Characterization of ovine strains of Mycobacterium paratuberculosis by restriction endonuclease analysis and DNA hybridization.

Restriction endonuclease analysis and DNA hybridization revealed five ovine strains of Mycobacterium paratuberculosis from South Africa had identical DNA patterns to an ovine strain from Canada. Genetically this strain type has features in common with the two major groups of M. paratuberculosis.     

Characterisation of the 33kDa piroplasm surface antigen of Theileria orientalis/sergenti/buffeli isolates from West Java, Indonesia.

The immunodominant 33/35kDa antigen of a Theileria isolate from West Java, Indonesia, was characterised and immuno-affinity purified by use of a monoclonal antibody, KUL-a4, and was shown to be representative of the T. orientalis/sergenti/buffeli group. The aminoterminal sequence of the purified 35kDa peptide (20 residues) was determined by automated Edman degradation and found to correspond to the predicted amino acid sequence of a prospective p33 gene previously sequenced from the same isolate. The cleavage site of a putative signal peptide was identified and conforms the (-3, -1) rule for signal peptidases. The existence of dimeric and trimeric forms of the p33/35 antigen is hypothesised from Western blot profiles. KUL-a4 appeared specific for the T. orientalis/sergenti/buffeli group. It did not recognise in indirect fluorescence antibody test (IFAT), intraerythrocytic bodies of Anaplasma marginale or piroplasms and schizonts of T. mutans, T. parva and T. annulata, whereas cattle antisera raised to these species showed cross-reactivity in IFAT. It however, appeared weakly cross-reactive in Western blot and ELISA, with the 34kDa piroplasm antigen of one T. annulata (Gharb) isolate. The present study indicates that the isolated antigen belongs to the p33/34 antigen family described within the T. sergenti/orientalis/buffeli group, and documents the group-specificity of one of its epitopes.     

A Tremor Syndrome With a Central Axonopathy in Scottish Terriers

A central axonopathy in 2 male and 1 female Scottish Terrier puppies from 3 different but related litters is reported. Clinical signs consisting of severe whole-body tremors and ataxia were first detected at the age of 10 to 12 weeks. They worsened with activity and excitement and diminished during rest or sleep. Two dogs also had paraparesis. In 1 dog the neurological deficits progressed over several months. Neuropathological examination revealed widespread axonal changes, vacuolation, and gliosis in the white matter of the central nervous system.

P‐20 Clinical,cytopathological and histopathological evaluation of sporotrichosis in experimentally infected cats

The purpose of this study was to evaluate the clinicopathological aspects of experimental sporotrichosis in cats and compare the sensitivity of cytopathology, histopathology and culture as diagnostic tools in different phases of the infection. Twenty adult, mixed‐breed cats (10 males and 10 females) were inoculated subcutaneously with 10⁶ fungal microorganisms. Clinical examination was performed weekly. Cytopathologic, histopathologic and culture examinations were performed at 15, 30 and 60 days postinoculation. Culture of multiple organs was performed after euthanasia at 30 (10 cats) and 60 (10 cats) days postinoculation. Friedman parametric and nonparametric statistical analysis were applied to the results. The nodular, tumoral and necrotic lesionsprogressed significantly until day 30 postinoculation, and partial spontaneous regression occurred at day 60, particularly in males. An intense inflammatory pyogranulatomous and lymphocytic infiltrate with rare giant cells and sparse fibrosis associated with numerous, pleomorphic, intra‐ and extracellular fungal cells were observed on day 30. These findings gradually decreased by day 60. Despite the inflammatory granuloma associated with feline sporotrichosis, a tendency for dissemination was observed, with fungal isolation in the lymph nodes, spleen and liver at the 30 and 60 days postinoculation. No significant differences were observed between cytopathology, histopathology and fungal culture during the different phases of the disease. Therefore, cytological examination was considered a simple, rapid and inexpensive diagnostic method at all stages of this disease. Funding: Self‐funded.     

The relationship between feeding and fertility in dairy cattle]

The possible relationship between nutrition and fertility in dairy cows are reviewed. Study of the literature shows that opinions on the effect of the various nutrients differ markedly. The conclusion which can be made is that in countries where standards of dairy farming are high, nutrition only plays a minor role in the problem of herd subfertility. In the course of time deficiencies in a large number of minerals, trace elements and vitamins have been stressed as a cause of inadequate fertility. However cows are at present generally fed with a lot of well balanced concentrates and therefore deficiencies are very unlikely to occur. During early lactation every high producing dairy cow has a negative energy balance. In a number of cows this results in anoestrus and in a long interval from parturition to first insemination. Overfeeding during late lactation and the dry period does not adversely affect subsequent reproduction but may cause puerperal disorders. The relation between protein nutrition and fertility is complex. Protein effects on fertility are minor. Problems can arise if there is a combination of a high percentage of crude protein (greater than 18%), a high rumen degradability of the protein and a low energy content of the diet. The value of metabolic profile tests is minimal because they do not differentiate between normal herds and herds with fertility problems.     

Evaluation of an electronic device used as a quick screening method to detect changes in the red cell content of the blood in horses participating in endurance trail rides

Dehydration, to varying degrees, will inevitably occur in horses participating in endurance trail rides. This water loss is directly related to the amount of body water lost through evaporative cooling, that in turn being related to the amount and rate of work performed, and to the environmental temperature and humidity.An electronic apparatus that can substitute for the conventional and time-consuming venous haematocrit was developed to measure the animals' hydration status. The mechanism of this electrometer is based upon the relationship between the red cell content and the electrical conductivity of the blood. The electrometer reading gave a significant correlation (p<0.001) with the venous haematocrit, thus indicating that the electrometer is a reliable apparatus for determining the animals' hydration status within seconds of venepuncture.
