oxidative deterioration of foods because the in vitro assays do not account for factors such as the physical location of the antioxidant, its interaction with other food components, and environmental conditions. To accurately evaluate the potential of antioxidants in foods, models must be developed that have the chemical, physical, and environmental conditions expected in food products. This paper outlines model systems of the evaluation of antioxidants in three types of foods: bulk oil, oil-in-water emulsions, and muscle foods. These model systems are not intended to be inclusive of all possible methods to measure lipid oxidation and antioxidant activity. However, use of these models would allow researchers to more easily compare research results from one paper to another.  相似文献
