We monitored breeding success of hornbills in Thailand for four sympatric species at Khao Yai National Park during 1981-2002 and six species at Budo-Sungai Padi National Park during 1994-2002. Within a 60 km2 study area at Khao Yai, the well protected area, use of available cavities ranged from 94% in 1984 to 50% in 1993. Competition for nesting cavities was 40% of cavities available indicating the shortage of suitable cavity. We monitored the cavity condition and modified 48 cavities and, between 1996 and 2000, these contributed annually 23-45% of nestings that were successfully fledged (n = 352). Within the 90 km2 at Budo Mountain, with heavy human disturbance, use of available cavities for nesting decreased from 81% in 1997 to 30% by 2001 and of successful fledging from 96% in 1997 to 55% in 1999, but both increased once our involvement with local communities had eradicated poaching and reduced disturbance. Competition for nesting cavities here was 26% of cavities available indicating the effect of poaching. Both strategies were successful in the short term but long-term management of cavities in large forest trees is required at Khao Yai and of forest disturbance at Budo.  相似文献

Bioassay-guided fractionation of the cytotoxic ethyl acetate extract from the stems of Dasymaschalon blumei (Annonaceae) led to the isolation of four aristololactam alkaloids, including the hitherto unknown 3,5-dihydroxy-2,4-dimethoxyaristolactam (1), as well as the three known compounds, aristolactam BI, goniopedaline, and griffithinam. Additionally, the cytotoxic extract from the combined leaves and twigs of the same plant yielded three known oxoaporphine alkaloids, oxodiscoguattine, dicentrinone, and duguevalline. The structures of aristolactams and oxoaporphine alkaloids were elucidated on the basis of spectroscopic methods. All isolates were evaluated for cytotoxicity against a panel of mammalian cancer cell lines and a noncancerous human embryonic kidney cell Hek 293.  相似文献

This study was conducted to compare the developmental competence of somatic nuclear transfer (NT) embryos, after either ionomycin or ethanol activation, in locally bred goats. Donor cells were prepared from the ear skin fibroblasts of a female goat. Cells, at passage 3-8, starved by culturing in 0.5% FCS for 4-8 d, were used for NT. Immature oocytes were obtained from FSH-stimulated goats and matured for 22 hr before enucleation and NT. After fusion, the reconstructed embryos were activated with either ionomycin or ethanol followed by culturing in 6-dimethylaminopurine (6-DMAP) and cytochalasin B (CB), for 3 hr. In experiment I, the fused NT embryos (n=63, ionomycin and n=68, ethanol treatments, respectively) were cultured in B2 with a Vero co-culture system and their developmental competence was evaluated through to Day 9. In experiment II, the NT embryos at the 2-4 cell stage on Day 2 derived from each treatment (ionomycin n=46, and ethanol n=37), were transferred into 10 synchronous recipients. There were no significant differences between the NT embryos derived from the ionomycin and ethanol groups, in fusion (86.3% versus 82.9%), cleavage (90.5% versus 82.4%) and for morula/blastocyst development rates (9.5% versus 5.9%). Sixty percent (3/5) of the recipients from ionomycin became pregnant by midterm (2.5 mts) while only 20% (1/5) from ethanol treatment was pregnant by Day 45. The results demonstrate that activation with either ionomycin or ethanol in combination with 6-DMAP-CB treatment does not affect the development of cloned goat embryos.  相似文献

Understanding organic matter characteristics(e.g., amount and state) of sediments is necessary for evaluating both the sediment and water environments; however, methods that have been proposed to investigate these characteristics are relatively expensive. This study attempted to evaluate the organic matter characteristics of littoral sediments by employing solely the loss on ignition(LOI)method, which is the most economical and accessible method in developing countries. Different types of sediments were first oven dried at 100℃ and then continuously burned at 200, 300, 400, 500, and 600℃ for 4 h at each temperature. The mass LOI at each temperature was calculated and compared with the 100℃ oven-dried weight. Our results suggested that the mass LOI across the temperature range of 200–300℃(LOI_(200-300)):mass LOI at 600℃(LOI_(600)) ratio(LOI_(200-300)/LOI_(600)) could represent different types of sediments as well as the organic matter:organic C ratio. Compared with the sediments unaffected by wastewater, the sediments that were more influenced by the inflow of domestic wastewater had a higher LOI_(200-300)/LOI_(600). Interestingly, LOI_(200-300)/LOI_(600) could also represent changes in the organic matter characteristics as a function of organic matter decomposition in sediments. In conclusion, the organic matter characteristics of littoral sediments could be evaluated using solely the LOI method, particularly with LOI_(200-300)/LOI_(600).  相似文献

Seven FDA pesticide laboratories collaborated to develop and validate an LC-MS/MS method to determine 173 pesticides in <20 min. The average determination coefficient (r2) was >0.99 for all but two compounds tested. The limits of detection were <20 ng/mL for all compounds and <10 ng/mL for 363 of the 368 transitions reported. The method was used to determine pesticides in two AOAC sponsored proficiency samples. The LC-MS/MS determination was used for the analysis of oranges, carrots and spinach using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged, Safe) method. Each matrix was fortified at 20, 100, 400, and 1000 ng/g. No false positive responses were detected in controls of the three matrices. 165 pesticides had recoveries between 70 and 130%, and 161 had minimum detection levels less than 10 ng/g. Recoveries of 169 compounds for the 1000 ng/g spikes were within 50-150%. A matrix effect study indicated all three matrices caused a small net suppressing effect, the most pronounced attributable to the citrus matrix. The procedure proved to be accurate, precise, linear, sensitive and rugged, and adds 100 pesticides to the scope of the FDA pesticide program.  相似文献

西农979是我国黄淮麦区优质高产、早熟耐寒兼抗赤霉病的小麦新品种。十倍体长穗偃麦草(Thinopyrum ponticum)7E染色体携带有抗赤霉病和抗叶锈病等多种抗性基因。为明确西农979品种的遗传基础,以西农979及其主要供体亲本小偃6号、早优504、陕229、陕213和西农881为材料进行系谱分析,结果表明,西农979及其主要供体亲本陕229、陕213、西农881均为小偃6号的衍生系;小偃6号是十倍体长穗偃麦草的衍生系。在此基础上,利用十倍体长穗偃麦草7E染色体上106个特异分子标记进行分析,发现有6个标记在西农979和小偃6号中扩增出了十倍体长穗偃麦草的特异片段,西农979和小偃6号均携带有十倍体长穗偃麦草7E染色体短臂上分子标记Xwmc653-Xwmc809之间的75.10~77.46cM区段,标记Xcfd31-Xgwm350之间的86.16~87.32cM区段,以及7E染色体长臂标记Xmag1932-Xdauk144之间的147.71~149.51cM区段。结果表明,西农979携带的十倍体长穗偃麦草7E染色体上的遗传物质源自小偃6号,这为进一步研究和利用西农979提供了理论参考。  相似文献

The controlled release of diclofenac sodium (DFNa) from a chitosan-oxidized konjac glucomannan (CTS-OKG) polymer film was studied. Konjac glucomannan (KGM) was initially oxidized by sodium periodate and then cross-linked to CTS via imine bonds (-C=N-) to form the new CTS-OKG copolymer. The DFNa loaded CTS-OKG polymers were characterized by Fourier transformed infrared spectroscopy (FT-IR) and X-ray diffractometry (XRD). Finally, the release profiles of DFNa from the CTS-OKG polymer matrices were evaluated in a simulated gastrointestinal fluid system comprised of two hours in simulated gastric fluid (SGF; pH 1.2) followed by 24 h in simulated intestinal fluid (SIF; pH 7.4). A 1:2:1 (w/w/w) ratio of CTS:OKG:DFNa prepared at room temperature for 3 hours gave the highest % encapsulation efficiency (EE) of 95.6 ± 0.6 and resulted in a minimal release of DFNa (<1% over 2 h) in SGF (pH 1.2) and a significantly improved sustained release in SIF (pH 7.4) with ~6% and 19% release over 8 and 24 h, respectively), some 15- and five-fold lower than that of the two commercial DFNa preparations, Diclosian and Voltaren. This formulation may be used for further study as a long term intestine controlled release drug model (at least 3 days).  相似文献

Host-plant resistance is an effective and economical means of controlling downy mildew (DM) diseases in maize (Zea mays L.). Crosses involving 10 inbred maize lines in all possible combinations (diallel cross) showed that susceptibility to sorghum DM (Peronosclerospora sorghi (Weston and Uppal) C. G. Shaw) is generally inherited as a dominant characteristic. However, progenies of crosses between one of the lines (CM105) resistant to DM, and susceptible lines, showed intermediate levels of resistance, indicating that several genes are involved and that their effects are additive. Progenies of crosses between resistant lines were more resistant than their parents, probably because of interactions between different resistance genes (epistasis). Both additive and non-additive gene action was detected by partitioning gene effects by generation mean in each 16 DM-resistant × DM-susceptible combinations, which indicates that the reactions of maize to DM are governed by several genes (i.e. polygenically) and that the inheritance of resistance is complex. An inbred line from Mexico, Tuxpeno Gpo. 48-Var. 168-1, is a particularly promising general source of resistance to DM.  相似文献

The development of germ cells has not been entirely documented in the cat especially the transition phase of the gonocyte to the spermatogonial stem cell (G/SSC). The aims of study were to examine testicular development and to identify the G/SSC transition in order to isolate and culture SSCs in vitro. Testes were divided into 3 groups according to donor age (I, < 4 months; II, 4–6 months; and III, > 6 months). In Exp. 1, we studied testicular development by histology, transmission electron microscopy and immunohistochemistry. In Exp. 2, we determined the expression of GFRα-1, DDX-4 and c-kit and performed flow cytometry. The SSCs isolated from groups II and III were characterized by RT-PCR and TEM (Exp. 3). Chronological changes in the G/SSC transition were demonstrated. The size, morphology and ultrastructure of SSCs were distinguishable from those of gonocytes. The results demonstrated that group II contained the highest numbers of SSCs per seminiferous cord/tubule (17.66 ± 2.20%) and GFRα-1+ cells (14.89 ± 5.66%) compared with the other groups. The findings coincided with an increased efficiency of SSC derivation in group II compared with group III (74.33 ± 2.64% vs. 23.33 ± 2.23%). The colonies expressed mRNA for GFRA1, ZBTB16, RET and POU5F1. Our study found that the G/SSC transition occurs at 4–6 months of age. This period is useful for isolation and improves the establishment efficiency of cat SSCs in vitro.  相似文献
