There is a growing interest in highly fermentable dietary fibers having the potential to reduce risks of disease through the production of short-chain fatty acids (SCFA). Recently a digestion-resistant retrograded maltodextrin (RRM), classified as type 3 resistant starch was developed. Systematic work to determine its molecular and physiological properties was carried out to determine (1) the fraction resistant to digestion in vitro and in vivo, (2) its postconsumption effect on blood glucose in healthy volunteers, and (3) its in vitro fermentation pattern, at different ages, by use of pooled fresh human fecal inoculum. RESULTS: The digestion resistant fraction obtained in vivo from ileostomy patients (59.4%) is similar to that obtained by the AOAC method for measuring retrograded resistant starch (59.7%). The relative glycemic response after consumption of 50 g of RRM was 58.5% compared to glucose set as 100%. When exposed to colonic microbiota, in vitro obtained indigestible fractions behave similarly to those obtained in vivo in ileostomy patients. Fermentation of RRM and production of butyric acid is negligible during the first months of life but develops subsequently during weaning. In adults, RRM fermentation results in a high yield of SCFA, with butyrate representing 21-31 mol % of total SCFA. The high yield of SCFA during colonic fermentation, observed from weaning age on, as well as the potential to help reduce glycemic load may be of benefit to a number of health-related functions in the host. Further study on clear clinical end points is warranted.  相似文献

Determinations of serum folate and cobalamin concentrations in dogs have proved of considerable value for the identification and characterisation of chronic small intestinal disorders, but the microbiological assays used are time-consuming and technically demanding. Dual isotope radio-assays are more convenient and have been developed for the determination of folate and cobalamin in human beings. This study has evaluated such an assay for the determination of serum folate and cobalamin concentrations in dogs by direct comparison with microbiological assays used previously. Assays were performed on samples from 77 dogs, including controls and animals with confirmed or suspected chronic small intestinal disease. Regression analysis demonstrated a significant relationship between the two assays for serum folate concentrations (R=0–85; P=0–0001) and a definite trend for radioassay to give lower results than bioassay. There was also a significant relationship between bioassay and radioassay data for serum cobalamin (R=0–91; P=0–0001) with comparable absolute values for these two assays. Radioassay of serum samples from 31 clinically healthy dogs gave control ranges of 3–7 to 8-8 4mUg/litre for folate and 205 to 490 ng/litre for cobalamin. These ranges were similar to those calculated by comparison with the established ranges for bioassay using regression analysis, which predicted ranges of 4-4 to 8-4 μg/litre and 217 to 398 ng/litre for radioassay of serum folate and cobalamin, respectively. These data indicate that a dual isotope radioassay of serum folate and cobalamin may be used for dogs, and emphasise the need for laboratories to validate and establish their own control ranges for different assays.  相似文献

Purpose

This study aimed at evaluating the acute effects of arsenic and zinc to the warmwater aquatic oligochaete Branchiura sowerbyi. Relative sensitivity with the coldwater species Tubifex tubifex was compared. Implications for the use of B. sowerbyi in the risk assessment of sediments in the tropics are discussed.

Materials and methods

Water-only (96 h) and sediment (14 days) toxicity tests were conducted with both species evaluating a concentration series of arsenic and zinc. The tests were conducted considering the environmental conditions in the natural habitat of T. tubifex (predominantly temperate) and B. sowerbyi (predominantly tropical). Both lethal and sublethal endpoints (autotomy of the posterior body parts, abnormal behavior and appearance) were determined in the tests. The lethal (LC10 and LC50) and effect (EC10 and EC50) concentrations were also determined to assess metal sensitivity for both species.

Results and discussion

Both test species were more sensitive to Zn than As in water-only tests, which is in agreement with previous studies evaluating the toxicity of these metals to aquatic oligochaetes. Sublethal effects were generally noted at concentrations lower than those leading to mortality. The warmwater oligochaete B. sowerbyi was more sensitive to both metals tested than the coldwater species T. tubifex.

Conclusions

Study findings support the need for using indigenous tropical species in risk assessments in the tropics. In addition, sublethal effect parameters should be included in toxicity testing with aquatic oligochaetes.

Landscapes in the humid tropics are undergoing a continuous change in land use. Deforestation is still taking its toll on forested areas, but at the same time more and more secondary forests emerge where formerly agricultural lands and pastures are being abandoned. Regarding soil hydrology, the extent to which secondary succession can recover soil hydrological properties disturbed by antecedent deforestation and pasture use is yet poorly understood. We investigated the effect of secondary succession on saturated hydraulic conductivity (Ks) at two soil depths (0-6 and 6-12 cm) using a space-for-time approach in a landscape mosaic in central Panama. The following four land-use classes were studied: pasture (P), secondary forest of 5-8 years of age (SF5), secondary forest of 12-15 years of age (SF12) and secondary forest of more than 100 years of age (SF100), each replicated altogether four times in different micro-catchments across the study region. The hydrological implications of differences in Ks in response to land-use change with land use, especially regarding overland flow generation, were assessed via comparisons with rainfall intensities.Recovery of Ks could be detected in the 0-6 cm depth after 12 years of secondary succession: P and SF5 held similar Ks values, but differed significantly (α = 0.05) from SF12 and SF100 which in turn were indistinguishable. Variability within the land cover classes was large but, due to sufficient replication in the study, Ks recovery could be detected nonetheless. Ks in the 6-12 cm depth did not show any differences between the land cover classes; only Ks of the uppermost soil layer was affected by land-use changes. Overland flow - as inferred from comparisons of Ks with rainfall intensities - is more likely on P and SF5 sites compared to SF12 and SF100 for the upper sample depth; however, generally low values at the 6-12 cm depth are likely to impede vertical percolation during high rainfall intensities regardless of land use.We conclude that Ks can recover from pasture use under secondary succession up to pre-pasture levels, but the process may take more than 8 years. In order to gain comprehensive understanding of Ks change with land use and its hydrological implications, more studies with detailed land-use histories and combined measurements of Ks, overland flow, precipitation and throughfall are essential.  相似文献

Interest in native species is growing across the tropics as reforestation of degraded lands becomes more widespread. In this study four tree species native to Panama - Cedrela odorata, Pachira quinata, Samanea saman, and Tabebuia rosea - were grown on rural farms at two dry tropical sites in Panama for up to five years. Survivorship and growth data at these “on-farm” trials are compared to data recorded from nearby experimental or “species selection” trial sites and evaluated in terms of soil fertility and management. Participant farmers were also asked about their interest in planting trees in general as well as their interest in 61 species grown in the species selection trial.Although, on-farm survivorship was variable and generally lower than that found on the species selection trial, one species (S. saman) experienced high and consistent survivorship. High survivorship combined with growth data from farms at both sites for this species suggests it would be a good candidate for extension projects working with rural farmers. Survivorship of other species appears more sensitive to farmer management and/or local site conditions. Generally lower growth on the Los Santos farms as compared to the species-selection trial is attributed to the lower soil fertility (plant available P) at the on-farm sites compared to the species selection trial. In contrast, only one species - P. quinata - had a growth variable found to be significantly lower between the on-farm and species selection trial sites in Rio Hato. C. odorata, P. quinata T. rosea can all be used in on-farm conditions with consideration to specific site and management conditions.By 2009, approximately 80% of the farmers planting trees still wished to participate in tree planting activities. All of the farmers no longer wishing to continue with the project expressed slow growth rates of trees as a principal reason. All but one of these farmers had growth rates for his/her trees markedly below those of the species selection trial nearby. Some farmers wishing to continue had very high mortality rates (>70% for all species), suggesting non-tangible benefits for participating in project activities. Other species that were not tested on-farm but grew well in the species-selection trials and were of interest to local farmers are discussed.As long as specific site and management conditions are carefully considered, data from species selection trials can be useful in informing tree planting projects with rural farmers; however, care should be taken to manage expectations.  相似文献

Mutational analysis of the tyrosine phosphatome in colorectal cancers   总被引:1,自引:0,他引:1  
Tyrosine phosphorylation, regulated by protein tyrosine phosphatases (PTPs) and kinases (PTKs), is important in signaling pathways underlying tumorigenesis. A mutational analysis of the tyrosine phosphatase gene superfamily in human cancers identified 83 somatic mutations in six PTPs (PTPRF, PTPRG, PTPRT, PTPN3, PTPN13, PTPN14), affecting 26% of colorectal cancers and a smaller fraction of lung, breast, and gastric cancers. Fifteen mutations were nonsense, frameshift, or splice-site alterations predicted to result in truncated proteins lacking phosphatase activity. Five missense mutations in the most commonly altered PTP (PTPRT) were biochemically examined and found to reduce phosphatase activity. Expression of wild-type but not a mutant PTPRT in human cancer cells inhibited cell growth. These observations suggest that the mutated tyrosine phosphatases are tumor suppressor genes, regulating cellular pathways that may be amenable to therapeutic intervention.  相似文献

The SAR11 clade consists of very small, heterotrophic marine alpha-proteobacteria that are found throughout the oceans, where they account for about 25% of all microbial cells. Pelagibacter ubique, the first cultured member of this clade, has the smallest genome and encodes the smallest number of predicted open reading frames known for a free-living microorganism. In contrast to parasitic bacteria and archaea with small genomes, P. ubique has complete biosynthetic pathways for all 20 amino acids and all but a few cofactors. P. ubique has no pseudogenes, introns, transposons, extrachromosomal elements, or inteins; few paralogs; and the shortest intergenic spacers yet observed for any cell.  相似文献

During the last decades, artificial night lighting has increased globally, which largely affected many plant and animal species. So far, current research highlights the importance of artificial light with smaller wavelengths in attracting moths, yet the effect of the spectral composition of artificial light on species richness and abundance of moths has not been studied systematically. Therefore, we tested the hypotheses that (1) higher species richness and higher abundances of moths are attracted to artificial light with smaller wavelengths than to light with larger wavelengths, and (2) this attraction is correlated with morphological characteristics of moths, especially their eye size. We indeed found higher species richness and abundances of moths in traps with lamps that emit light with smaller wavelengths. These lamps attracted moths with on average larger body mass, larger wing dimensions and larger eyes. Cascading effects on biodiversity and ecosystem functioning, e.g. pollination, can be expected when larger moth species are attracted to these lights. Predatory species with a diet of mainly larger moth species and plant species pollinated by larger moth species might then decline. Moreover, our results indicate a size-bias in trapping moths, resulting in an overrepresentation of larger moth species in lamps with small wavelengths. Our study indicates the potential use of lamps with larger wavelengths to effectively reduce the negative effect of light pollution on moth population dynamics and communities where moths play an important role.  相似文献

Background, Aim and Scope   Coastal and river plains are the surfaces of depositional systems, to which sediment input is a parameter of key-importance. Their habitation and economic development usually requires protection with dikes, quays, etc., which are effective in retaining floods but have the side effect of retarding sedimentation in their hinterlands. The flood-protected Dutch lowlands (so-called dike-ring areas) have been sediment-starved for up to about a millennium. In addition to this, peat decomposition and soil compaction, brought about by land drainage, have caused significant land subsidence. Sediment deficiency, defined as the combined effect of sediment-starvation and drainage-induced volume losses, has already been substantial in this area, and it is expected to become urgent in view of the forecasted effects of climate change (sea-level rise, intensified precipitation and run-off). We therefore explore this deficiency, compare it with natural (Holocene) and current human sediment inputs, and discuss it in terms of long-term land-use options. Materials and Methods: We use available 3D geological models to define natural sediment inputs to our study area. Recent progress in large-scale modelling of peat oxidation and compaction enables us to address volume loss associated with these processes. Human sediment inputs are based on published minerals statistics. All results are given as first-order approximations. Results: The current sediment deficit in the diked lowlands of the Netherlands is estimated at 136 ± 67 million m3/a. About 85% of this volume is the hypothetical amount of sediment required to keep up with sea-level rise, and 15% is the effect of land drainage (peat decomposition and compaction). The average Holocene sediment input to our study area (based on a total of 145 km3) is ~14 million m3/a, and the maximum (millennium-averaged) input ~26 million m3/a. Historical sediment deficiency has resulted in an unused sediment accommodation space of about 13.3 km3. Net human input of sediment material currently amounts to ~23 million m3/a. Discussion: As sedimentary processes in the Dutch lowlands have been retarded, the depositional system's natural resilience to sea-level rise is low, and all that is left to cope is human countermeasure. Preserving some sort of status quo with water management solutions may reach its limits in the foreseeable future. The most viable long-term solutions therefore seem a combination of allowing for more water in open country (anything from flood-buffer zones to open water) and raising lands that are to be built up (enabling their lasting protection). As to the latter, doubling or tripling the use of filling sand in a planned and sustained effort may resolve up to one half of the Dutch sediment deficiency problems in about a century. Conclusions: Conclusions, Recommendations and Perspectives. We conclude that sediment deficiency – past, present and future – challenges the sustainable habitation of the Dutch lowlands. In order to explore possible solutions, we recommend the development of long-term scenarios for the changing lowland physiography, that include the effects of Global Change, compensation measures, costs and benefits, and the implications for long-term land-use options.
