The isolate BOR-3, collected in Slovakia in 1996, was recently identified as a natural recombinant between an M and D type of Plum pox virus (PPV). Biological assays demonstrated its capacity to be aphid- and graft-transmitted to various Prunus spp. hosts. A study was carried out to determine the further presence of PPV recombinants in two epidemiologically distinct areas – Slovakia and France. Tools based on PPV-M and D subgroup typing, targeting P3–6K1, CI and CP regions of the PPV genome were used for recombinant identification. Closely related recombinant variants were detected in different Prunus spp. during a survey conducted in Slovakia in 2001, but not within a set of selected PPV isolates from France collected between 1985 and 2001. Sequence analysis of the (Cter)NIb–(Nter)CP region of 10 recombinant isolates from Slovakia showed their high homology, reaching more than 98%. All the recombinant isolates shared the same recombination breakpoint situated in the C terminus of the NIb gene. Our study demonstrates that the PPV recombinants are viable and competitive with conventional PPV-M and D isolates. The present work indicates that the occurrence of recombinants within PPV isolates might be more common than previously assumed.  相似文献

Characterization of a novel potyvirus isolated from maize in Israel   总被引:1,自引:0,他引:1  
A potyvirus (proposed name of Zea mosaic virus [ZeMV]) isolated from maize in Israel was analyzed by serology, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) of capsid proteins, symptomatology, and sequencing. Parts of the nuclear inclusion b, coat protein, and 3' regions were sequenced; the amino acid sequence of ZeMV capsid was determined by time-of-flight mass spectrometry (TOFMS). The results of these analyses were compared with those of similar analyses of the following potyviruses: Maize dwarf mosaic virus (MDMV), Sugarcane mosaic virus strain MDB (SCMV-MDB), Johnsongrass mosaic virus(JGMV), Sorghum mosaic virus (SrMV), and an isolate of MDMV from Israel. Indirect enzyme-linked immunosorbent assay using ZeMV antiserum detected only ZeMV, and reciprocal tests using MDMV, JGMV, or SrMV antisera failed to detect ZeMV. ZeMV cross-reacted weakly when SCMV-MDB antiserum was used. The mass of ZeMV capsid was determined to be 36,810 Da by SDS-PAGE and 34,216 Da by TOFMS. The ZeMV systemically infected johnsongrass (Sorghum halepense), but did not infect oat (Avena sativa), pearl millet (Pennisetum glaucum), barley (Hordeum vulgare), or rye (Secale cereale). Necrosis was caused in 19 sorghum lines by SrMV, in 15 by ZeMV, in 14 by MDMV, and in 5 by JGMV and SCMV-MDB. The nucleic acid and amino acid sequences of ZeMV clearly showed that it is not a strain of JGMV, MDMV, SCMV, or SrMV.  相似文献

Trace metals: Cd, Cu, Cr, Ni, Pb and Zn and persistent organicpollutants: polycyclic aromatic hydrocarbons (PAHs) and polychlorobiphenyls (PCBs) were investigated in a fraction ofsediments characterized by a granulometry < 200 μm. Coresfrom 17 to 54 cm depth, were collected at two sites of theSeine river upstream (Port à l'Anglais) and downstream (Saint-Cloud) from Paris and at one site of the Marne river (Saint-Maurice). Marne river sediments displayed higher Cu andPb levels than those of the Seine river. Except for Cd and Cr,contents did not vary notably in the Seine river sediments.Metal contents, except Pb, were significantly correlated withthe sediment fraction which granulometry is < 50 μm,particularly at Port à l'Anglais (p < 0.001). Correlationcoefficients between Cr, Cu, Ni and Zn contents and clayfractions of the cores ranged from 0.85 to 0.99 at both sitesof the Seine river. PCB levels varied widely, ranging from 27to 689 μg kg-1 and were highly correlated with bothsediment fraction < 50μm and particulate organic carbon(POC) ratio. PAH levels ranged from 2.30 to 41.3 μg g-1 and displayed the highest values close to dams; theyshowed no correlations with sediment fraction < 50 μm and POC.  相似文献

The effect of triglycerides (Tg) on GRF-mediated GH secretion was examined in 2 groups of twelve ten-day old male calves. Twelve calves were intravenously infused with a lipid-heparin solution (5 mg Tg and 0.3 IU heparin/kg body wt/min for 90 min). The twelve control calves received in the same way, the same volume of saline. Thirty minutes after the start of infusion, GRF 1–29 (human amide, 0.16 μg/kg body wt) was intravenously injected in six animals of each group.

Mean plasma GH levels reached peak concentrations in the 2 groups 5 min after GRF injection. However the area under the GH response curve, when lipid-heparin was given, was significantly diminished compared to the response when saline was given. In the same time, lipid-heparin treatment increased plasma SRIF concentration. These data suggest that an increase in plasma Tg concentration, induced by lipid-heparin infusion, inhibits GRF-mediated GH secretion, possibly through stimulation of SRIF secretion.  相似文献

The transportation pathways of triazines were investigatedfrom their origins onwards, in the Marne and the Seine rivers(France). According to the 1991 survey, the highest total triazine concentration levels (956 ng L-1) were found in the river Marne: 580 ng L-1 for atrazine and 200 ng L-1 for simazine, in relation with agricultural practices.The total triazine levels reached 829 ng L-1 in the riverSeine. The other triazines remained low, from 5 to 75 ng L-1. The triazine fluxes were 226 and 404 kg for a 118 d period in the Marne and the Seine rivers, respectively. Out of the treatment period, triazine peaks were related to riverflows with a 48 hr delay between the peak and the top of theflood. After herbicide treatment, peak concentrations wereclosely related with the precipitations. Concerning theriver Marne tributaries, highest atrazine concentrationswere found in June, in those which drained the vineyardarea. In January, the concentrations were higher inperiurban areas underlining their contribution to theherbicide inputs in the rivers. On the whole, the ratio oftriazines transported in the two rivers represented 0.2%of the total amount applicated annually on their catchment basins.  相似文献

This study was conducted to determine whether the daily consumption for 84 days of tyrosol and hydroxytyrosol, the main olive oil phenolic compounds, and olive oil mill wastewater (OMWW), a byproduct of olive oil production, rich in micronutrients, may improve bone loss in ovariectomized rats (an experimental model of postmenopausal osteoporosis) and in ovariectomized rats with granulomatosis inflammation (a model set up for senile osteoporosis). As expected, an induced chronic inflammation provoked further bone loss at total, metaphyseal, and diaphyseal sites in ovariectomized rats. Tyrosol and hydroxytyrosol prevented this osteopenia by increasing bone formation ( p < 0.05), probably because of their antioxidant properties. The two doses of OMWW extracts had the same protective effect on bone ( p < 0.05), whereas OMWW did not reverse established osteopenia. In conclusion, polyphenol consumption seems to be an interesting way to prevent bone loss.  相似文献

The relationships between some organochlorines and metals in the water and in the zebra mussel of the river Seine have been studied in the Parisian area. The bioconcentration of PCBs by the mussel (Kb = 17 × 103) was about 10 fold higher than for other organochlorines. The accumulation and the release response of the mussel to alterations of the water organochlorine levels occurred within a period smaller than 7 days. The correlation between the water and the mussel levels is highly significant for γHCH and PCBs (r=0.94, p<0.01 and r=0.87, p<0.05, respectively). The Pb bioconcentration (0.15 × 103) was about 20 fold lower than the Cu and Cr (3.4 and 3.0 × 103, respectively). The Cu and the Pb levels in the water and in the mussel followed a parallel course. Cu and Pb appear to be taken up and released relatively rapidly within a 7 day period. Comparisons between the different stations along the river Seine showed that the highest pollutant evels in the water were found downstream Paris and were fairly well reflected by the mussel levels. However, when pollution reaches top values, the mussel metabolic activities are affected and thus, the bioconcentration ability too.  相似文献
