The number of goblet cells containing neutral and acidic mucins, including sulphomucins and sialomucins, was investigated in the small intestine of goat kids fed with lyophilized bovine colostrum in the period of passive immunity acquisition. At 0, 7 and 14 h of life, 15 male newborns received 5% of body weight of lyophilized bovine colostrum (LBC) and 14 male newborns received goat colostrum (GC), both with 55 mg/mL of IgG. Three additional animals were sampled at birth, without colostrum intake. Duodenum, jejunum and ileum samples were collected at 18, 36 and 96 h of life. Histological stains, periodic acid-Schiff, 1% alcian blue pH 2.5 and 1% alcian blue pH 1.0 were used to identify neutral and acidic mucins and acidic sulphated mucins, respectively. The number of goblet cells containing neutral and acidic mucins, including sulphomucins and sialomucins, does not differ in the duodenum (P>0.05). In the jejunum, LBC showed a higher number of goblet cells containing sialomucins compared to GC (P<0.05). The highest number of goblet cells containing acidic and neutral mucins and total number of goblet cells were observed at 96 h (P<0.05). In this segment, vacuoles of colostrum were present at 18 and 36 h mainly in the upper region of the villi, while the goblet cells were located at the bottom. At 96 h, vacuoles of colostrum were not detected, only goblet cells distributed throughout the villi. In the ileum, the number of goblet cells containing sulphomucins was higher (P<0.05) at 96 h than at 18 h. The LBC group showed higher (P<0.05) number of goblet cells containing sulphomucins at 96 h and total number of goblet cells at 36 and 96 h than the 0-h group. The present work revealed that the greater the absorption of colostrum in the goat kids' jejunum epithelium, the smaller the number of goblet cells. Considering this segment, feeding newborns with heterologous colostrum caused alteration in the number of goblet cells containing sialomucin. This condition suggested a reaction of the intestinal epithelium with increasing secretion due to the presence of non-recognized substances from the lyophilized bovine colostrum. 相似文献

Enzyme activity was evaluated in the intestine of juvenile pacu, Piaractus mesopotamicus, fed diets containing 0, 10 or 20 % of lyophilized bovine colostrum (LBC) inclusion for either 30 or 60 days. The enzymes intestinal acid and alkaline phosphatase (ACP and ALP, respectively), nonspecific esterase (NSE), lipase (LIP), dipeptidyl aminopeptidase IV (DAP IV) and leucine aminopeptidase (LAP) were studied using histochemistry in four intestinal segments (S1, S2, S3 and rectum). Moderate activity of the DAP IV was detected in the three last intestinal segments, but no differences among the treatments were detected. Enzymes LAP, NSE and LIP were weakly stained in all intestinal segments and the inclusion of 10 or 20 % of LBC in the diet commanded a moderate reaction to NSE in the S3 segment at day 60. ACP activity was detected only in the brush border of the S1 segment of fish fed 0 % LBC for either 30 or 60 days. The activity of ALP was very strong in the first intestinal segment, but a weak reaction was seen in the last segments. The inclusion of 20 % of LBC changed the pattern of staining to the ALP, eliciting moderate staining in S2 at day 30 and S1 at day 60. The consumption of diets containing LBC by juvenile pacu did not have significant implications in intestinal enzymatic activity, which still was not fully stimulated. 相似文献

Proline plays important roles in various stages and processes of plant development. However, there are few reports on the effect of endogenous proline accumulation on germination and seedling vigour under suitable conditions of germination and development. The aim of this work was to evaluate the effect of endogenous proline in transgenic tobacco (Nicotiana tabacum L.) constitutively expressing the VaP5CSF129A gene in germination and seedling vigour. Under optimal conditions of germination, the transgenic seeds had high proline content when compared to the non-transgenic plants. However, this higher accumulation did not result in better germination or seedling vigour, contrasting to reported results for exogenous proline application. The pre-treatment by water submersion indicated that the lowest initial seed germination with the highest proline concentration may be due to their greater post-harvest dormancy. When newly germinated seedlings were subjected to osmotic stress (?0.9 MPa), the free proline content increased proportionally in all genotypes and the transgenic events seedlings showed greater root length compared to those of the non-transformed control. This can be advantageous as, in theory, seedlings with longer roots may have a better chance of growing and exploring the different soil layers, allowing the transgenic events to be more tolerant to edaphic constraints. 相似文献

Immunoglobulins in piglets from sows heat-stressed prepartum 总被引:1,自引:0,他引:1
Sows were subjected to moderate heat stress in a chamber (32 C) from d 100 of pregnancy until less than 8 h before delivery of first piglet, while control sows were in a thermoneutral chamber (21 C) or farrowing house (22 C). Blood serum and colostrum at parturition of heat-stressed sows and their piglets' serum at birth had elevated cortisol concentrations. Total protein, globulin and immunoglobulin G (IgG) concentrations in sow serum tended to decrease as parturition time was approached; albumin did not change. Total protein and IgG concentrations in colostrum at parturition and in milk 24 and 48 h later tended to be lower in heat-stressed sows. Concentrations of these four protein fractions (total, globulin, IgG and albumin) in piglet serum at birth did not differ among treatment groups, but soon after colostrum ingestion they increased markedly in all groups. Therefore, in all groups total protein remained constant while globulin and IgG decreased. Globulin concentration on d 1 was lowest in piglets from heat-stressed sows, but its rate of decrease after d 1 was not affected by sow treatment. Immunoglobulin G concentration was 11 mg/ml lower, but its rate of decrease through postnatal d 20 was slower in piglets from heat-stressed sows than in those from control sows; a 10-mg/ml difference in IgG concentration on postnatal d 1 has been associated with increased preweaning mortality in piglets. Higher cortisol concentration in serum and lower IgG in colostrum of sows under heat stress was associated in their piglets with higher serum cortisol at birth and lower serum IgG for the first 20 d postnatum. 相似文献
