To ascertain the distribution of alpha chains within the collagen molecule, intramolecular cross-links were introduced into tropocollagen by reacting formaldehyde with dilute homogeneous molecular dispersions of collagen extracted from lathyritic guinea pig skin. Denaturation, chromatography on carboxymethyl cellulose, measurement of molecular weight, and analyses of the amino acids of the cross-linked product indicate that most, if not all, of the collagen molecules consist of two alpha1 chains and one alpha2 chain [(alpha1)(2)(alpha2)(1)].  相似文献

For characterization of ether-extractable fat content (EE), L*, a*, and b* color, and water-holding capacity (WHC), 12 muscles or muscle groups were dissected from 48 pork carcasses of boars, barrows, or gilts that were fed diets either at minimum (LO) or 1% above (HI) their protein requirements and slaughtered in two separate trials at 100 or 110 kg. In both trials across muscles, gilts and boars had lower (P < .05) EE than barrows. In the 110-kg trial, boars had lower (P < .001) EE than gilts. In the 100-kg trial, boars on LO diets had lower (P < .001) WHC than all other groups, and both boar groups had lower (P < .05) WHC than gilts. No differences (P > .05) in WHC were seen in the 110-kg trial. In the 100-kg trial, gilts had lower L* (P < .05) than boars and barrows, but in the 110-kg trial boars had lower L* (P < .05) than barrows and gilts. The lowest (P < .05) a* values were for boars in the 100-kg trial and for boars on LO diets in the 110-kg trial. In both trials, the serratus ventralis had more (P < .001) EE than all other muscles. In both trials, the semitendinosus had higher (P < .001) L* and the longissimus had lower (P < .01) a* and b* than all other muscles. The numerous differences observed among muscles may help identify optimal uses for the entire pork carcass.  相似文献

An antigen-capture enzyme-linked immunosorbent assay (cELISA) based on monoclonal antibodies (mAb) was set up and evaluated for selective detection of salmonid antibody responses to the antigen P1, which is a weakly immunogenic exoprotease of typical Aeromonas salmonicida. This new assay permits a specific determination of anti-protease-antibodies, without antigen purification. Serum antibodies induced by the strongly immunogenic lipopolysaccharide could reliably be discriminated from anti-P1-antibodies. Antibody titres of 45 experimental antisera recorded by cELISA were moderately correlated with titres determined by routinely used indirect ELISA (iELISA) by detecting partially different antibody populations (r=0.753). Substitutions of immunoreactants and confirmatory immunoblotting strongly suggest that the mAb-based assay selectively recognises antibodies directed to epitopes of native protease. A conjugate of inhibited protease and cationized bovine serum albumin (cBSA) was found to engender a significant anti-protease-response in three salmonid species (P<0.05), whereas the unconjugated antigen and Apoject 1-Fural were proved to be ineffective. Recorded specific antibody titres were as high as 1:381,400, indicating a considerable enhanced immunogenicity of cBSA-conjugated P1 and high assay sensitivity. The established cELISA offers a promising approach to further improvement of monitoring fish humoral immune response to surface accessible epitopes of the immunosuppressive exoprotease, P1, and to scrutinize its protective significance.  相似文献

In two separate 2 x 2 factorial arrangements, 69 male, crossbred lambs were allotted randomly to the following treatments: 1) nonimplanted (NI) rams, 2) implanted (I) rams, 3) NI wethers, and 4) I wethers. In Trial 1, 36 lambs were allotted to treatment groups at birth (n = 9) and I lambs were implanted with 12 mg of zeranol between 1 and 3 d of age and again at weaning (average age of 62 d). Lambs were slaughtered at three time-constant end points of 78, 93, and 107 d on feed postweaning (average age of 155 d). Rams grew faster postweaning, were more efficient in their feed conversion, were heavier at slaughter, and had lower numerical yield grades than did wethers (P less than .05). Implanted lambs tended (P = .08) to grow faster and were (P less than .05) more efficient in their feed conversion than NI lambs. Rams produced heavier (P less than .05) trimmed subprimal shoulders, loins, and legs and had (P less than .05) a higher percentage of their carcass weight in the subprimal shoulder than did wethers. During Trial 2, NI rams (n = 8), I rams (n = 8), NI wethers (n = 8), and I wethers (n = 9) were allotted to treatment groups, and I lambs were implanted at average ages of 14, 55, and 98 d. Thirty-five zeranol-implanted (I) and nonimplanted (NI) ram and wether lambs representing four treatments (implanted rams [IR], nonimplanted rams [NIR], implanted wethers [IW], and nonimplanted wethers [NIW]) were evaluated for meat palatability and muscle collagen characteristics. Rib (longissimus muscle, LM) chops from I lambs were juicier (P less than .05) than rib chops from NI lambs. Chops from IR lambs had more (P less than .05) detectable connective tissue and lower myofibrillar and overall tenderness scores than chops from NIR, IW, or NIW lambs. Warner-Bratzler shear (WBS) values tended to be higher (P = .06) for LM chops from rams than for those from wethers, but WBS values for Biceps femoris (BF) chops were similar (P greater than .05) for rams and wethers. Implanting did not affect (P greater than .05) WBS values. Rams had more (P less than .05) LM heat-labile (soluble, SC), nonheat-labile (insoluble, IC), and total collagen (TC) and a higher (P less than .05) percentage of SC (SC/TC) than did wethers. Soluble collagen, TC, and percentage of SC for the BF were higher (P less than .05) and IC tended (P = .09) to be higher in chops from rams than in those from wethers. Implanting did not affect (P greater than .05) collagen amount or solubility. Serum nonprotein hydroxyproline (NPHP) was higher (P less than .05) in rams than in wethers throughout the feeding period and tended (P = .05) to be higher at slaughter.
