Effects of Nylon and Polyglactin 910 Suture Material on Perilimbal Corneal Wound Healing in the Dog

The effects of fine suture materials on corneal wound healing in the dog were studied. In 20 dogs, standardized perforating perilimbal clear corneal wounds were made and closed with either monofilament Polyglactin 910 or nylon. Five dogs each were euthanized 8,12,16, and 21 days postoperatively. Results of gross, biomicroscopic, and histologic examinations at 8,12,16, and 21 days showed nylon and Polyglactin 910 to cause similar inflammatory responses. Epithelialization and suppuration around the suture tracts were observed more frequently when Polyglactin 910 was used. Both materials were associated with a foreign body (granulomatous) response. There was no loss of wound integrity with either material. Strengths of the incised corneal tissues with and without the sutures intact were determined. At day 16 postsurgery, there was a statistically significant difference in tissue strengths between cases in which sutures were and were not intact. This difference was not apparent at 21 days postsurgery, which suggested that the suture dependent phase ends between the 16th and 21st postsurgical days. In addition, at day 16 there was no statistical difference between the tissue strength of wounds sutured with nylon or Polyglactin 910. Corneal suture materials studied should remain in place for at least 16 days, and absorbable material is only appropriate if it retains tensile strength for 16 days.     

Probleme der biologischen Insektizidprüfung. V. Überlegungen zur Einrichtung biologischer Mittelprüfungslaboratorien

Zusammenfassung Die toxikologische Prüfung von Insektiziden wird sich künftig auch der Insekten als Modellversuchstiere stärker bedienen. Dadurch erwachsen dem Laboratoriumssektor der biologischen Mittelprüfung an Insekten neue Aufgaben. Diese vermag sie nur bei einer Verschärfung sorgfältiger Versuchsbedingungen zu lösen. Eine der dazu erforderlichen Voraussetzungen ist die saubere bauliche Trennung zwischen Zuchträumen, Testräumen, Mittelvorratsräumen und eventuell noch den Räumen für die Zucht von Insekten im insektizidhaltigen Milieu. Hierfür wird ein Organisationsschema beschrieben und abgebildet.Von den bisherigen Folgen dieser Reihe sind erschienen: Teile I. (Bauch 1961) in der Zeitschrift Angewandte Parasitologie2: 1, II. High-velocity pulsars in the galactic halo

It is proposed that high-velocity pulsars are produced in extended galactic halos, and possibly in extragalactic space, from primordial (population III) stars. Such a population of neutron stars could provide an explanation for the gamma-ray bursters and would then accommodate the possibility that most bursters are not in the visible parts of galaxies.

Diversity of human copy number variation and multicopy genes

Copy number variants affect both disease and normal phenotypic variation, but those lying within heavily duplicated, highly identical sequence have been difficult to assay. By analyzing short-read mapping depth for 159 human genomes, we demonstrated accurate estimation of absolute copy number for duplications as small as 1.9 kilobase pairs, ranging from 0 to 48 copies. We identified 4.1 million "singly unique nucleotide" positions informative in distinguishing specific copies and used them to genotype the copy and content of specific paralogs within highly duplicated gene families. These data identify human-specific expansions in genes associated with brain development, reveal extensive population genetic diversity, and detect signatures consistent with gene conversion in the human species. Our approach makes ~1000 genes accessible to genetic studies of disease association.     

cDNA-cloning and expression of VP1-specific sequences of foot-and-mouth disease virus types A5 and O1

The RNA genome of foot- and mouth disease virus strains A5 Westerwald and O1 Lausanne has been reverse-transcribed and cloned in lambdaphages or plasmids. Identification of cDNA-clones containing VP1-specific sequences was achieved by hybridization, restriction mapping, and sequence analysis. VP1-coding cDNA-fragments were subcloned into the expression vector pEX which led to synthesis of fusion proteins with beta-galactosidase. These fusion proteins reacted with anti-VP1 antibodies on a Western blot, but were not capable of inducing neutralizing antibodies to mice. This seemed to suggest a tertiary structure of the VP1-epitopes unlike those of native VP1. Other attempts are discussed to construct VP1-fusion proteins folding similarly to the native viral protein structure.
