This study was aimed to develop captive breeding and nurture the young of stonesucker (Garra ceylonensis, Bleeker), a small threatened freshwater fish species endemic to Sri Lanka. Breeding of stonesucker was possible under indoor conditions without hormonal stimulation. They were single spawners having a fecundity of 562±176 eggs per fish (total length of 7.9±0.6 cm) with an egg size of 770–1877 μm. Duration of development stages were, egg to hatchling 36–48 h, hatchling to postlarva stage 3–4 days, postlarva to juvenile stage 5–64 days and adult stage in 300–365 days at 26–28°C. Juveniles exhibited bottom dwelling habit in 70–75 days. Postlarva fed with formulated feed exhibited the highest growth (P<0.05) compared with live diets, Artemia and microworm (Anguillula sp.). Although the juveniles revealed a significantly high growth performance (P<0.05) with swine liver diet up to day 45 compared with formulated feed and combination of detritus and plankton, formulated feed exhibited an increase in growth from day 50. Therefore, formulated feed could be used for rearing of postlarva and juvenile stages without any live feeds.  相似文献

A high plasma cholesterol level, especially low-density lipoprotein cholesterol, indicates increased risk of cardiovascular diseases. Plasma cholesterol levels are influenced by diet and cholesterol biosynthesis, uptake, and secretion. Cholesterol uptake involves solubilization into complex phospholipid spherical bodies termed micelles that facilitate the transport of lipids through the gut brush border membrane into enterocytes. In vitro assays reported to date to determine potential cholesterol-lowering effects of various compounds require artificial micelle preparations that are elaborate and time-consuming to prepare. The aims of this study were to compare the efficacy of artificially prepared micelles with naturally derived micelles from pig's bile and to test their ability to assess potential inhibitors of cholesterol uptake. The suitability of pig's bile-derived micelles was tested both at the level of the micelle and at cellular uptake using cultured Caco-2 cells. Known cholesterol uptake inhibitors at the micelle (green tea catechins) and at the Caco-2 cell (beta-lactoglobulin-derived peptide, IIAEK) were used as reference inhibitory compounds. It was concluded that pig's bile was a rapid, reproducible, convenient, and cost-effective source of micelles for cholesterol micelle solubility and cellular uptake assay systems and is suitable for screening purposes focused on identifying potential cholesterol-lowering agents.  相似文献

Crude extracts of evening primrose meal were prepared in 56% (v/v) acetone and separated into six fractions (I-VI) using a Sephadex LH-20 column. Qualitative tests for phenolic and vanillin positive compounds produced positive results for all fractions. Silica gel thin-layer chromatography of fractions III and V allowed the location and isolation of two spots containing moderate to strong antioxidative compounds. High-performance liquid chromatography of the spot isolated from fraction III showed the resolution of two structurally related compounds, whereas that of the spot from fraction V showed the presence of one compound. Nuclear magnetic resonance spectroscopy and electron impact mass spectrometry produced sufficient evidence to identify the isolated compounds as (+)-catechin, (-)-epicatechin, and gallic acid. These compounds accounted for about 10.5 and 1.7% of the dry mass of the crude extracts and meal, respectively.  相似文献

The effect of germination and growth under illuminated and dark environments on canola seed reserves was investigated. Depletion of proteins and lipids in whole seedlings and their top (leaf/cotyledons) and bottom parts (stem/roots/seed coat) was independent of light, whereas the protein solubility increased at a faster rate under an illuminated environment than in the dark. A rapid increase in free fatty acids but a net decrease of dry matter content in seedlings grown in the dark environment was observed. The dry matter content of seedlings grown in the illuminated environment increased due to photosynthetic biomass accumulation.  相似文献

Twelve edible plant extracts rich in polyphenols were screened for their potential to inhibit oxidation of low-density lipoprotein (LDL) in vitro and to modulate LDL receptor (LDLr) activity in cultured HepG2 cells. The antioxidant activity (inhibition of LDL oxidation) was determined by measuring the formation of conjugated dienes (lag time) and thiobarbituric acid reagent substances (TBARS). Betel leaf (94%), cashew shoot (63%), Japanese mint (52%), semambu leaf (50%), palm frond (41%), sweet potato shoot, chilli fruit, papaya shoot, roselle calyx, and maman showed significantly increased lag time (>55 min, P < 0.05) and inhibition of TBARS formation (P < 0.05) compared to control. LDLr was significantly up-regulated (P < 0.05) by Japanese mint (67%), semambu (51%), cashew (50%), and noni (49%). Except for noni and betel leaf, most plant extracts studied demonstrated a positive association between antioxidant activity and the ability to up-regulate LDL receptor. Findings suggest that reported protective actions of plant polyphenols on lipoprotein metabolism might be exerted at different biochemical mechanisms.  相似文献

The effect of canola (Brassica napus L.) seed germination under illuminated and dark environments on the total concentration and the composition of tocopherols and phytosterols in seedlings and extracted oil were investigated. During the first 10 days of germination, a decrease in gamma-tocopherol was offset by an increase in alpha-tocopherol, indicating the interconversion of these isomers. From day 10 to day 20 under illumination, there was a net increase in alpha-tocopherol and total tocopherols suggesting the synthesis of new tocopherols, whereas there was no net increase in tocopherols in dark. Tocopherols were mainly concentrated in the leafy seedling tops rather than in the non-photosynthesizing bottoms, whereas phytosterols were equally distributed across both sections. The total tocopherol content of oil extracted from 20-day-old seedlings was 4.3- to 6.5-fold higher than that of intact seeds. On a dry seedling basis, the content and composition of phytosterols did not change significantly (p > 0.05) over the sprouting period, but the concentration of total phytosterols in the oil fraction increased 4.2- to 5.2-fold. The concentration of these valuable phytochemicals in the oil fraction is largely due to the depletion of oil reserves during germination, as well as the de novo synthesis of new alpha-tocopherol stimulated by the presence of light. Germination may represent a viable means to naturally concentrate these high-value constituents in canola oil, offering improvements in oil quality based on the nutritional value and oxidative protection offered by tocopherols and the health benefits provided by both tocopherols and phytosterols.  相似文献

Nutrient deficiency, high rate of evapotranspiration, and insufficient and erratic rainfall are the critical challenges for crop production in the dryland areas (DLAs) of Sub-Saharan Africa, including Tanzania, where 61% of arable land is prone to drought. In addressing these challenges, field trials were conducted in central Tanzania to evaluate the interactive effects of ripping and tie-ridges with organic (FYM) and inorganic fertilizers (N) on the mitigation of the critical period of soil moisture stress (CPSMS) for sorghum yield performance. Both in situ rainwater harvesting techniques (IRWHT) and flat-cultivated land were integrated with 8 Mg FYM ha–1, 70 Kg N ha–1, and a combination of 35 Kg N ha–1 and 4 Mg FYM ha–1 (N+ FYM). Among the IRWHT, tie-ridges stored a significant water volume of 577 and 457 m3 ha–1, which mitigated the CPSMS by the maximum of 95% and 37% for the above-average rainfall and below-average rainfall season, respectively. However, it only registered the highest grain yield (2.02 Mg ha–1) and biomass (3.46 Mg ha–1) in a below-average rainfall season. The highest overall grain yield (5.73 Mg ha) and biomass (12.09 Mg ha–1) were harvested in ripping with combined fertilizer treatments in an above-average rainfall season, while the lowest grain yield (0.5 Mg ha–1) and biomass (1.2 Mg ha–1) were registered in the flat-cultivation control in the below-average rainfall season. In the latter season, IRWHT increased the mitigation potential in the order; flat cultivation < ripping < tie-ridges; and sorghum yield, highly correlated with drought mitigation index. The results showed that sorghum grain yield and final biomass performance depend on the influence of IRWHT applied, rainfall amount, soil moisture level, integrated fertilizer, and length of the CPSMS. In the above-average rainfall seasons, fertilizers mask the influence of the IRWHT. The opposite is true in the below-average rainfall season. Although ripping N+ FYM resulted in the highest overall yield, the study recommends practicing tie-ridges integrated with N+ FYM due to regular occurrences of low and unreliable rainfall in the dryland areas.  相似文献

Free-radical scavenging, reducing, and phase II enzyme-inducing activities of aqueous and 5% aqueous ethanol extracts of freeze-dried root tissue of four beet (Beta vulgaris L.) strains (red, white, orange, and high-pigment (red) phenotypes) were determined. Aqueous and ethanolic tissue extracts of the regular and high-pigment red phenotypes were most capable of inhibiting metmyoglobin/H(2)O(2)-mediated oxidation of 2-2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) and 2,2'-azobis-(2-amidinopropane) dihydrochloride (AAPH)-mediated bleaching of beta-carotene. These same extracts were also most efficient at reducing ABTS radical cation and inducing quinone reductase in murine hepatoma (Hepa 1c1c7) cells in vitro.  相似文献
