Clavibacter michiganensis subsp. sepedonicus, a Gram positive bacterium that causes bacterial ring rot of potato, was studied in eggplant, an alternate host, using strains that differed in phenotype. Two factors affecting virulence, the ability to induce a hypersensitive response (HR) and cellulase production, were studied. A plasmid-free isolate of C. michiganensis subsp. sepedonicus that causes HR on tobacco but is unable to produce cellulase multiplied efficiently in planta, but caused only weak symptoms. In contrast, a strain that is unable to induce HR on tobacco but produces cellulase was impaired in the ability to multiply in the host and caused no symptoms. When the two non-virulent strains were coinoculated into eggplants, typical disease symptoms developed. This enhancement was not due to formation of a new phenotype or significant increases in population density of either of the strains. Our results suggest that both cellulase production and the ability to induce HR are required for a successful infection process and disease induction by C. michiganensis subsp. sepedonicus. Our results additionally suggest that the ability to induce HR on non-host plants is required for multiplication in the host plant, whereas cellulase expression is necessary for induction of disease symptoms.  相似文献

This experiment was a part of a research on the soil community in anthropogenous birch stands in Finland. In that study it was found that communities of Collembola are similar in birch stands of different origin (cultivated field or spruce forest), while the communities of Oribatida are essentially different. When compared to original spruce forest, the communities of both groups are different.Cultivation eliminates the populations of most microarthropod species, that have to disperse after reforestation from the surrounding areas. The aim of the experiment was to study the ambulatory dispersal of soil microarthropods. It was carried out in plastic boxes filled with an intact block of defaunated surface soil taken from a birch stand that was established ca. 30 years earlier on a cultivated field. A strip of intact spruce forest soil was placed at one end of the boxes to harbour the source populations. At the opposite end we placed a row of pitfall traps. Soil samples were taken every two weeks at increasing distances from the source soil using a metal corer, and animals were extracted in a high gradient apparatus. The pitfalls were also emptied every two weeks. The experiment lasted for ten weeks.Pitfall and soil sample data gave an estimation of the maximum dispersal rate for each species or genus. The distance that the populations could potentially disperse in 30 years (age of the stand) indicated that some, but not all of the species could have actively migrated to the central parts of the birch stand (30 m).  相似文献

The combined and separate effects of Cd and wood ash on Archaea from coniferous forest humus were studied in a microcosm experiment. Nonmetric multidimensional scaling of the denaturing gradient gel analysis of polymerase chain reaction amplified 0.9 kb 16S ribosomal DNA fragments revealed changes in archaeal communities due to the ash treatments. Cd with or without ash did not further influence the result. Representatives of the ash and control communities were cloned, grouped by restriction fragment length polymorphism analysis and finally sequenced. All sequences belonged to non-thermophilic Crenarchaea.  相似文献

Natural regeneration of native tree species in the understoreys of fast-growing plantation tree species (Acacia mangium, Gmelina arborea and Paraserianthes falcataria) and in adjacent natural riverine forest and non-cultivated grassland areas were studied on Imperata cylindrica grassland sites in Riam Kiwa plantation area, South Kalimantan, Indonesia. Seedling and sapling densities as well as species composition and richness were assessed in a total of 35 ha by a systematic line plot survey.Floristic composition was similar in A. mangium and P. falcataria stands which differed distinctly from G. arborea stands and Imperata grassland area. Riverine forest was characterized by later successional species. In A. mangium and P. falcataria stands, a total of 63 species from 24 families were recorded. Seedling stock consisted merely of pioneer and secondary species, but also several fairly commonly occurring primary forest species. Euphorbiaceae was the most common family representing 42% of sapling and 29% of seedling densities. The most abundant regeneration occurred in A. mangium stands (4,706 seedling/sapling ha-1), followed by P. falcataria (2,242 ha-1) and G. arborea (554 ha-1). Seedling recruitment was significantly lower in plots dominated by grass vegetation than those dominated by no vegetation or shrub vegetation. Breast height diameter and basal area of canopy trees as well as coverage and height of ground vegetation did not explain seedling/sapling density. Results show that plantation establishment with suitable fast-growing tree species facilitates recruitment of a variety of native tree species. This provides potential for rapid restoration of forest ecosystem and for further development of the mixed stands of fast-growing exotics and naturally regenerated native species.  相似文献

Arctic soil carbon (C) stocks are threatened by the rapidly advancing global warming. In addition to temperature, increasing amounts of leaf litter fall following from the expansion of deciduous shrubs and trees in northern ecosystems may alter biogeochemical cycling of C and nutrients. Our aim was to assess how factorial warming and litter addition in a long-term field experiment on a subarctic heath affect resource limitation of soil microbial communities (measured by thymidine and leucine incorporation techniques), net growing-season mineralization of nitrogen (N) and phosphorus (P), and carbon turnover (measured as changes in the pools during a growing-season-long field incubation of soil cores in situ). The mainly N limited bacterial communities had shifted slightly towards limitation by C and P in response to seven growing seasons of warming. This and the significantly increased bacterial growth rate under warming may partly explain the observed higher C loss from the warmed soil. This is furthermore consistent with the less dramatic increase in the contents of dissolved organic carbon (DOC) and dissolved organic N (DON) in the warmed soil than in the soil from ambient temperature during the field incubation. The added litter did not affect the carbon content, but it was a source of nutrients to the soil, and it also tended to increase bacterial growth rate and net mineralization of P. The inorganic N pool decreased during the field incubation of soil cores, especially in the separate warming and litter addition treatments, while gross mineralized N was immobilized in the biomass of microbes and plants transplanted into the incubates soil cores, but without any significant effect of the treatments. The effects of warming plus litter addition on bacterial growth rates and of warming on C and N transformations during field incubation suggest that microbial activity is an important control on the carbon balance of arctic soils under climate change.  相似文献

The aim of the current study was to identify factors influencing the technological functionality of fermented bran. The influences of fermentation type and type of wheat bran on the microbial community, bioactivity, arabinoxylans (AX), and activity of xylanases were studied in the bran ferments. Furthermore, technological quality of ferments was established by using them to replace wheat in baking with a 20% substitution level. Solubilization of AX and endogenous xylanase activity of bran were influenced by the type of bran, fermentation type, and conditions. Peeled bran had a clearly reduced microbial load and different microbial community in comparison to native bran. Bran from peeled kernels contained 10‐fold lower activities of endogenous xylanases in comparison to native bran. Yeast fermentation of bran from peeled kernels increased the level of folates (+40%), free phenolic acids (+500%), and soluble AX (+60%). Bread containing yeast‐fermented peeled bran had improved volume (+10–15%) and crumb softness (25–35% softer) in comparison to unfermented counterparts. Solubilization of AX during the 20 hr fermentation and decreased endogenous xylanase activity are proposed as the main reasons for the improved technological functionality of fermented bran.  相似文献

Protecting biodiversity within separate set-aside conservation areas has not been effective enough to halt its loss. Thus, new approaches to conserve biodiversity alongside production are needed. The non-market values of a forest may play an essential role when the forest owner decides the use of their land. However, so far the service offerings other than related to timber production, have been scant. The mismatch between decision support services offered and the service interests of forest owners may result in the objectives of forest owners remaining unfulfilled. The aims of this study were to explore the links between family forest owners’ forest management preferences and their objectives for the forest and secondly their preferences for decision support services. Data were collected in a postal survey in the Northern Karelia region, Finland in spring 2014. Data consist of 298 survey answers that were analysed using multi-variate analyses. Two typologies were combined: clustering of forest ownership objectives and the preferred forest management style. We found that the forest owner’s objectives were demonstrated by their preferred way of managing the forest. Opinions about different decision aid services varied between cluster groups. The groups emphasizing nature values considered biodiversity related information about their forest more necessary than other groups. They were also less satisfied with the usability of the forest management plan. Forest advisory services should better acknowledge the prevalence of multiple objectives also among forest owners who are interested in timber selling. Developing services for forest owners with diverse socio-economic backgrounds, information needs and objectives is important.  相似文献

Ethnobotanical studies and scorecard-based assessments have been used in identification and prioritisation of indigenous fruit trees with domestication and income potential at local levels. Less has been done to systematically identify species with potential for international markets. This study compared the applicability of the two methods to identify indigenous fruits with economic potential for export. The ethnobotanical study consisted of household and market surveys based on questionnaires, focus group discussions and key-informant interviews. The scorecard was based on pre-determined scoring criteria applied to species identified in the ethnobotanical study. Priority indigenous fruits identified in the ethnobotanical study included Canarium schweinfurtii, Vangueria apiculata, Garcinia buchananii and Tamarindus indica whereas the scorecard assessment identified a different set of species; i.e. V. apiculata, G. buchananii, Myrianthus arboreus, Pseudospondias microcarpa, Phoenix reclinata and Rhus vulgaris as having export potential. The ethnobotanical methods were effective in identifying species with local economic potential and hence high likelihood of adoption by local farmers while the scorecard method was found more useful to identify species with export potential.  相似文献

Peroxidase activity and isoenzymes of fresh wood samples of the third shoot of 12-year old trees and from the sapwood, transition zone and heartwood of c. 60-year old stems of Scots pine (Pinus sylvestris L.) were investigated. Wood samples were ground at −30°C, extracted, and the extracts concentrated c. 20-fold for peroxidase activity assays (guaiacol method) and for IEF-PAGE. At least 11 major isoenzymes could be found in the gels. Even the heartwood contained some peroxidase isoenzymes. Isoenzyme patterns of the juvenile wood did not change with the season. However, juvenile wood showed the highest peroxidase activity at the end of the growing season. Peroxidase activity decreased from the outer sapwood towards the heartwood. Thin sections of different wood zones stained for peroxidase revealed activity in ray parenchyma and resin canal epithelial cells. Intensive staining was localised in the bordered pits of vertical and ray tracheids, and in the end walls of ray parenchyma cells.  相似文献
