Captive breeding is used to supplement populations of many species that are declining in the wild. The suitability of and long-term species survival from such programs remain largely untested, however. We measured lifetime reproductive success of the first two generations of steelhead trout that were reared in captivity and bred in the wild after they were released. By reconstructing a three-generation pedigree with microsatellite markers, we show that genetic effects of domestication reduce subsequent reproductive capabilities by approximately 40% per captive-reared generation when fish are moved to natural environments. These results suggest that even a few generations of domestication may have negative effects on natural reproduction in the wild and that the repeated use of captive-reared parents to supplement wild populations should be carefully reconsidered.  相似文献

Lead poisoning is commonly diagnosed in cattle. In this study, 3 groups of cattle from different herds accidentally exposed to discarded lead batteries on pasture were intensively studied to determine the extent and severity of exposure. The losses from acute death due to lead toxicity were substantial in all the 3 study groups at 12%, 17%, and 4%. Blood samples were taken from all cattle around the time of the first diagnosis and then later in 2 of the 3 herds to monitor the change in lead concentrations over time. Asymptomatic lead toxicosis was observed in these herds. In these 3 groups, between 4% and 12% of asymptomatic cattle had blood lead concentrations consistent with acute lead poisoning (> 0.35 ppm), and between 7% and 40% of these asymptomatic animals were in the high-normal range (0.1-0.35 ppm). Because of the consistently high number of asymptomatic cattle with elevated lead levels, all cattle potentially exposed to a lead source should be tested before sale or slaughter to minimize the entry of lead into the food chain. The blood lead concentrations, which were monitored for months after the initial diagnosis, decreased slowly after the cattle were removed from the lead source. The prolonged retention of lead may be due to continued release and absorption of lead from metal particles in the reticulum or rumen. The mean reduction in the lead level was 0.046 ppm (95% CI, 0.017-0.075 ppm) every 30 days for these 2 herds. Using a single-component exponential model, the half-life of lead in the animals retested from Herds 1 and 2 was highly variable. The median half-life was 63 days (interquartile range, 34-107 days). One out of 8 pregnant heifers with high blood levels had a stillborn calf. There were no abortions or calf mortalities in this group. Blood samples were'collected from the calves around the time of birth. The concentrations of lead in the blood of the calves exposed in utero were low (0.010-0.095 ppm).  相似文献

Background – Staphylococcus pseudintermedius is an important canine pathogen, and the emergence and widespread dissemination of meticillin‐resistant strains (MRSP) is of significant concern. Multidrug‐resistant infections may require alternative approaches, such as the use of topical therapy. There is minimal information about the in vitro susceptibility of meticillin‐susceptible S. pseudintermedius (MSSP) and MRSP to biocides and topical antimicrobials. Hypothesis/Objectives – The hypothesis was that clinical isolates of MSSP and MRSP would not have universal susceptibility to topical biocides and antimicrobials. The goal of this study was to assess the susceptibility of a collection of S. pseudintermedius isolates to selected antimicrobials and biocides. Animals – The study was performed on clinical isolates of MSSP and MRSP from dogs with skin and soft tissue infections collected throughout North America between 2006 and 2008. Methods – The minimal inhibitory concentrations (MICs) of chlorhexidine digluconate, benzalkonium chloride, triclosan, accelerated hydrogen peroxide, geranium oil, tea tree oil and grapefruit seed extract were tested for 25 MRSP and 25 MSSP isolates from dogs using the agar dilution method. The MICs of fusidic acid, bacitracin and mupirocin were determined using Etests. Results – Triclosan demonstrated excellent activity against all bacterial isolates, with no growth at the lowest concentration evaluated (MIC ≤ 0.5 μg/mL). Conversely, grapefruit seed extract did not inhibit growth at the highest concentration tested (MIC > 3.84 μg/mL). All isolates were susceptible to mupirocin, fusidic acid and bacitracin. There were no significant differences noted in the range, MIC50 or MIC90 between MSSP and MRSP isolates. Conclusions and clinical importance – While isolates were susceptible to most of the tested compounds, universal susceptibility to all compounds with potential antimicrobial activity cannot be assumed, and specific testing is required.  相似文献

Soil carbon dioxide (CO2) flux is an integrative measure of ecosystem functioning representing both biotic and physical controls over carbon (C) balance. In the McMurdo Dry Valleys of Antarctica, soil CO2 fluxes (approximately −0.1-0.15 μmol m−2 s−1) are generally low, and negative fluxes (uptake of CO2) are sometimes observed. A combination of biological respiration and physical mechanisms, driven by temperature and mediated by soil moisture and mineralogy, determine CO2 flux and, therefore, soil organic C balance. The physical factors important to CO2 flux are being altered with climate variability in many ecosystems including arid forms such as the Antarctic terrestrial ecosystems, making it critical to understand how climate factors interact with biotic drivers to control soil CO2 fluxes and C balances. We measured soil CO2 flux in experimental field manipulations, microcosm incubations and across natural environmental gradients of soil moisture to estimate biotic soil respiration and abiotic sources of CO2 flux in soils over a range of physical and biotic conditions. We determined that temperature fluctuations were the most important factor influencing diel variation in CO2 flux. Variation within these diel CO2 cycles was explained by differences in soil moisture. Increased temperature (as opposed to temperature fluctuations) had little or no effect on CO2 flux if moisture was not also increased. We conclude that CO2 flux in dry valley soils is driven primarily by physical factors such as soil temperature and moisture, indicating that future climate change may alter the dry valley soil C cycle. Negative CO2 fluxes in arid soils have recently been identified as potential net C sinks. We demonstrate the potential for arid polar soils to take up CO2, driven largely by abiotic factors associated with climate change. The low levels of CO2 absorption into soils we observed may not constitute a significant sink of atmospheric CO2, but will influence the interpretation of CO2 flux for the dry valley soil C cycle and possibly other arid environments where biotic controls over C cycling are secondary to physical drivers.  相似文献

Larvae of Siganus lineatus were hatched and reared through metamorphosis in a 7 000-1 tank covered with a translucent roof. The larvae were fed phytoplankton (mixed cultures), rotifers (Brachionus plicatilis), copepods (Oithona sp.), and Artemia nauplii. Thirty-five days were required for all the larvae to metamorphose; survival was 800 out of 5 000 (16%). The rearing period and rearing criteria are identical to those required for S. canaliculatus. Three important developmental stages were identified: the dark-head carnivorous stage (the gut is a simple coil), the brown-head omnivorous stage (the gut is elongated and convoluted), and the juvenile herbivorous stage (the gut is highly elongated and convoluted into a complex coil). Over-feeding on Artemia nauplii may result in deaths among brown-head and juvenile fishes; adjustment of the feeding regime when larvae begin to develop into the brown-head stage is considered important.
