Microsatellites or Simple Sequence Repeats(SSRs) are informative molecular genetic markers in many crop species. SSRs are PCR-based, highly polymorphic, abundant, widely distributed throughout the genome and inherited in a co-dominant manner in most cases. Here we describe the presence of SSRs in cDNAs of cotton. Thirty one SSR primer pairs of 220 (∼14%) tested led to PCR amplification of discrete fragments using cotton leaf cDNA as template. Sequence analysis showed 25% of 24randomly selected cDNA clones amplified with different SSR primer pairs contained repeat motifs. We further showed that sequences from the SSR-containing cDNAs were conserved across G. barbadense and G. hirsutum, revealing the importance of the SSR markers for comparative mapping of transcribed genes. Data mining for plant SSR-ESTs from the publicly available databases identified SSRs motifs in many plant species,including cotton, in a range of 1.1 to4.8% of the submitted ESTs for a given species. Tumor necrosis factor (TNF) is a key regulator of inflammatory responses and has been implicated in many pathological conditions. We used structure-based design to engineer variant TNF proteins that rapidly form heterotrimers with native TNF to give complexes that neither bind to nor stimulate signaling through TNF receptors. Thus, TNF is inactivated by sequestration. Dominant-negative TNFs represent a possible approach to anti-inflammatory biotherapeutics, and experiments in animal models show that the strategy can attenuate TNF-mediated pathology. Similar rational design could be used to engineer inhibitors of additional TNF superfamily cytokines as well as other multimeric ligands.  相似文献

Four-week old plants of pepper (Capsicum annuum L.) cultivar ‘Jawala’ were sprayed with an aqueous solution of chlorflurenol at concentrations of 50, 100, 150 or 200 mg/l. Chlorflurenol reduced the growth of the main shoot and the area of the leaf, weakened apical dominance, increased the growth and number of lateral branches, and reduced the growth and number of stamens. Induction of stamenless (pistillate) flowers was an interesting effect. The size, viability and germination of pollen grains were reduced by chlorflurenol. It caused an appreciable promotion in fruit-set up to a concentration of 150 mg/l and a drastic reduction at 200 mg/l, accompanied by increase and decrease of yield, respectively. Parthenocarpy was observed at 100, 150 and 200 mg/l.  相似文献

The oxidant/antioxidant balance of rabbits naturally infected with Psoroptes cuniculi and treated with ivermectin +/− vitamins A, D3, E, and H supplementation was investigated. Two groups of seven mixed ♂ and ♀, 6-to-8 month-old New Zealand White rabbits, diagnosed Psoroptes mites-positive by skin scraping examination and seven clinically healthy control rabbits were examined. Blood samples were obtained on day 0 and at 28 days post-therapy to determine oxidative stress indices. On day 0, the levels of lipid peroxides were significantly higher (P ≤ 0.01) in the Psoroptes-infected rabbits compared with the healthy controls while those of reduced glutathione and the activities of the antioxidant enzymes glutathione peroxidase, glutathione-S-transferase, catalase, and superoxide dismutase were significantly lower (P ≤ 0.01). Vitamin supplementation of the ivermectin-treated rabbits revealed both faster clinical (14 days) and parasitological (10 days) recovery. It was concluded that significant alteration of oxidant/antioxidant balance is a factor in the pathogenesis of P. cuniculi infestation of rabbits, and recovery can be enhanced by combining ivermectin treatment with vitamin A, D3, E, and H supplementation.  相似文献

Phytoremediation is a promising approach for reclamation of salt-affected soil. Phytoextraction is the most commonly used process, which exploits plants to absorb, immobilize, and accumulate salt in their shoots. In this study, halotolerant plant growth-promoting rhizobacteria (PGPR) were isolated from the rhizosphere of wild grasses growing naturally in salt-affected areas of Lucknow, Uttar Pradesh (India) and were tested for their efficacies of salt-tolerance and plant growth-promoting (PGP) abilities. Based on 16S rRNA sequences, the most efficient halotolerant isolates possessing PGP traits were identified as Pseudomonas plecoglossicida (KM233646), Acinetobacter calcoaceticus (KM233647), Bacillus flexus (KM233648), and Bacillus safensis (KM233652). Application of these isolates as bio-inoculants significantly (P < 0.05) increased the growth and bacoside A yield of a medicinal plant, Bacopa monnieri (L.) Nash, grown on natural salt-affected soil. The phytoremediation of salt-affected soil was evident by the substantial increase in shoot Na+:K+ ratio of bio-inoculant-treated plants. When compared to un-inoculated control plants, the soil physico-chemical properties of bio-inoculant-treated plants were improved. The shoot and root biomass (fresh and dry weights), soil enzymes, and soil nutrient parameters showed significant positive correlations with the shoot Na+:K+ ratio. Consequently, the halotolerant PGPR screened in this study could be useful for the reclamation of saline soils concomitant with improved plant growth and bacoside A yield.  相似文献

Experiments were conducted to enhance the growth of common carp (Cyprinus carpio), rohu (Labeo rohita) and Mozambique tilapia (Oreochromis mossambicus) through use of sugarcane bagasse as substrate. Bagasse was suspended in water with or without supplementation with fertilizers. Bagasse supplemented with cattle dung and urea favoured higher zooplankton production and significantly (p < 0.05) increased fish growth by over 50% compared to bagasse or fertilizers on their own. This higher production of fish is attributed to bacterial biofilm promoted on the substrate which, apart from forming food for zooplankton and fish, contributed to improved water quality by lowering ammonia.  相似文献
