BACKGROUND: Serum protein analysis in both humans and experimental animal species has so far been carried out by labor-intensive techniques, such as agarose gel electrophoresis (AGE). OBJECTIVE: The objective of this study was to evaluate capillary electrophoresis (CE) as an alternative technique to AGE for the analysis of serum proteins from healthy animals. METHODS: Blood samples were collected into tubes without anticoagulant from 6 fasted healthy male mice, rats, dogs, marmosets, and humans. Serum proteins were separated by CE using a technique standardized for the analysis of human proteins, and the results (efficiency, resolution, and precision) were compared with those obtained through AGE. RESULTS: Compared with AGE, CE resulted in narrower peaks and more peaks. The efficiency of protein separation by CE was significantly higher for all species, and resolution (R) was significantly higher in samples from dogs. Using rat serum, intraday reproducibility was lower for all protein fractions, and interday reproducibility was lower for most peaks, compared with AGE. CONCLUSIONS: We conclude that CE is a viable alternative to AGE for the determination of protein electrophoresis in a routine veterinary clinical pathology laboratory. The minimal sample requirement (2 microL), complete automation, and quantitative results make CE an especially valuable technique for protein analysis in experimental animal models.  相似文献

Growth of cassava was investigated in Central Africa (Congo) on crops attacked by two phytophagous mites: Mononychellus progresivus and Oligonychus gossypii. Change and distribution of dry matter in leaves, stems and roots were monitored for the 24 month crop cycle on infested and mite-free plants. Leaf dry matter increased during the rainy season and decreased during the dry season. Stems contain the reserves used for initiating plant growth after the dry season. Dry matter increased in the roots which form true storage organs. At the start of the second year of cultivation, one third of the root biomass was used to support the recovery of plant growth and development. Mite densities fewer than 50 mobile forms per leaf (maximum number of mites) were not sufficient to cause significant loss of leaf, stem and root dry matter. Nevertheless the mites diverted part of the biomass allocated to the roots thereby reducing the efficiency of storage root production in the infested plot.  相似文献

A thin film of well-ordered anatase TiO2 nanotubes prepared by anodic oxidation of titanium metal were synthesised and used as adsorbent medium for the purification of water from aqueous uranium and lead. The amount of subtracted metal ions was quantified by using X-ray photoelectron spectroscopy at the surface of the reacted TiO2 surface. Batch experiments for the sorption of U and Pb at the surface of the titania substrate were carried out in separated solution equilibrated with air of uranyl acetate and lead nitrate, in the pH range 3?C9. For uranium, the experiments were also repeated in anoxic (N2) atmosphere. The amount of metal ions adsorbed onto the titania medium was quantified by measurements of the surface coverage expressed in atomic percent, by recording high-resolution XPS spectra in the Ti2p, U4f and Pb4f photoelectron regions. Adsorption of the uranyl species in air atmosphere as a function of pH showed an adsorption edge near pH 4 with a maximum at pH 7. At higher pH the presence of very stable uranyl?Ccarbonate complexes prevented any further adsorption. Further adsorption increased until pH 8.5 was obtained when the uranyl solution was purged from dissolved CO2. Lead ion showed a sorption edge at pH 6, with a maximum uptake at pH 8. The results showed that the uptake of uranium and lead on the selected titania medium is remarkably sensitive to the solution pH. This study demonstrates the reliability of this type of material for treating water polluted with heavy metals as well as leachates from radioactive nuclear wastes.  相似文献

Bemisia tabaci adults were collected from pepper and melon at different commercial production greenhouses in Argentina and Uruguay. The biotype status of adults was then established using cytochrome oxidase I gene (mtCOI) as molecular marker. Only the Q biotype was found on all plants sampled. 1. This study aimed to provide information on the response of laying-type pullets to dietary threonine (THR) during three periods of growth prior to the onset of lay. Different batches of Dekalb White pullets were used in three separate trial periods (from 4 to 6, 8 to 11 and 13 to 16 weeks of age) using 8 dietary THR concentrations in each period, using a completely randomised design, and with each treatment being replicated 6 times, using 15 birds per replication in period 1 and 8 birds in periods 2 and 3. In period 1 the THR content (THRc) ranged from 2.3 to 7.6 mg/g, in period 2 from 1.7 to 5.5 mg/g, and in period 3 from 1.4 to 4.7 mg THR/g feed.

2. Body weight gain, food intake and the deposition of protein and lipid in the feather-free body and in the feathers were measured in each period. Linear regressions were fitted to all data falling below the break point defined by the broken stick regression, to estimate the efficiency of utilisation of THR. The maximum protein growth rate was 4.0 ± 0.2, 5.3 ± 0.4 and 3.5 ± 0.5 g/d in periods 1, 2 and 3, respectively.

3. The efficiency of utilisation of dietary THR for THR deposition in each period was the same, at 0.85 ± 0.1 mg/mg. As dietary THRc decreased, the amount of body lipid deposition increased.

The extensive use of chlorotriazines as selectiveherbicides in agriculture and their relatively highpersistence imply that these compounds are now presentin the environment, contaminating surface and groundwater. In European countries, United States andCanada, the drinking water ordinance demands a limitedconcentration of 0.5 μg L-1 for the sum of allpesticides and 0.1 μg L-1 with respect to eachcompound, implying on the necessity of sensitive andselective analytical methods. In the present study wedescribe two methods for the analysis of atrazine,simazine and ametryn residues in surface and groundwater collected from the Espraiado Stream watershed,Ribeirão Preto region, SP, Brazil. The HPLC methodused for sample screening was based on herbicideextraction with dichloromethane:isopropanol (9:1, v/v)followed by reversed-phase chromatography (RP-8) withdetection at 220 nm. The presence of herbicides wasconfirmed by GC-MS after ethyl acetate extraction. Atotal of 250 samples collected at different sites fromOctober 1995 to July 1996 were analyzed. Ametrynresidues were detected in 17 samples but almost alwaysat concentrations below those maximum levels recommended by international agencies of environmental control.  相似文献
