Eastern equine encephalitis (EEE) was diagnosed in a flock of emus in southeastern Louisiana. The outbreak involved juvenile and adult breeders ranging in age from 20 to 36 months, with an attack rate of 76% and a case fatality rate of 87%. The diagnosis was confirmed by isolation and characterization of the viral agent, and by detection of EEE antibody in two recovered emus. High mortality was preceded by marked depression, hemorrhagic diarrhea, and emesis of blood-stained ingesta. On postmortem examination, hemorrhagic enteritis and multiple petechia of viscera were observed. Microscopic changes included severe necrosis of hepatocytes, intestinal mucosa, and necrotizing vasculitis of the spleen and lamina propria of the intestine. No nervous system lesions were observed. This outbreak occurred concurrently with EEE in horses and was attributed to unseasonably heavy rainfall with an abundance of arthropod vectors and proximity to free-living reservoir host species.  相似文献

In the fall of 1999, West Nile virus (WNV) was isolated for the first time in the Western Hemisphere during an outbreak of neurologic disease in humans, horses, and wild and zoo birds in the northeastern United States. Chickens are a potential reservoir for WNV, and little is known about the pathogenicity of WNV in domestic chickens. Seven-week-old chickens derived from a specific-pathogen-free flock were inoculated subcutaneously with 1.8 x 10(3) 50% tissue culture infectious dose of a crow isolate of WNV in order to observe clinical signs and evaluate the viremic phase, gross and microscopic lesions, contact transmission, and immunologic response. There were no observable clinical signs in the WNV-inoculated chickens during the 21-day observation period. However, histopathologic examination of tissues revealed myocardial necrosis, nephritis, and pneumonitis at 5 and 10 days postinoculation (DPI); moderate to severe nonsuppurative encephalitis also was observed in brain tissue from one of four inoculated birds examined at 21 DPI. WNV was recovered from blood plasma for up to 8 DPI. Virus titers as high as 10(5)/ml in plasma were observed at 4 DPI. Fecal shedding of virus was detected in cloacal swabs on 4 and 5 DPI only. The WNV also was isolated from myocardium, spleen, kidney, lung, and intestine collected from chickens euthanatized at 3, 5, and 10 DPI. No virus was isolated from inoculated chickens after 10 DPI. Antibodies specific to WNV were detected in inoculated chickens as early as 5 DPI by the plaque reduction neutralization test and 7 DPI by the indirect fluorescent antibody test. Chickens placed in contact with inoculated chickens at 1 DPI lacked WNV-specific antibodies, and no WNV was isolated from their blood plasma or cloacal swabs throughout the 21 days of the experiment.  相似文献

Individual effect of different field scale management interventions for water saving in rice viz. changing date of transplanting, cultivar and irrigation schedule on yield, water saving and water productivity is well documented in the literature. However, little is known about their integrated effect. To study that, field experimentation and modeling approach was used. Field experiments were conducted for 2 years (2006 and 2007) at Punjab Agricultural University Farm, Ludhiana on a deep alluvial loamy sand Typic Ustipsamment soils developed under hyper-thermic regime. Treatments included three dates of transplanting (25 May, 10 June and 25 June), two cultivars (PR 118 inbred and RH 257 hybrid) and two irrigation schedules (2-days drainage period and at soil water suction of 16 kPa). The model used was CropSyst, which has already been calibrated for growth (periodic biomass and LAI) of rice and soil water content in two independent experiments. The main findings of the field and simulation studies conducted are compared to any individual, integrated management of transplanting date, cultivar and irrigation, sustained yield (6.3-7.5 t ha−1) and saved substantial amount of water in rice. For example, with two management interventions, i.e. shifting of transplanting date to lower evaporative demand (from 5 May to 25 June) concomitant with growing of short duration hybrid variety (90 days from transplanting to harvest), the total real water saving (wet saving) through reduction in evapotranspiration (ET) was 140 mm, which was almost double than managing the single, i.e. 66 mm by shifting transplanting or 71 mm by growing short duration hybrid variety. Shifting the transplanting date saved water through reduction in soil water evaporation component while growing of short duration variety through reduction in both evaporation and transpiration components of water balance. Managing irrigation water schedule based on soil water suction of 16 kPa at 15-20 cm soil depth, compared to 2-day drainage, did not save water in real (wet saving), however, it resulted into apparent water saving (dry saving). The real crop water productivity (marketable yield/ET) was more by 17% in 25th June transplanted rice than 25th May, 23% in short duration variety than long and 2% in irrigation treatment of 16 kPa soil water suction than 2-days drainage. The corresponding values for the apparent crop water productivity (marketable yield/irrigation water applied) were 16, 20 and 50%, respectively. Pooled experimental data of 2 years showed that with managing irrigation scheduling based on soil water suction of 16 kPa at 15-20 cm soil depth, though 700 mm irrigation water was saved but the associated yield was reduced by 277 kg ha−1.  相似文献

Immunogenicity of an oil-emulsified Escherichia coli (serotype O1) pili vaccine was evaluated in chickens. Chickens were vaccinated with 116 micrograms or 29 micrograms of pili protein and challenged with the homologous E. coli via the posterior thoracic air sac. Unvaccinated chickens were challenged to serve as positive controls or left unchallenged to serve as negative controls. Vaccinated chickens were protected against challenge because they suffered low or no mortality; had mild gross lesions in air sacs, pericardial sacs, and livers, and the scored values were significantly (P less than or equal to 0.05) lower than those in the positive controls; and eliminated E. coli from tissues more efficiently than the positive controls.  相似文献

The rhizomes of Acorus calamus are empirically used in the treatment of a wide variety of human diseases. Administration of the 50% ethanolic extract (100 and 200 mg/kg) as well as saponins (10 mg/kg) isolated from the extract demonstrated significant hypolipidemic activity. Identification of a small core germplasm set representing the available genetic diversity is essential for its proper evaluation and subsequent utilization in rice improvement programmes. For constituting a small diverse mini‐core panel of Indian rice germplasm, a representative set of 6912 accessions drawn based on their geographic origin from the whole rice germplasm collection available in the National Gene Bank was genotyped using 36 microsatellite markers. Automated fragment analysis of amplicons yielded a total of 435 alleles, with an average 12.4 and range of 3–29 alleles per locus. Polymorphism information content (PIC) ranged from 0.08 (RGNMS190) to 0.86 (RM552) with an average of 0.528. Based on genotyping data, a mini‐core consisting of 98 genotypes was identified. Ninety‐four per cent of the alleles present in the core set were present in the mini‐core. The identified small but diverse panel will be useful for further intensive trait‐specific evaluation and utilization in allele mining.  相似文献

Avian pneumovirus (APV) or turkey rhinotracheitis virus (TRTV) is an important respiratory pathogen of domesticated poultry in many countries in Europe, Africa, and Asia. Until recently, the United States was considered free of APV. In late 1996, an atypical upper respiratory tract infection appeared in turkey flocks in Colorado and shortly thereafter in turkey flocks in Minnesota. An avian pneumovirus (APV-US) that was serologically distinct from the previously described TRTV was isolated as the primary cause of the new syndrome. The nucleotide sequence of a fragment of the APV-US fusion gene was determined and used to develop a polymerase chain reaction-based assay that specifically detects APV-US viral nucleic acid sequences in RNA extracts of tracheal swabs and turbinate homogenates. The assay is highly sensitive in that it can detect <0.01 TCID50 of APV. The availability of this assay enables the rapid and accurate determination of APV-US in infected poultry flocks.  相似文献

Pilus proteins from Escherichia coli serotypes O1, O2, and O78 pathogenic to poultry were compared with regard to their antigenic relatedness and partial amino acid sequences. Agglutination, immunodiffusion, and immunoblot assays with polyclonal antibodies to these pili showed that these pili not only share some common antigens but also contain antigens unique to each pilus. The partial amino-terminal amino acid sequences support our earlier findings that the pili are different but contain some structural homologies.  相似文献
