Profilin is generally thought to regulate actin polymerization, but the observation that acidic phospholipids dissociate the complex of profilin and actin raised the possibility that profilin might also regulate lipid metabolism. Profilin isolated from platelets binds with high affinity to small clusters of phosphatidylinositol 4,5-bisphosphate (PIP2) molecules in micelles and also in bilayers with other phospholipids. The molar ratio of the complex of profilin with PIP2 is 1:7 in micelles of pure PIP2 and 1:5 in bilayers composed largely of other phospholipids. Profilin competes efficiently with platelet cytosolic phosphoinositide-specific phospholipase C for interaction with the PIP2 substrate and thereby inhibits PIP2 hydrolysis by this enzyme. The cellular concentrations and binding characteristics of these molecules are consistent with profilin being a negative regulator of the phosphoinositide signaling pathway in addition to its established function as an inhibitor of actin polymerization.  相似文献

The effects of inbreeding in livestock species breeds have been well documented and they have a negative impact on profitability. The objective of this study was to evaluate the levels of inbreeding in Sarda (SAR, n = 785) and Valle del Belice (VdB, n = 473) dairy sheep breeds and their impact on milk production traits. Two inbreeding coefficients (F) were estimated: using pedigree (FPED), or runs of homozygosity (ROH; FROH) at different minimum ROH lengths and different ROH classes. After the quality control, 38,779 single nucleotide polymorphisms remained for further analyses. A mixed-linear model was used to evaluate the impact of inbreeding coefficients on production traits within each breed. VdB showed higher inbreeding coefficients compared to SAR, with both breeds showing lower estimates as the minimum ROH length increased. Significant inbreeding depression was found only for milk yield, with a loss of around 7 g/day (for SAR) and 9 g/day (VdB) for a 1% increase of FROH. The present study confirms how the use of genomic information can be used to manage intra-breed diversity and to calculate the effects of inbreeding on phenotypic traits.  相似文献

The objectives of this work were to investigate alternative selection criteria for milk yield traits in the Valle del Belice sheep breed, which are either corrected for lactation length or not affected by lactation length, to estimate genetic parameters for these alternative selection criteria and to compare the estimated breeding values. The genetic correlations show that corrected milk yield (CMY), maximum test-day yield (MTY) and milk yield at hundred days (MYH), are moderately or weakly correlated with lactation length (LL) (rg = 0.58, 0.16 and 0.39, respectively). Higher genetic correlation was found between total milk yield (TMY) and LL (rg = 0.73). Rank correlations between selection criteria for estimated breeding values for the entire data set were above 0.90 for CMY vs. TMY and CMY vs. MYH and were similar for rams and ewes. Very low were the rank correlations for LL vs. MTY and LL vs. MYH in comparison with 0.75 for LL vs. TMY. Under high selection intensity, rank correlations between breeding values from CMY vs. TMY, CMY vs. MYH and MTY vs. MYH were lower, ranging from 0.53 to 0.75, but higher than all other contrasts between selection criteria. The general results obtained in this study show that MYH is a selection criterion that could improve the genetic evaluation in the Valle del Belice dairy sheep.  相似文献

A time-to-event study for mastitis at first-lactation in Valle del Belice ewes was conducted, using survival analysis with an animal model. The goals were to evaluate the effect of lambing season and level of milk production on the time from lambing to the day when a ewe experienced a test-day with a recorded SCC greater than or equal to 750,000 cells/ml, and to estimate, for this trait, its heritability and the percentage of variation explained by the flock-year of lambing effect. A dataset with 2468 first-lactation records, collected from 1998 to 2003 in Valle del Belice ewes allocated in 17 flocks, was used. The Cox model used included lambing season and total milk yield adjusted for lactation length as fixed effects and flock-year of lambing effect and individual additive genetic effect as random effects. In total 40.5% of the records were censored. Results indicated that ewes lambing from April to July were at a higher risk of mastitis than those lambing from August to November (conventional season), and that ewes in the highest class of milk production were at a higher risk of mastitis than those in the lowest level. The heritability for the time interval between lambing and first test-day with mastitis was 3% on the logarithmic scale and 4% on the real scale. The proportion of variation, in the time interval between lambing and first test-day with mastitis, explained by the flock-year of lambing effect was 19% on the logarithmic scale and 27% on the real scale; this seems to stress the importance of flock management.  相似文献

The aim of this paper was to characterize the diversity among haplotypes based on 22 single nucleotide polymorphisms (SNPs) and one deletion within four casein genes in two geographically distant goat populations, the Sicilian Girgentana breed and the Norwegian goat breed. Forty Girgentana goats were genotyped for the aforementioned polymorphisms and the resulting data set was compared with 436 goats from the Norwegian population previously genotyped for these markers. Several casein gene polymorphisms were not in Hardy-Weinberg equilibrium either in Girgentana, or in the Norwegian breed. The SNP haplotype frequencies for the four casein genes were calculated and despite the large geographical distance and phenotypic divergence between these breeds, a proportion of casein loci haplotypes were found to be identical between both Norwegian and Girgentana goats. However, for the CSN2 gene there were no common haplotypes between the two populations. The level of linkage disequilibrium between the casein genes was less in the Girgentana population than in the Norwegian population.  相似文献

Selective breeding has led to modifications in the genome of many livestock breeds. In this study, we identified the genomic regions that may explain some of the phenotypic differences between two closely related breeds from Sardinia. A total of 44 animals, 20 Sardinian Ancestral Black (SAB) and 24 Sardinian White (SW), were genotyped using the Illumina Ovine 50K array. A total of 68, 38 and 15 significant markers were identified using the case–control genome‐wide association study (GWAS), the Bayesian population differentiation analysis (FST) and the Rsb metric, respectively. Comparisons among the approaches revealed a total of 22 overlapping markers between GWAS and FST and one marker between GWAS and Rsb. Three markers detected by Rsb were also located near (<2 Mb) to highly significant regions identified by GWAS and FST analyses. Moreover, one candidate marker identified by GWAS and FST approaches was located in a run of homozygosity island that was shared by both breeds. We identified several genes involved in many phenotypic differences (such as stature and growth, reproduction, ear size, coat colour, behaviour) between the two analysed breeds. This study shows that combining several genome‐wide approaches could improve discovery of regions involved in the variability of breeding traits and responsible for the phenotypic diversity even between closely related breeds. Overall, the combination of such genome‐wide methods can be extended to other livestock breeds that share between them a similar genetic background, to understand the process that shapes the patterns of genetic variability between closely related populations.  相似文献

High aquacultural rearing density and handling of fish may frequently result in skin or gills wounds, thereby facilitating the onset of secondary infections. The capacity of the zebrafish to regenerate tissues, as well as fins and other organs, makes it an ideal animal model for studying the mechanisms of tissue regeneration. Since macrophages are involved in tissue regeneration, a diet including ß‐glucans might positively affect the process through activation of macrophages and other immune pathways. Consequently, the aim of this study was to investigate the effects of the oral administration of 1,3‐1,6 β‐glucans on the regeneration process of the caudal fin after its amputation in zebrafish. One hundred and twenty zebrafish were randomly distributed into four groups with three replicates each: an untreated non‐amputated group (CNA) and an untreated amputated group (CA) fed a control diet; two treated and amputated groups (MI and MII) fed for 14 days the same diet with the addition of two differently extracted 1,3‐1,6 ß‐glucans (MacroGard® and Experimental MacroGard®, Biorigin©). ß‐glucans were added to allowed the administration of 12.5 mg/kg of fish body weight (0.35 g/kg of feed). Results showed that 1,3‐1,6 ß‐glucans decreased fish mortality rate and enhanced both daily and cumulative regenerated fin area, independent of the specific ß‐glucan extraction method used. Based on the mechanisms similarities of the innate immune system and tissue regeneration among different teleost species, these results may likely be extended to species of interest for the aquaculture sector.  相似文献
