A field population of Plutella xylostella from Malaysia (SERD4) was divided into five sub-populations and four were selected (G2-G5) with the Bacillus thuringiensis insecticidal crystal (Cry) toxins Cry1Ac, Cry1Ab, Cry1Ca and Cry1Da. Bioassay at G6 gave resistance ratios of 88, 5, 2 and 3 for Cry1Ac, Cry1Ab, Cry1Ca and Cry1Da respectively compared with the unselected sub-population (UNSEL-SERD4). The Cry1Ac-selected population showed little cross-resistance to Cry1Ab, Cry1Ca and Cry1Da, (3-, 2- and 3-fold compared with UNSEL-SERD4), whereas the Cry1Ab-SEL sub-population showed marked cross-resistance to Cry1Ac (40-fold), much greater than Cry1Ab itself. In contrast, the Cry1Ca- and Cry1Da-SEL sub-population showed little if any cross-resistance to Cry1Ac and Cry1Ab. The mode of inheritance of resistance to Cry1Ac was examined in Cry1Ac-selected SERD4 by standard reciprocal crosses and back-crosses using a laboratory insecticide-susceptible population (ROTH). Logit regression analysis of F1 reciprocal crosses indicated that resistance to Cry1Ac was inherited as an incompletely dominant trait. At the highest dose of Cry1Ac tested, resistance was recessive, while at the lowest dose it was almost completely dominant. The F2 progeny from a back-cross of F1 progeny with ROTH were tested with a concentration of Cry1Ac that would kill 100% of ROTH. The mortality ranged between 50 and 95% in seven families of back-cross progeny, which indicated that more than one allele on separate loci were responsible for resistance to Cry1Ac. 相似文献

Genetics and mechanism of resistance to deltamethrin in a field population of Spodoptera litura (Lepidoptera: Noctuidae) 总被引:2,自引:0,他引:2
BACKGROUND: Spodoptera litura (F.) causes enormous losses in many economically important crops. The genetics of insecticide resistance has been extensively studied in several insect pests, but there is a lack of information on S. litura. Therefore, the genetics and mechanisms of the resistance of S. litura to deltamethrin were investigated. RESULTS: Bioassays at generation G1 gave resistance ratios of 9, 5, 41, 52 and 49 for deltamethrin, cypermethrin, profenofos, chlorpyrifos and triazofos respectively, when compared with the susceptible Lab-PK strain. Bioassays at G4 with a deltamethrin-selected population (Delta-SEL) showed that selection gave resistance ratios of 63 and 7 for deltamethrin when compared with the Lab-PK and UNSEL strains respectively. Cross-resistance to other insecticides tested was observed in the selected population. A notable feature of the Delta-SEL strain was that resistance to deltamethrin, cypermethrin, profenofos and chlorpyrifos did not decline over the course of five generations. Synergism tests with microsomal oxidase (MO) and esterase-specific inhibitors indicated that the deltamethrin resistance was associated with MO and, possibly, esterase activity. Reciprocal crosses between the Delta-SEL and Lab-PK strains indicated that resistance was autosomal and incompletely dominant. A direct test of monogenic inheritance suggested that resistance to deltamethrin was controlled by more than one locus. CONCLUSION: Stability and dominance of resistance and cross-resistance suggest that insecticides with different modes of action should be recommended to reduce pyrethroid selection pressure. 相似文献

Background: Through its membrane and intracellular receptors, vitamin D regulates many vital functions in the body including its well known actions on musculoskeletal system. Growing body of evidences demonstrate that vitamin D undergoes some of behavioral aspects of neurocognition. The present study was designed to evaluate the effect of food regimens without vitamin D or with a supplement of 1,25(OH)2D3 on spatial performance of adult rats. Methods: The animals were trained in the Morris water maze to find a hidden platform. The time spent and the distance traveled to find the platform, speed of navigation and the percentage of unsuccessful trials were considered for assessment of the task learning. Results: Our findings indicated that the vitamin D-deprived rats had a significant lower performance compared to both the controls and the animals receiving 1,25(OH)2D3 supplementation. Concerning the unsuccessful trials, lack of vitamin D resulted in the highest failures in the maze navigation. The regimen with additional 1,25(OH)2D3 did not considerably influence learning of the maze task. Conclusion: We concluded that while vitamin D deficiency deteriorates the spatial task learning, the 1,25(OH)2D3 supplementation did not effectively underlie the maze performance. Polystyrene (PS) composites with nanofibrous structure consisting of multi-walled carbon nanotubes (MWCNTs) with 0-10 wt.% of nanofiller have been fabricated via electrospinning technique. The surface morphology and thermal properties of the composites were evaluated by scanning electron microscopy (SEM) and thermo-gravimetric analysis (TGA). The SEM analysis of the composite nanofibers samples revealed that the average diameter of the nanofibers increases with increasing MWCNTs content. The resultant MWCNTs/PS composite nanofibers diameters were in the range of 391±63 to 586±132 nm. The thermal stability of composites was increased after addition of MWCNTs to PS matrix. The electrical conductivity of the composites with different weight percentage of MWCNTs was investigated at room temperature. Electrical conductivity of MWCNTs/PS composite nanofiber followed percolation theory having a percolation threshold V c= 0.45 vol% (~0.75 wt. %) and critical exponent q=1.21. The electrical conductivity and thermal properties confirmed the presence of good dispersion and alignment MWCNTs encapsulated within the electrospun nanofibers. The electromagnetic interference (EMI) shielding effectiveness of the MWCNTs/PS composites was examined in the measurement frequency range of 8.2-12.4 GHz (X-band). The total EMI shielding efficiency of MWCNTs/PS composite nanofibers increased up to 32 dB. The EMI shielding results for MWCNTs/PS composite nanofibers showed that absorption loss was the major shielding mechanism and reflection was the secondary mechanism. The present study has shown the possibility of utilizing MWCNTs/PS composite nanofibers as EMI shielding/absorption materials. 相似文献

Genetic inheritance of resistance to Bacillus thuringiensis var kurstaki (BTK) was examined in a diamondback moth (Plutella xylostella) population collected from the Melaka region of Malaysia. A BTK‐selected sub‐population (BTK‐SEL) which was more than 100‐fold resistant to BTK compared with a susceptible (ROTH) population of P xylostella was used with standard reciprocal crosses and back‐crosses between ROTH and BTK‐SEL. Logit regression analysis of F 1 reciprocal crosses indicated that BTK resistance was inherited as an incompletely recessive autosomal trait and controlled by a single locus. In contrast, other studies have shown that resistance to Cry1Ac is inherited as an incompletely dominant autosomal trait in a Cry1Ac‐selected sub‐population of the same Melaka population. Spermatozoa concentration, ionic composition, osmolality, glucose and total protein contents of seminal plasma and sperm motility were determined in Barbus sharpeyi (Cyprinidae, Teleosotei). Spermatozoa concentration ranged from 9.77 to 20.20 × 109 spermatozoa mL?1. Osmolality (mOsmol kg?1) and ionic contents (mM L?1) of the seminal plasma were 274.5±9.0, 70.0±3.4 Na+, 28.8±0.9 K+, 101.7±3.1 Cl?, 0.9±0.1 Mg2+ and 2.1±0.1 Ca2+ respectively. Total protein and glucose were 5.3±0.2 g L?1 and 76.7±4.3 mM L?1 respectively. Sperm motility was initiated in a hypo‐osmotic condition, composed of either an ionic (KCl or NaCl) or a non‐ionic (sucrose) activation medium. Duration of sperm motility was very short: <2 min after activation in distilled water. Percentage of motile spermatozoa was significantly higher in an activation medium containing NaCl compared with that of distilled water. An activating medium containing NaCl or KCl higher than 150 mM or sucrose higher than 275 mM totally inhibited the activation of sperm motility. Immediately after sperm activation, wave(s) propagated along the flagellum, but waves were restricted to the proximal part of the flagellum (close to the head) at 1 min post activation. Sperm quality of Barbus barbus L. was compared among the three following dietary regimes: Group A, fed 100% commercial diet (Karpico™ containing 33% crude
protein and 6% fat), Group B, fed 78% commercial diet and 22% frozen chironomid (Chironomus plumosus) larvae, and Group C, fed 56% commercial diet and 44% frozen chironomid larvae. Concentrations of polyunsaturated fatty acids
(PUFAs) in Group A, B, and C were 39.1, 42.0, and 44.6, respectively, as a percentage of total fatty acids. Sperm morphology,
volume, concentration and motility, total number of spermatozoa, and osmolality of the seminal plasma were compared during
the spawning season. Dietary regime did not influence sperm volume, concentration, or total number of spermatozoa, osmolality
of seminal plasma, or the percentage of motile sperm, but significantly affected sperm morphology (except for anterior and
posterior parts of the midpiece) and sperm velocity (P < 0.05). Groups B and C showed similar sperm characteristics during the spawning season compared to Group A. Almost all parameters
changed either among or within groups during the spawning season, suggesting differences in terms of the optimal time for
sperm collection. The best time for sperm collection was March for Group A, but April for Groups B and C, when the osmolality
of the seminal plasma measured 289 mOsmol kg−1 and sperm motility was maximal. Spermatogenesis, hydration, and cell decomposition were confirmed as the three major parameters
controlling sperm characteristics during the spawning season. The possible correlation between sperm morphology and motility

The present investigation was carried out to estimate the interrelationships and repeatabilities of various variability parameters,
i.e., genotypic (σ2g) and phenotypic (σ2p) variances, genotypic (GCV) and phenotypic (PCV) coefficients of variation, broad-sense heritability (hb2), genetic advance (GA), and genetic gain (GG), using data derived from a number of plant characters [seed yield (SY), days
to heading (DH) and maturity (DM), plant height (PH), and thousand kernel weight (TKW)] of 17 spring safflower genotypes grown
in 27 environments in Iran during 2003–2005. The analysis of variance for the five characters revealed significant differences
among the genotypes in most of the environments, indicating a very high variability within the genotype. High genotypic and
phenotypic variances in the quantitative traits, particularly for SY, were markers of increased success. A close correlation
between GCV and PCV was observed for all the traits, indicating that all of the characters were less influenced by the environment
and that the variability which exists in these characters is under genetic influence; however, the relatively higher values
of PCV indicate the predominant role of environment. High expected genetic advances were observed for SY, but this trait showed
large fluctuations in different environments (97.4%). High and low mean values of heritability were observed for DH (74.3%)
and SY (59.7%), respectively, whereas high and low mean values of genetic gain were found for SY (60.3%) and DM (4.2%), respectively.
High heritability coupled with low GA as a percentage of the mean was observed for DH and MD, whereas moderate heritability
with moderate GA as a percentage of the mean was exhibited for SY. Analysis of correlation among the parameters showed that
they were strongly and constantly correlated with each other, but none of them were consistently well correlated with the
heritability parameter (hb2). Based on estimates of the genetic variability parameters within each trial, the ranking of genotypes in the low-performance
subset for all traits differed from that in the high-performance subset. This result indicates poor repeatability for the
genetic variability parameters. The estimates of the parameters in the multi-year trials were almost constant and repeatable,
whereas the responses over years showed poor repeatability. 相似文献

Mango (Mangifera indica L.) is one of the most economically important trees which has been cultivated for several centuries. The tree is susceptible
to insect damage and is attacked by leaf eaters, termites, root grubs, sapsuckers, gall formers, stem borers, pod borers and
fruit borers. The mango mealybug Drosicha mangiferae Green (Hemiptera: Monophlebidae) is one of the most destructive pests in the Indo-Pak subcontinent, sometimes causing premature
fruit drop. A combination of several management strategies such as use of entomopathogens, chemical insecticides and burning
of adult females has been used to control D. mangiferae. Overreliance on insecticides has eliminated natural enemies, and we were therefore interested in investigating the impact
of alternative insect pest management strategies such as cultural and mechanical techniques on D. mangiferae. The results of our studies suggested that different bands significantly stopped the upward movement of overwintering and
surviving populations of D. mangiferae. To augment the bands’ results, mounding the tree with debris on a plastic sheet once a year was the most effective technique
and resulted in maximum reduction of the nymphal population of D. mangiferae. The newly developed Haider band—designed in our laboratory in the present studies—was very successful in the field in controlling
the upward movement of newly hatched nymphs on trees: only 0.8% of the nymphs crossed the band. Only 7% to 10% of the nymphs
crossed plastic sheets, polyethylene sheets and funnel bands. The cotton and gunny bag (jute) bands were the least effective:
42% of the nymphs managed to cross them. The results suggested that employing cultural practices and bands together could
reduce the need for insecticide treatments and offer a sustainable method for D. mangiferae control. 相似文献

BACKGROUND: Spodoptera litura (F.) is a cosmopolitan pest that has developed resistance to several insecticides. The aim of the present study was to establish whether an emamectin‐selected (Ema‐SEL) population could render cross‐resistance to other insecticides, and to investigate the genetics of resistance. RESULTS: Bioassays at G1 gave resistance ratios (RRs) of 80‐, 2980‐, 3050‐ and 2800‐fold for emamectin, abamectin, indoxacarb and acetamiprid, respectively, compared with a laboratory susceptible population Lab‐PK. After three rounds of selection, resistance to emamectin in Ema‐SEL increased significantly, with RRs of 730‐fold and 13‐fold compared with the Lab‐PK and unselected (UNSEL) population respectively. Further studies revealed that three generations were required for a tenfold increase in resistance to emamectin. Resistance to abamectin, indoxacarb, acetamiprid and emamectin in UNSEL declined significantly compared with the field population at G1. Furthermore, selection with emamectin reduced resistance to abamectin, indoxacarb and acetamiprid on a par with UNSEL. Crosses between Ema‐SEL and Lab‐PK indicated autosomal and incomplete dominance of resistance. A direct test of a monogenic model and Land's method suggested that resistance to emamectin was controlled by more than one locus. CONCLUSION: Instability of resistance and lack of cross‐resistance to other insecticides suggest that insecticides with different modes of action should be recommended to reduce emamectin selection pressure.
