Twenty-seven crossbred calves (1/2 Simmental, 1/4 Hereford, 1/4 Brahman) averaging 257 +/- 11 d of age were either hot-iron-branded (H), freeze-branded (F), or sham-branded (S). Calves were blocked for temperament, weight, and sex and were randomly assigned to day and order in which treatments were applied. To reduce stress from handling at treatment time, each calf was herded through the squeeze chute daily for 5 d before the experiment. Jugular cannulas were inserted in each calf 1 d before application of treatment. Blood samples and heart rate measures were obtained at -5, -3, 0, .5, 1, 3, 5, 10, 15, and 20 min after application of the treatments. Mean concentrations of plasma epinephrine (EPI) were higher for H calves at time .5 min than for either S or F calves (P = .10). To account for individual differences, prebranding heart rates and hormone concentrations were subtracted from subsequent samples and were also used to calculate a proportion for each subsequent sample. Analyses of subtracted values found that EPI concentrations were greater for H calves than for either S or F calves (P = .007) at .5 min postbranding. No other differences were found for the subtracted analyses. Analyses of proportion data also revealed that H calves had greater EPI than did either S or F calves (P = .027) at .5 min postbranding. Only three animals vocalized during branding, one H calf and two F calves. Adult wethers (n = 750) were lot-fed for 13 days, 8 days or 3 days before a simulated voyage lasting 18 days to examine whether the period of lot-feeding affected the proportion of sheep that ate pelleted feed and their body weight change during simulated shipping. There was no significant difference in the proportion of non-feeders between treatment groups on days 7 and 14 of the voyage. Body weights were not significantly different between the treatment groups on days 14 and 18 of the voyage. Overall body weight loss, from the farm to the end of simulated shipping, was 4.08 kg (+/- 0.28, s.e.m.), 4.58 kg (+/- 0.28) and 4.51 kg (+/- 0.28) in sheep lot-fed for 13 days, 8 days and 3 days, respectively, and was not significantly different between treatments. It was concluded that lot-feeding for 13 days conferred no advantage in body weight or numbers of non-feeders compared with shorter periods in this study.  相似文献

Echinococcus granulosus worm excretory/secretory antigens (WES) were used in ELISA for diagnosis of E. granulosus infection in dogs and compared with protoscolex somatic antigens (PSM). Sera from 224 dogs were tested. There was no correlation between ELISA absorbance values and E. granulosus worm burdens using either antigen. There was a significant linear relationship between absorbance values of sera tested in the ELISA using WES (W-ELISA) and the ELISA using PSM (P-ELISA). However, there was a small but significant difference between the absorbance values of the sera tested against the two antigens. Western blot analysis of WES using sera from E. granulosus-infected and uninfected dogs revealed antigenic components of relative molecular mass (Mr) larger than 94,000, Mr 94,000-68,000 and Mr 43,000-39,000 in worms, and these were specific for E. granulosus and not identified in PSM; these antigenic differences may be responsible for differences in reactivity in ELISA. The sensitivities of W-ELISA and P-ELISA were 80.8% and 75.6%, respectively. The specificities of W-ELISA and P-ELISA were 93.7% and 97.9%, respectively. The reduced specificity in W-ELISA was mainly attributable to increased background reactivity of sera from Taenia hydatigena-infected dogs. Despite the reduction in specificity, both ELISAs are valuable epidemiological tools to determine the prevalence of antibody to E. granulosus in dog populations and to monitor the success of hydatid control campaigns.  相似文献

Following an antigenic dose of 10 microgram or a tolerogenic dose of 200 microgram of Escherichia coli O138 lipopolysaccharide (LPS), BALB/c mice were examined on day 14 for percentages of theta-bearing cells. A considerable increase in T cells was noticed in lymphocytes from tolerant draining lymph nodes, and furthermore these cells did not possess receptors for LPS when tested for rosette inhibition. However, when the supernatant from 1 X 10(7) macrophages, pretreated with 150 microgram LPS, was given to tolerant mice on day 7, by day 14 tolerance was found to be broken, anti-LPS IgG was present in circulation and the draining lymph node contained T cells specifically committed to LPS. The change from suppressor to helper T cell activity is discussed in relation to enhancement of the immune response.  相似文献

The present study was designed to examine the effects of cell-cycle synchronization protocols, such as confluent, roscovitine treatment and serum starvation, in bovine foetal fibroblasts on synchronization accuracy at G0/G1, viability, apoptosis, necrosis and ploidy for use as a nuclei donor. The cells in 5-10 passages were randomly allocated into three treated groups. Cells were cultured either in Dulbecco's modified Eagle's medium (DMEM) + 10% foetal bovine serum (FBS) until 90% confluent (group 1, confluent), in DMEM + 10% FBS + 30 microM roscovitine for 12 h (group 2, roscovitine), or in DMEM + 0.5% FBS for 5 days (group 3, serum starvation). Most of the cells (>80%) in all groups were arrested at the G0/G1 stage. Although the rates did not differ, cells in group 1 showed an increased cell population arrested at the G0/G1 phase. Significantly (p < 0.05) higher rates of apoptosis occurred in group 3 than in group 1 and 2 (10% vs 6% and 6%, respectively). No differences in chromosomal abnormality were observed among groups. However, by increasing the number of cell culture passages up to 15, significantly (p < 0.05) higher chromosomal abnormality was observed than in 5 and 10 passages (39% vs 28% and 23%, respectively) in group 1. The results clearly indicated that bovine foetal fibroblasts could be effectively synchronized at G0/G1 stages by all the three different treatments, confluent, roscovitine and serum starvation. However, cells in confluent showed reduced apoptosis and necrosis when they underwent 5-10 passages, exhibiting increased percentage of cells with stable chromosome diversity. Hence, cells in confluent merit further studies before they could be used as nuclear donors.  相似文献

The implementation of nutrient management plans for confined animal feeding operations requires recording N and P loads from land-applied manure, including nutrients applied in irrigation water from manure treatment lagoons. By regulation, lagoon irrigation water nutrient records in Mississippi must be based on at least one lagoon water nutrient analysis annually. Research in Mississippi has shown that N and P levels in lagoon water, and the N:P ratio, vary significantly through the year. Nutrient estimates based on one annual analysis do not account for this variability and may overestimate or underestimate N and P loads. The present study reports an improved method to more precisely estimate N and P loads in irrigation water from swine manure lagoons. The method is based on predictable annual cycles of N and P levels in lagoon water and employs simple curve-fitting of lagoon-specific formulas derived by analyses of historical data. Similarity of curves from analyses of Mississippi lagoons and other lagoon studies suggests that the method can be applied using the often limited nutrient data for a lagoon to more precisely estimate seasonal shifts of N and P and to improve the precision of estimates for N and P in irrigation water. Although the present study focused on swine manure lagoons in the southern US, recognition that the annual N cycle in lagoon water is temperature driven, suggests that additional research incorporating temperature into future models could extend these models to other types of waste treatment lagoons and climates.  相似文献
