This study investigated the effects of dietary poly‐β‐hydroxybutyrate (PHB) and mannan oligosaccharide (MOS) on the growth performance, digestive enzyme activities, and disease resistance of rainbow trout fingerlings. A total of 420 fingerlings, distributed equally into four different groups, each with three replicates, were fed a basal diet (control), 2% PHB (PHB), 0.2% MOS (MOS), or a diet supplemented with a mixture of 2% PHB and 0.2% MOS (PHB+MOS) for 60 d. Results showed that dietary PHB and/or MOS did not cause significant improvement in the growth performance of the fingerlings. The gut pH was markedly reduced; however, the activities of the digestive enzymes (except for pepsin and amylase by dietary PHB) were not significantly improved by the dietary supplements. Results also showed that dietary PHB and/or MOS markedly increased the resistance of the fingerlings toward Yersinia ruckeri infection, while the lowest total antibody was observed in fish fed control and MOS, indicating that MOS exerts its protective effects by modulating other mechanisms. Overall, this study provided a first screening effort to determine the effects of PHB and/or MOS as dietary supplements in rainbow trout fingerlings to develop an optimal prebiotic mix for improving the growth and health status of rainbow trout.  相似文献

Urban and industrial air pollution can cause elevated heritable mutation rates in birds and rodents. The relative importance of airborne particulate matter versus gas-phase substances in causing these genetic effects under ambient conditions has been unclear. Here we show that high-efficiency particulate-air (HEPA) filtration of ambient air significantly reduced heritable mutation rates at repetitive DNA loci in laboratory mice housed outdoors near a major highway and two integrated steel mills. These findings implicate exposure to airborne particulate matter as a principal factor contributing to elevated mutation rates in sentinel mice and add to accumulating evidence that air pollution may pose genetic risks to humans and wildlife.  相似文献

Introduction: Antibiotic supplements are regularly used in neuronal culture media to control contamination; however, they can interfere with the neuronal excitability and affect electrophysiological properties. Therefore, in this study, the effect of penicillin/streptomycin supplements on the spontaneous electrophysiological activity of hippocampal pyramidal neurons was examined. Methods: Electrophysiological whole-cell patch-clamp recordings from rat hippocampal pyramidal cells in primary culture were performed to investigate the effects of antibiotic supplements on the intrinsic excitability of cultured cells. Results: The present findings indicated that presence of antibiotic supplements (penicillin/streptomycin) in the culture medium altered the intrinsic electrical activity of hippocampal pyramidal neurons in primary culture. These alterations included: 1) depolarized resting membrane potential; 2) a significant enhancement in the after-hyperpolarization amplitude; 3) a significant increase in the area under the action potential and in the decay and rise time of the action potential; 4) a significant broadening of action potential and 5) a significant reduction in the firing frequency. Conclusion: These findings suggest that addition of antibiotic supplements to culture media influences the neuronal excitability and alters the electrophysiological properties of cultured neurons, possibly through changing the ionic conductance underlying neuronal excitability. Pyrus L. is a taxonomically difficult genus mainly because of interspecific and intergeneric hybridization. A majority of the wild species of Pyrus occurs in Iran, making this region very important for any study of the systematics of the genus. In this investigation, seven quantitative and seven qualitative characters of leaf epidermis, particularly epidermal cells shape, anticlinal walls patterns, type and size of stomata, and cuticular ornamentation of 22 taxa of Pyrus plus Malus orientalis are studied for assessment of their taxonomical value at different levels. Scanning electron microscopy and light microscopy were used for evaluation of epidermis features of these taxa. Some characters such as shape and anticlinal wall pattern of epidermal cells, presence of adaxial stomata, pattern of cuticular ornamentation and epicuticular wax types are useful in separation of some Pyrus taxa. Moreover, Malus orientalis is recognized easily from all Pyrus species via the density of stomata. According to ANOVA analysis, all quantitative variables show high level of between-group differentiation. Also, based on the hierarchical clustering results gained by multiple correspondence analysis and factor analysis of mixed data analyses of the current study, micromorphological characters of epidermal cells reflect the ecological conditions of pear taxa, so most of the mesophyte and xerophyte taxa can be separated from each other in accordance with epidermal cells characters, specially the anticlinal wall pattern. Totally, the application of some other biological markers, particularly suitable molecular data are needed to present well-resolved relationships among these taxa.  相似文献

BACKGROUND: Trichophyton tonsurans is one of the dermatophyte fungi which invades the skin and hair of human. Several properties of this fungus have been investigated so far. However a few studies were carried out in the field of molecular biology of this fungus. In the present study, we tried to identify the Squalene epoxidase gene which is related to synthesis of ergosterol in this fungus. METHODS: Pairs of 23 and 24 nucleotides primers were designed from highly conserved regions of the similar genes in other fungi. Mentioned primers were utilized in PCR by using isolated genomic DNA of T. tonsurans whereas the PCR fragments were then sequenced. RESULTS AND CONCLUSION: Nucleotides (n = 558) have been sequenced from this new gene which encodes a polypeptide with 186 amino acids. Sequences comparison in gene data banks (NCBI, NIH) for this part of DNA and its deduced amino acid revealed significant homology with members of the eukaryotic Squalene epoxidase.  相似文献

The possibility of manufacturing rubberwood and kenaf (Hibiscus cannabinus L.) stem medium density fibreboard (MDF) panels at different pressure and resin content were investigated. The effect of mechanisms of interacted independent variables (resin content and pressure) on MDF properties was analyzed. The board performance was evaluated by measuring internal bond (IB) strength, modulus of rupture (MOR), modulus of elasticity (MOE), water absorption (WA) and thickness swell (TS). The test results were statistically analyzed by using response surface method (RSM) to determine the significant independent variables that influenced MDF properties. A mathematical simulation or response surface models were developed to predict the MDF properties (MOR, MOE, IB, WA and TS). The obtained results showed that MDF density and all interactions between the experimental variables were significant factors that influenced the mechanical properties of MDF. At 8 bar and 14 % resin content, the MDF recorded WA of 83.12 % and TS of 20.2 %. It can be inferred that two parameters (resin content and pressure) had positive effect on physical and mechanical properties of MDF. We concluded that resin content show more significant effects on MDF manufacturing as compared to pressure parameters.  相似文献

Biofloc technology is considered as a method that degrades organic waste by microorganisms and produces microbial flocs. A 30-day experiment was performed to investigate the effects of partial replacement of daily feeding intake with biofloc on the growth performances, digestive enzymes activity and liver histology of the common carp Cyprinus carpio L. fingerlings. Two hundred and eight healthy fingerlings (58.6 ± 0.2 g) were randomly distributed in 12 tanks (30 L) at a density of 25.4 kg m?3 and fed experimental treatments (100 % daily feeding rate as a control, biofloc + 75 % daily feeding rate, biofloc + 50 % daily feeding rate, biofloc + 25 % daily feeding rate). At the end of experiment, the results indicated that the highest weight gain was observed in the fish fed BFT 75 % and control which differed significantly from those fed BFT 25 % (P < 0.05). Diet BFT 75 % improved total protease and pepsin activity compared with BFT 25 and 50 % (P > 0.05). No significant difference was observed in case of lipase, amylase and alkaline phosphatase activity between the treatments. In the liver, histological alterations were found in the treatments, and feeding the fish with BFT 75 % significantly improved hepatocellular quantification and qualification than the other groups. The results obtained in this experiment suggest that the biofloc improves growth performances, digestive enzyme activity and liver condition of the common carp fingerlings when 25 % of daily feeding rate (BFT 75 %) was replaced with one carbohydrate such as molasses in zero-water exchange system.  相似文献

Background:Hearing loss, a congenital genetic disorder in human, is difficult to diagnose. WES is a powerful approach for ethiological disgnosis of such disorders. Methods:One Iranian family with two patients were attented in the study. Sequencing of known NSHL genes was carried out to recognize the genetic causes of HL. Results:Molecular analyses identified a novel stop loss mutation, c.1048T>G (p.Term350Glu), whitin the P2RX2 gene, causing a termination-site modification.This event would lead to continued translation into the 3'' UTR of the gene, which in turn may result in a longer protein product. The mutation was segregating with the disease phenotype and predicted to be pathogenic by bioinformatic tools. Conclusion:This study is the first Iranian case report of a diagnosis of ADNSHL caused by P2RX2 mutation. The recognition of other causative mutations in P2RX2 gene more supports the probable function of this gene in causing ADNSHL. BACKGROUND: The aim of this study was to compare invasive and non-invasive strains of Shigella flexneri isolated from Tehran by a 120 kDa protein band by SDS-PAGE, electron microscopy of cell culture and Congo red dye methods. METHODS: S. flexneri strains were isolated by standard bacterial methods from fecal specimens of children attending to the 3 children's hospitals. Phenotype analysis for screening virulent of strains of S. flexneri was done on a plate of tryptic soy agar contained 0.003% Congo red dye. Whole membrane protein preparations were used to examine the protein profiles of the inner and outer membrane of these Gram-negative bacteria. The protein mixture was electrophoresed through a polyacrylamide gel. The gel was stained with Coomassie brilliant blue R250 and destained with ethanol and acetic acid. HeLa cell culture was done by two-step preparations: one for light microscopy and the other for electron microscopy. RESULTS: Some of S. flexneri (46%) were Congo red positive colonies. S. flexneri with negative Congo red phenotype could not enter the HeLa cell culture. A 120 kDa protein band was found in 46% of these bacteria which could enter into HeLa cell culture. Pseudopod structures which facilitate bacterial cell-to-cell spread were readily identified by electron microscopy. DISCUSSION: Since the existence of 120-kDa protein band was corresponded to enter of S. flexneri into the HeLa cell culture and correlated with Congo red dye positive, for identification of invasive and non-invasive S. flexneri strains, the use of a 120-kDa protein band by SDS-PAGE or a simple, rapid and very cheap Congo red dye method is recommended. Because, there are some deaths due to Shigella sp. in our country, notification on the isolation of these bacteria in both children hospitals laboratories and private clinical laboratories is important.  相似文献
