Orf virus (ORFV), a member of parapoxvirus, is an enveloped virus with genome
of double-stranded DNA. ORFV causes contagious pustular dermatitis or contagious ecthyma
in sheep and goats worldwide. In general, detection of viral DNA and observing ORFV virion
in tissues of afflicted animals are two methods commonly used for diagnosis of orf
infection; however, isolation of the ORFV in cell culture using virus-containing tissue as
inoculum is known to be difficult. In this work, the ORFV (Hoping strain) isolated in
central Taiwan was successfully grown in cell culture. We further examined the biochemical
characteristic of our isolate, including viral genotyping, viral mRNA and protein
expression. By electron microscopy, one unique form of viral particle from ORFV infected
cellular lysate was demonstrated in the negative-stained field. Moreover, immunomodulating
and anti-influenza virus properties of this ORFV were investigated. ORFV stimulated human
monocytes (THP-1) secreting proinflammatory cytokines IL-8 and TNF-α. And, pre-treatment
of ORFV-infected cell medium prevents A549 cells from subsequent type A influenza virus
(IAV) infection. Similarly, mice infected with ORFV via both intramuscular and
subcutaneous routes at two days prior to IAV infection significantly decreased the
replication of IAV. In summary, the results of a current study indicated our Hoping strain
harbors the immune modulator property; with such a bio-adjuvanticity, we further proved
that pre-exposure of ORFV protects animals from subsequent IAV infection. 相似文献

The discrimination between wild and released Taiwan abalone Haliotis diversicolor was performed by stable carbon isotope analysis. Abalone samples were collected from Mao Aw Gulf in north-eastern Taiwan. Live abalone and dead shells were collected by divers with an 80 × 80 cm frame. The densities and amounts of wild and released abalone, as well as the survival rates of released abalone, in 1997 and 1998 were estimated. The age of the specimens was also determined from the specific temperature of each month and the inverse relationship between oxygen isotope values and temperatures. Results show that examining stable isotopic profiles proves to be a practical and feasible method for stock discrimination as well as density and abundance estimation. These data would be helpful in assessing the success of abalone culture and release programs, which aim to enhance this valuable marine resource. 相似文献

Use of phages for identifying the citrus canker bacterium Xanthomonas campestris pv. citri in Taiwan
Phages CP115 and CP122, which were isolated from canker lesions on grapefruit and Liucheng sweet orange, respectively, showed a high degree of specificity with respect to lysis of test bacterial strains. When used jointly, they lysed 135 (97·8%) out of 138 Xanthomonas campestris pv. citri strains isolated from the canker lesions on leaves, twigs, and fruits of various citrus species, cultivars, and hybrids grown throughout Taiwan, but they did not lyse other X. campestris pathovars and other phytopathogenic bacteria, nor other bacteria isolated from soil, clinical or environmental samples. Of 252 CP115/CP122-sensitive and 78 CP115/CP122-resistant bacterial strains with colony characteristics typical of or similar to those of X. campestris pv. citri , isolated from canker lesions of various citrus plants in diverse growing regions in Taiwan, 250 (99·2%) and 76 (97·4%) strains were pathogenic and non-pathogenic, respectively, when inoculated into Liucheng sweet orange or Mexican lime. Thus, phages CP115 and CP122, when used jointly, appear to be applicable for identifying X. campestris pv. citri in Taiwan. 相似文献

洋麻炭疽病的种子消毒处理,在現有方法中,虽以冷水溫湯浸种为最好,但杀菌效果仍不能彻底,效果也不稳定。試驗証明,預浸后种子的含水量是决定溫浸效果的关鍵性因素。一般种子在20℃的冷水中預浸12小时,含水量由10%增加到40%,以后在50℃的温水中处理15分钟或52℃处理10分钟,就可获得彻底杀菌的效果。随着預浸时間的延长,种子的含水量相应增加,同时种子間含水量的差异也逐漸减少,更易达到彻底的目的。 相似文献

This study determined the
antimicrobial resistance profiles of Escherichia coli isolates from dogs
with a presumptive diagnosis of urinary tract infection (UTI). Urine samples from 201 dogs
with UTI diagnosed through clinical examination and urinalysis were processed for
isolation of Escherichia coli. Colonies from pure cultures were
identified by biochemical reactions (n=114) and were tested for susceptibility to 18
antimicrobials. The two most frequent antimicrobials showing resistance in Urinary
E. coli isolates were oxytetracycline and ampicillin. Among the
resistant isolates, 17 resistance patterns were observed, with 12 patterns involving
multidrug resistance (MDR). Of the 69 tetracycline-resistant E. coli
isolates, tet(B) was the predominant resistance determinant and was
detected in 50.9% of the isolates, whereas the remaining 25.5% isolates carried the
tet(A) determinant. Most ampicillin and/or amoxicillin-resistant
E. coli isolates carried blaTEM-1 genes.
Class 1 integrons were prevalent (28.9%) and contained previously described gene cassettes
that are implicated primarily in resistance to aminoglycosides and trimethoprim
(dfrA1, dfrA17-aadA5). Of the 44 quinolone-resistant
E. coli isolates, 38 were resistant to nalidixic acid, and 6 were
resistant to nalidixic acid, ciprofloxacin and enrofloxacin. Chromosomal point mutations
were found in the GyrA (Ser83Leu) and ParC (Ser80Ile) genes. Furthermore, the
aminoglycoside resistance gene aacC2, the chloramphenicol resistant gene
cmlA and the florfenicol resistant gene floR were also
identified. This study revealed an alarming rate of antimicrobial resistance among

Reasons for performing study: Differences in racing times have been noted on synthetic track surfaces that appear to depend on the temperature of the track. No published study to date has considered this effect in a systematic manner. Objectives: To investigate the relationship between temperature of track and speed of horses racing on a synthetic surface. Potential changes in the wax component of the synthetic track were investigated as one possible cause of changes in the track speed at the temperatures observed. Methods: At Del Mar racetrack (California, USA), the air, surface and subsurface temperatures at 4 depths in the synthetic race surface were measured periodically throughout the day over a 42 day period. The 6 furlong (1.2 km) race (afternoon) and fast training ‘work’ (morning) times were also compiled. Samples of the track were obtained and the wax separated using a solvent separation technique. Differential scanning calorimetry was used to determine the range of temperatures at which the wax from the track underwent softening and other material changes. Transformation temperatures were compared to temperatures acquired from the track to evaluate the likelihood of changes in the wax properties during racing. Results: Average air, surface and subsurface temperatures changed significantly throughout the day. Temperatures were higher during the afternoon race sessions and race times were significantly slower compared to morning work times. Temperatures at which some of the components of the wax began to soften were found to be within the range of temperature measured during track operation. Conclusions: A correlation was found between temperature of the synthetic track and speed of horse. Wax separated from the track showed that the temperatures experienced in the surface during normal operation exceed the temperatures at which the wax begins to experience thermal transformation. It is therefore hypothesised that the wax may be a cause of the observed changes in the track performance. Potential relevance: Future work should include a study of components of the synthetic track responsible for the change and epidemiological association of risk of injury. 相似文献

This study investigates the seasonal variation of germination ability of buried seeds of Monochoria vaginalis (Burm.f.) Presl var. plantaginea Solms. The field-collected seeds were buried in a flooded or an upland field and then exhumed monthly. The exhumed seeds were germinated under four temperature regimes. The seeds exhumed from the flooded soil were dormant at the beginning of burial and proceeded into a conditional dormancy/non-dormancy/conditional dormancy cycle throughout the remaining period of the experiment. The seeds exhumed monthly from the non-flooded soil exhibited an annual dormant cycle, which is dormancy/conditional dormancy/non-dormancy/conditional dormancy/dormancy. At day and night temperatures of 25/20 °C, the exhumed seeds from both the flooded and the upland soil resembled each other in terms of seasonal variation of the germination percentage. In September and October, more seeds exhumed from upland soil failed to germinate under higher temperature than from flooded soil. Strictly avoiding exposure to light during seed exhuming and seed testing prevented the seeds from germinating. A short exposure of the exhumed seeds to light during preparation promoted dark germination when the seeds were at the non-dormant stage. The potential implications of our results for weed management strategies in rice production are discussed. 相似文献

萤光显微法常用于实验室研究胶黏剂在人造板里的分布,用这个方法作为人造板生产线品质控制的指标还不广泛。本实验利用萤光显微技术配合电脑图像分析,先测定实验室制作的一组高密度纤维样板里的异氰酸酯含胶量,再用同法测定工厂产的高密度纤维板里的异氰酸酯含胶量。实验室样板施胶量为2%、3%、4%、6%及8%的高密度纤维板各2张,3张高密度纤维样板是从工厂生产线上在4h之内不同时间取得。结果显示,实验室各种施胶量的样板,用萤光显微法测得的含胶量分别为2.06%,2.901%,4.00%,6.17%及9.07%,测得含胶量与施胶量的相关系数为r=0.996 0。用同法测得3张工厂样板的含胶量分别为3.53%、3.54%及3.56%,与生产时设定的3.50%施胶极为接近。本实验结果显示,萤光显微分析法可迅速并准确地测出高密度纤维板里的异氰酸酯含胶量,用该法检测工厂样板的结果表示,该工厂在4h取样期间内施胶情形良好而且稳定,胶黏剂效率高,产品品质优良。 相似文献
