1. The effects of dietary or parenteral administration of ascorbic acid on the adverse effects of excess dietary tyrosine were investigated with young male White Leghorn chicks in a 2‐week experiment.

2. Addition of 10 g ascorbic acid/kg to the control diet (without excess tyrosine) produced no beneficial effects on performance. Excess dietary tyrosine caused depressions in all measures of performance.

3. Adding 0.1, 1, 10 or 20 g ascorbic acid/kg to the diet containing excess tyrosine tended to improve performance.

4. Subcutaneous injection of 50 mg ascorbic acid/bird d to chicks receiving excess tyrosine brought about a significant improvement in body‐weight gain.

5. The elevation of plasma free tyrosine caused by excess dietary tyrosine decreased as dietary ascorbic acid increased.

6. Ascorbic acid can alleviate, though not completely counteract, the adverse effect of excess dietary tyrosine.  相似文献

Grain hardness, amylose content, and glutenin subunit composition are critical determinants for end‐use properties of wheat. To improve the end‐use properties of domestic wheats, we studied these traits between the Australian and North American wheat classes exported to Japan in 2009 and 2011 by analyzing the corresponding alleles. Most hard classes had Pina‐D1b or Pinb‐D1b. A partial waxy allele (Wx‐B1b) was found in all Australian Standard White (ASW) seeds in 2009 and two‐thirds of ASW seeds in 2011. All or most American hard wheat seeds had Glu‐D1d. Most U.S. Western White (WW) seeds had a null allele (Glu‐A1c) or alleles that lacked one of the two Glu‐B1 subunits. Most hard red winter (HRW) seeds had Glu‐B3b or Glu‐B3g. Quality characteristics of these classes seemed to be consistent with these results. In addition, we also found new Glu‐1 and Glu‐3 alleles in HRW and WW. These results suggested that although there are variations in its allelic composition from year to year, each class has unique quality‐related alleles corresponding to its end use. We proposed two matrices for classification of starch properties on the basis of Pin and Wx allelic combinations and for classification of gluten strength on the basis of glutenin allelic combinations.  相似文献

For developmental competence of porcine embryos in vitro, it is important to improve the culture environment. The present study was performed to evaluate four different culture systems for in vitro matured porcine oocytes following intracytoplasmic sperm injection (ICSI); drop, well and two sizes of the well of the well (WOW) systems (500 and 1,000 microm in diameter). The cleavage rate on Day 2 and the mean cell number in blastocysts on Day 6 were not significantly different among the four treatments. However, the 1,000 microm WOW (24.6%) resulted in a significantly higher (P<0.05) blastocyst rate than those in the other culture systems (12.9, 14.8, and 7.1% for drop, well, and 500 microm WOW, respectively). The present study indicates that the microenvironment created by the 1,000 microm diameter WOW improves blastocyst production of in vitro matured porcine oocytes after ICSI, and that the effectiveness of the WOW system is dependent on the size (diameter) of the WOW.  相似文献

Giardia intestinalis antigen in fecal samples was examined in 361 dogs of 14 breeding kennels located at various areas in Japan, using a commercial enzyme-linked immunosorbent assay (ELISA) kit. G. intestinalis antigen was detected in 37.4% of the fecal specimens. All of the 14 breeding kennels were positive for G. intestinalis antigen with the range from 6.7 to 59.3%. The prevalence in puppies (54.5%) was significantly (p < 0.01) higher than that in adults (30.9%). There was no difference in prevalence between males and females, and between the puppies from the mother dogs positive and negative for Giardia antigen. In conclusion, G. intestinalis widely invaded the breeding kennels in Japan.  相似文献

The interferon-stimulated gene 15 (ISG15) is induced by type I interferon (IFN). Recent studies have revealed that like ubiquitin, ISG15 is conjugated with target proteins. In this study, the feline ISG15 (FeISG15) gene was cloned from feline IFNomega (FeIFNomega)-stimulated feline kidney epithelial (CRFK) cells. According to gene sequence results, cDNA was 474bp long and encoded a protein of 157 amino acids. The putative amino acid sequences showed 62.5-72.1% identity with those of other mammalian ISG15s. Similar to human and mouse ISG15, FeISG15 included tandem ubiquitin-like domains; its homology with feline ubiquitin was 36.3-39.5%. The LRLRGG conjugating motif was located only in the carboxyl terminal ubiquitin-like domain. FeISG15 also lacked the carboxyl terminal extension after the LRLRGG motif, which is present in mouse and human ISG15. Recombinant FeISG15 protein was expressed as a His-tagged fusion protein in Escherichia coli and purified by ion-exchange chromatography followed by affinity chromatography. Monoclonal anti-FeISG15 antibodies revealed free FeISG15 and FeISG15 conjugated with target proteins in cells after IFNomega stimulation by Western blotting analysis. Furthermore, mRNA of IFNgamma was detected from peripheral blood mononuclear cells (PBMCs) after stimulation with rFeISG15 extracellularly by RT-PCR. Taken together, these results suggested that FeISG15 had ubiquitin- and cytokine-like activity, as in other species.  相似文献

In Japan, Lactococcus garvieae infection has been the main fish disease in aquaculture. Although commercial oral and injectable vaccines have been used to prevent L. garvieae infection in Japan, L. garvieae has been isolated not only from unvaccinated fish but also from vaccinated fish in which immunity induced by vaccination had diminished. In order to obtain epidemiological information on this fish pathogen, we conducted biased sinusoidal field gel electrophoresis (BSFGE) pattern analysis and phage typing of L. garvieae isolates (n = 427) from fish in Japan. These isolates were obtained from 13 different fish species between 1980 and 2007. In the BSFGE analysis, L. garvieae isolates were classified into 17 groups (S1–S17) based on the SmaI digestion patterns and into four groups (A1–A4) based on the ApaI digestion patterns. Phage typing revealed five different phage susceptibility profiles (A–E) in L. garvieae isolates. Since 2005, comparisons of the results of phage typing and BSFGE have indicated the presence of a novel genotype (S16/A4) with phage type E. All the strains belonging to this type showed lincomycin sensitivity.  相似文献

In the present study, we tried to detect DNA for ribosomal RNA genes of piroplasma parasites from the liver or blood of 43 Japanese serows (Capricornis crispus) in Iwate Prefecture of Japan by polymerase chain reaction. Approximately 500-bp amplicons were obtained in 35 (81.4%) of the 43 samples by amplification for V4 hyper-variable regions of the 18S rRNA gene, and the amplicons were considered to be DNA of Theileria species. The complete nucleotide sequences (1,700 or 1709 bp) of the 18S rRNA gene were determined in 20 samples and were divided into 5 genotypes that were phylogenetically separated into two different lineages showing a polyphyletic relation. The Theileria DNAs of the two different lineages were considered to be those of distinct species.  相似文献

To assess the possible utility of a fungal gene for manganese-dependent peroxidase (MnP) produced by a transgenic plant in phytoremediation, we transformed hybrid aspen with a chimeric gene for MnP. Our gene construct allowed expression of the gene for MnP in plants and relatively high MnP activity was detected in the hydroponic medium in which roots of plants that expressed the transgene had been cultured. Some of our transgenic plants were able to remove bisphenol A from the medium more efficiently than wild-type plants. Our results demonstrate that, without any modification of the coding sequence, a chimeric gene for fungal MnP can be expressed in a woody plant, with secretion of active MnP from roots into the rhizosphere. Our strategy suggests new options using woody plants for phytoremediation.  相似文献

To determine the genotypes of Giardia intestinalis from domestic and wild animals in Japan, Giardia isolates obtained from feces of 24 dogs kept in households and breeding kennels, three companion cats, five dairy calves and three wild monkeys, Macaca fuscata, were genotyped using the 177 bp sequence of the glutamete dehydrogenase gene (gdh). The genotypes were assemblages A, C, D or A/D for dog isolates, Assemblage F for cat isolates, assemblages A or E for calf isolates and assemblage B for monkey isolates. This is the first report on the genotypes of Giardia isolates from cats, calves and wild monkeys in Japan.  相似文献

A gene encoding the mature Escherichia coli heat-labile enterotoxin (LT) lacking the nick site in the A subunit by deleting tripeptides was introduced in a vector pNH301 and expressed extracellularly as mutant molecule of holotoxin at high levels in Bacilus brevis HPD31-S5 of the host bacterium. The mucosal adjuvant activities of the produced mutant LT (mLT) preparation were studied in pigs and cattle. Intranasal immunization of pigs with the recombinant subunit vaccine of Erysipelothrix rhusiopathiae or the component vaccine of Bordetella bronchiseptica mixed with the mLT resulted in a substantial enhancement of both mucosal and serum-specific antibody levels. The immunized pigs were also protected when challenge-exposed intradermally with a highly virulent E. rhusiopathiae strain or challenge-exposed intranasally with a highly virulent strain of B. bronchiseptica. The mLT intranasally administered with recombinant intimin (an outer membrane adhesin) of E. coli O157:H7 also induced an elevation of IgA-specific antibody in the nasal secretion and saliva of calves as well as an elevation of IgG1-specific antibody level against the intimin in the sera and colostrum of cows. The three kinds tested protein antigens were poorly immunogenic when antigen administered intranasally alone. The mLT intranasally administered at a higher effective dose did not induce local adverse reactions or diarrhea in pigs and cattle. The present study demonstrates that the recombinant mLT produced using the B. brevis expression system might represent promising immunoadjuvants for the potential application of intranasal vaccines directed against infectious diseases in pigs and cattle.  相似文献
