Canine histiocytic sarcoma (HS) is an aggressive tumor type originating from histiocytic
cell lineages. This disease is characterized by poor response to chemotherapy and short
survival time. Therefore, it is of critical importance to identify and develop effective
antitumor drugs against HS. The objectives of this study were to examine the drug
sensitivities of 10 antitumor drugs. Using a real-time RT-PCR system, the mRNA expression
levels of 16 genes related to drug resistance in 4 canine HS cell lines established from
dogs with disseminated HS were determined and compared to 2 canine lymphoma cell lines
(B-cell and T-cell). These 4 canine HS cell lines showed sensitivities toward microtubule
inhibitors (vincristine, vinblastine and paclitaxel), comparable to those in the canine
B-cell lymphoma cell line. Moreover, it was shown that P-gp in the HS cell lines used in
this study did not have enough function to efflux its substrate. Sensitivities to
melphalan, nimustine, methotrexate, cytarabine, doxorubicin and etoposide were lower in
the 4 HS cell lines than in the 2 canine lymphoma cell lines. The data obtained in this
study using cultured cell lines could prove helpful in the developing of advanced and
effective chemotherapies for treating dogs that are suffering from HS. 相似文献

Contrast-enhanced ultrasound using Sonazoid, a novel contrast medium with a liver-specific Kupffer phase, was evaluated in canine focal liver lesions Twenty-five dogs with a liver mass were given intravenous Sonazoid, and the enhancement pattern in the arterial, portal, and parenchymal phase was characterized. An enhancement defect in the lesion in the parenchymal phase was observed in all malignant lesions, whereas only one of nine benign lesions had a filling defect. The diagnostic value of the presence of a filling defect for malignancy was statistically significant (100% sensitivity, 88.9% specificity, 94.1% positive predictive value, 100% negative predictive value), and was equal to that of hypoenhancement in the portal or delayed phase. The defect pattern (clear or irregular defect) was dependent ( P <0.05) on the types of malignancy (i.e., hepatocellular carcinoma and other types of malignancies). In the arterial phase, five of the six hepatocellular carcinomas had hypervascularity, whereas no other lesion was characterized by hypervascularity. In some dogs, additional lesions that could not be observed with conventional B-mode ultrasonography were detected in the parenchymal phase. The enhancement pattern of Sonazoid, especially in the parenchymal phase, has potential as a diagnostic tool for canine focal liver lesions. 相似文献

Nasal lymphoma (NL) is the most common nasal tumor in cats, and radiotherapy, chemotherapy, or a combination of these treatments have been described as the treatment for this disease. However, the previous studies included various machines and protocols of radiotherapy. Therefore, we aimed to retrospectively compare the prognosis among cases treated with palliative hypofractionated radiotherapy, chemotherapy, and a combination of them with united machine and protocol of radiotherapy. When compared overall survival and progression free survival, there was no significant difference among these three groups. The data of this study suggested that similar efficacy could be achieved by palliative hypofractionated radiotherapy, chemotherapy, or a combination of them. 相似文献

A Pomeranian dog, 1 year- and 8 month-old neutered female, was presented with persistent
respiratory distress and recurrent generalized demodicosis. Physical examination revealed
cyanosis, rough respiratory sounds, multifocal alopecia and dermal erosions on the dorsal
side of the forelimbs, perineal area and skin around the eyes. A severe diffuse
interstitial lung pattern was observed on thoracic radiographs. The blood examination
revealed neutrophilia and hypoglobulinemia. Serum immunoglobulin concentrations of IgG and
IgA were low. Histopathological examination revealed severe diffuse interstitial pneumonia
with Pneumocystis carinii infection. Severe lymphoid depletion was
observed in the spleen and other organs with lymphoid follicles consisted mainly of
CD3-positive T cells and few cells of B-cell lineage. B-cell hypoplasia with subsequent

Solid state13C nuclear magnetic resonance(NMR)spectroscopy is a common tool to study the structure of soil humic fractions;however,knowledge regarding carbon structural relationships in humic fractions is limited.In this study,mobile humic acid(MHA)and recalcitrant calcium humate(CaHA)fractions were extracted from eight soils collected from six US states and representing a variety of soils and ecoregions,characterized by this spectroscopic technique and analyzed for statistical significance at P≤0.05.We found that the abundances of COO and N–C=O functional groups in the MHA fractions were negatively correlated to soil sand content,but were positively correlated to silt,total N and soil organic carbon contents.In contrast,the abundances of the COO and N–C=O functional groups were only positively correlated to the content of clay in the CaHA fractions,indicating that the two humic fractions were associated with diferent soil components.The two13C NMR peaks representing alkyls and OCH3/NCH were negatively correlated to the peaks representing aromatics,aromatic C–O and N–C=O/COO.Comparison of the sets of data from13C NMR spectroscopy and ultrahigh resolution mass spectrometry revealed that the aromatic components identified by the two methods were highly consistent.The comparison further revealed that protein in MHA was associated with,or bound to,the nonpolar alkyl groups,but a component competitively against(or complementary to)aromatic groups in the MHA composition.These observations provided insight on the internal correlations of the functional groups of soil humic fractions. 相似文献

Protein-losing enteropathy (PLE) is known to induce hypercoagulability and resultant thromboembolism in dogs. We hypothesized that hypercoagulability would improve if remission was obtained in dogs with PLE after treatment. This study aimed to evaluate the changes in the coagulation parameters after treatment in dogs diagnosed with PLE. As coagulation parameters, prothrombin time (PT), activated partial thromboplastin time (aPTT), fibrinogen, thrombin-antithrombin complex (TAT), D-dimer, and antithrombin (AT) were measured. In addition to these parameters, rotational thromboelastometry (ROTEM), which evaluates the comprehensive coagulation and fibrinolysis reactions of whole blood, was conducted and the data of clotting time (CT), clot formation time (CFT), α angle (α), maximum clot firmness (MCF) and lysis index at 60 min (LI60) were obtained. Eleven of the 14 dogs diagnosed with PLE were classified as responders to the treatment based on the changes in their plasma albumin (ALB) concentration after treatment. Significant increase in CFT and decrease of α and MCF indicating the resolution of hypercoagulability were found after treatment in responder dogs; however, there was no significant change in the coagulation and fibrinolysis parameters other than those measured by ROTEM. This study demonstrated that the hypercoagulability detected by ROTEM was significantly improved after treatment in dogs with PLE. 相似文献
