It has been shown that Escherichia coli isolates from lesions of cellulitis belong to a limited number of clonal groups distinct from those of isolates found in the environment of these birds. In this study, different in vitro methods were used to evaluate adherence properties of E. coli isolates from cellulitis lesions and environments of high- and low-cellulitis prevalence broiler flocks. One hundred isolates were tested by hemagglutination. Adherence to frozen sections of chicken skin and binding to soluble fibronectin were examined for 40 of these 100 isolates by immunofluorescence and by immunocytofluorometry, respectively. Localization of bacterial adherence to skin tissues was confirmed by immunohistochemistry. It was demonstrated that O78:K80 isolates from cellulitis lesions adhered to skin sections to a much greater extent in deeper than in superficial tissue layers. A greater bacterial adherence following growth in TSB at 37 C was demonstrated for isolates from flocks with high prevalence of cellulitis than for isolates from flocks with low prevalence of cellulitis. MANOVA analysis results showed a significant difference between superficial and deep tissue layers only for one set of isolates from flocks with high prevalence of cellulitis. Hemagglutinating activity was variable among the O78:K80 isolates obtained from flocks with high prevalence of cellulitis. The results obtained for some O78:K80 isolates following growth in TSB suggest a role for type 1 fimbriae or F1 in adherence to skin sections. This was reinforced by the finding that adherence was inhibited by D-mannose. Poultry E. coli isolates that express F1 had no affinity for soluble fibronectin, although localization of the adherence in skin sections suggested a role for extracellular matrix components such as collagen and insoluble fibronectin.  相似文献

Only a few of the registered insecticides against Cydia pomonella L. are still effective in areas where insecticide resistance has emerged in this pest. Resistance mechanisms are multiple, and their lone or cumulative effects in a single population are not completely understood. A detailed estimation of resistance spectrum is still required to define the suitable insecticides to use against a given population. The efficacy of ten insecticides was therefore investigated together with the resistance mechanisms expressed in four laboratory strains and 47 field populations of C. pomonella from five countries. Bioassays were performed using topical applications of diagnostic concentrations on diapausing larvae, and resistance mechanisms were analysed on adults emerging from control insects. All populations exhibited a reduced susceptibility to at least one insecticide when compared with the susceptible laboratory strain. Cross-resistances were observed between azinphos-methyl or phosalone and more recent compounds such as spinosad and thiacloprid. Resistances to azinphos-methyl, diflubenzuron, spinosad, tebufenozide and thiacloprid were significantly correlated with mixed-function oxidase activity, while increased glutathione-S-transferase and reduced non-specific esterase activities were correlated with resistance to azinphos-methyl and emamectin, respectively. Conversely, resistances to azinphos-methyl, tebufenozide and thiacloprid were negatively correlated with increased esterase activity. None of the observed mechanisms explained the loss of susceptibility of populations to chlorpyrifos-ethyl, and no significant correlation was detected between resistance to deltamethrin and the presence of the kdr mutation. The suitability of such non-target instars to monitor insecticide resistance in field populations is discussed.  相似文献

Context

Parasites can induce strong effects on their host’s growth, not only as a result of host resource exploitation (growth loss) but also with a potential adaptive value for host (tolerance response) and themselves (increased transmission).

Aims

We assessed these three types of phenotypic changes in oak seedlings infected by powdery mildew.

Methods

A manipulative field experiment with three levels of parasite inoculum was designed in order to tease apart infection from genetic effects on oak growth. Seedlings were monitored during 3 years for height growth, phenology and infection.

Results

Powdery mildew infection induced both significant growth loss and qualitative changes in plant architecture. The most striking and unexpected change was increased growth polycyclism in infected seedlings. This benefitted both the host as a form of compensation for infection-caused height loss, and the pathogen, by increasing sporulation.

Conclusion

The study highlights the effect of parasites in the expression of plant phenotypic traits, such as phenology and ultimately tree architecture. Both host tolerance and parasitic manipulation may be involved in the observed changes in growth patterns. These results suggest a complex interplay between development and defence in trees and emphasize the need to better assess tolerance mechanisms when considering the defence strategies of trees against pathogens.  相似文献

Context

Climate change is expected to increase forest vulnerability through disturbances such as windstorms and droughts. Forest managers are therefore investigating strategies to increase forest resistance and resilience, especially by promoting uneven-aged and mixed forests through group selection, and by reducing stand stocking and large trees proportion. However, there is little information on the long-term impacts of these two practices.

Aims

The objectives of this study were (1) to develop an original silviculture algorithm designed for uneven-aged management and (2) to use it to assess the effects of the above-mentioned management methods in long-term simulations.

Methods

We simulated individual and group selection techniques in order to study the effects of group size, harvesting intensity and their interactions on wood production, stand heterogeneity, and regeneration in mountain spruce–fir forests. We used the spatially explicit individual-based forest model Samsara2 to simulate forest dynamics.

Results

Our simulation results confirmed the positive effect of group selection practices on structure diversity and regeneration but not on spruce maintenance. Increasing harvesting intensity enabled forest destocking but decreased structure diversity and led to non-sustained yields for the most intensive scenarios.

Conclusion

As adaptation measure, we thus recommend moderate group selection harvesting creating 500 m2 gaps.  相似文献

Following a survey involving wild oil palm groves in Côte d’Ivoire as a whole in 1968, 17 parents were chosen at Yocoboué (southernCôte d’Ivoire). Agronomic performance of the 17 parents was assessed through Deli × Yocoboué hybrids in 3 trials grown at La Mé (Côte d’Ivoire), Mondoni and La Dibamba (Cameroon).When compared to the Deli × La Mé progenies used as controls, the Deli × Yocoboué hybrids produced 91% for total bunch weight and86% for oil yields. However, the genetic variability within the Deli × Yocoboué progenies allows to select individual progenies as productive as the Deli × La Mé controls, for breeding purposes. A study of major agronomic traits led to the choice of parent YO 11 T , which combines the largest number of favourable traits. On average, Deli × Yocobouéprogenies have slower vertical growth and are susceptible to Fusarium wilt, except 2 parents (YO 3 T and YO 9 T), which seem to be resistant.  相似文献

• Introduction   

Phosphorus (P) is often the first or second element limiting aboveground net primary productivity of forests. Besides low available inorganic orthophosphate (Pi) concentrations, soil may contain high total P contents, as insoluble mineral P or as organic P. Most plants form mycorrhizal associations that improve their P nutrition. Three main hypotheses have been proposed to explain this positive effect through an increase of (1) P mobilisation from mineral P, (2) P mobilisation from organic P and (3) soil exploration and P uptake. However, the positive effect of mycorrhizal symbiosis may be variable with the fungal species forming the association. This could be due to the different abilities of mycorrhizal fungi to mobilise P and/or to take up Pi from the soil.  相似文献

Oscillations of the Sun have been used to understand its interior structure. The extension of similar studies to more distant stars has raised many difficulties despite the strong efforts of the international community over the past decades. The CoRoT (Convection Rotation and Planetary Transits) satellite, launched in December 2006, has now measured oscillations and the stellar granulation signature in three main sequence stars that are noticeably hotter than the sun. The oscillation amplitudes are about 1.5 times as large as those in the Sun; the stellar granulation is up to three times as high. The stellar amplitudes are about 25% below the theoretic values, providing a measurement of the nonadiabaticity of the process ruling the oscillations in the outer layers of the stars.  相似文献

In the codling moth Cydia pomonella (L), insecticide resistance genes have been associated with pleiotropic effects affecting phenology. In this paper, we investigated whether an increase in the frequency of insecticide resistance in field populations of C pomonella was likely to entail significant divergences in the temporal occurrence of both susceptible and insecticide-resistant individuals. For this purpose, we built a phenological model that provided suitable predictions of the distinct and diverging seasonal evolutions of populations of a susceptible and two insecticide-resistant (at two and three loci) homozygous genotypes of C pomonella. Model simulations for each genotype were further compared with pheromone trap catches recorded in a field insecticide-treated population over an 8-year period (from 1992 to 2000), which reflected the progressive annual increase in the frequency of resistance in southeastern France. We found a significant delay in field adult emergence relative to those predicted by the homozygous susceptible model, and the magnitude of such a delay was positively correlated with increasing frequencies of insecticide resistance in the sampled field population of C pomonella. Adult emergence predicted in the theoretical population that was homozygous for resistance at two loci converged with those recorded in the field during the investigated 8-year period. This suggested that the pleiotropic effects of resistance were likely to result in a significant phenological segregation of insecticide-resistant alleles in the field. The results of this study emphasized the potential for pest populations exposed to chemical selection to evolve qualitatively with respect to phenology. This may raise critical questions regarding the use of phenological modelling as a forecasting tool for appropriate resistance management strategies that would take into account the diverging seasonal evolutions of both insecticide resistance and susceptibility.  相似文献
