Longitudinal infection data on Mycobacterium avium subspecies paratuberculosis (MAP) was collected on three dairy farms in Northeastern United States during approximately 10 years. Precise data on animal characteristics and animal location within farm were collected on these farms. Cows were followed over time with regard to MAP status during biannual fecal and serum sampling and quarterly serum sampling. Approximately 13 000 serum samples, 6500 fecal samples and 2000 tissue samples were collected during these years. Prevalence of positive samples was 1.4% for serological samples, 2.2% in fecal samples and 16.7% in tissue samples. Infection dynamics of MAP was studied and resulted in a number of potential changes in our understanding of MAP infection dynamics. First, a high prevalence of MAP infection was observed in these herds due to lifetime follow up of cows, including slaughter. Second, two distinctly different infection patterns were observed, so called non-progressors and progressors. Non-progressors were characterized by intermittent and low shedding of MAP bacteria and a virtual absence of a humoral immune response. Progressors were characterized by continuous and progressive shedding and a clearly detectable and progressive humoral immune response. Strain typing of MAP isolates on the three farms identified on two of three farms a dominant strain type, indicating that some strains are more successful in terms of transmission and infection progression. Continuous high quality longitudinal data collection turned out to be an essential tool in our understanding of pathobiology and epidemiology of MAP infections in dairy herds.  相似文献

Covalent organic frameworks (COFs) have been designed and successfully synthesized by condensation reactions of phenyl diboronic acid {C6H4[B(OH)2]2} and hexahydroxytriphenylene [C18H6(OH)6]. Powder x-ray diffraction studies of the highly crystalline products (C3H2BO)6.(C9H12)1 (COF-1) and C9H4BO2 (COF-5) revealed expanded porous graphitic layers that are either staggered (COF-1, P6(3)/mmc) or eclipsed (COF-5, P6/mmm). Their crystal structures are entirely held by strong bonds between B, C, and O atoms to form rigid porous architectures with pore sizes ranging from 7 to 27 angstroms. COF-1 and COF-5 exhibit high thermal stability (to temperatures up to 500 degrees to 600 degrees C), permanent porosity, and high surface areas (711 and 1590 square meters per gram, respectively).  相似文献

We investigated the influence of arbuscular mycorrhizae (AM) and dark septate fungi (DSF) colonisation on cadmium (Cd) accumulation in Arrhenatherum elatius from heavy metal-contaminated sites. AM colonisation disappeared when Cd concentrations in soil increased, while DSF infection was weak but constant throughout the experiment indicating that soil heavy metals are toxic to AM but not to DSF. AM colonisation was greatest when plant Cd concentrations were highest providing evidence that AM colonisation may influence Cd accumulation. In addition, the disappearance of AM and the concomitant reduction of Cd in shoots during seed maturation result in our suggestion that seasonal variation in AM may play a role in protecting developing seeds from soil pollution.  相似文献

Using reproduction parameters as indicators for cattle welfare has limitations and, at best, these parameters should only be viewed as indirect indicators of welfare. On a farm level, measures such as fertility rates emphasize biological performance of the herd but fail to consider the welfare of individual animals. Even on an individual level, the relationship between reproductive effectiveness and animal welfare is complex. Good reproductive performance does not automatically signify good welfare, as domestication and targeted breeding programmes have led to prioritization of high productive and reproductive performance in most modern farm animal species. In this review, we synthesize literature regarding cattle husbandry, reproduction, welfare and their multidimensional relationships. We argue that practices such as artificial insemination or the use of sexed semen may provide potential welfare advantages as these practices reduce the risk of disease transmission and injury or enable selection of specific beneficial traits. Furthermore, they may offer a solution to current practices jeopardizing welfare, such as the management of surplus bull calves in the dairy industry. Conversely, the animals’ ability to perform natural behaviours such as oestrous expression, an aspect arguably contributing to welfare, is often limited on commercial farms; this limitation is particularly evident in housing systems such as tie stalls where movement is restricted. Moreover, common management practices such as oestrus manipulation may lead to negative attitudes in citizens who often regard naturalness as important element of animal welfare.  相似文献

Purpose

The present research aimed to assess the influence of two phosphorous (P) amendments on metal speciation in rhizosphere soil and the soil–plant transfer of metals.

Materials and methods

Complementary experiments were performed: field experiments on a contaminated cultivated soil and laboratory experiments on an uncultivated contaminated soil to highlight the mechanisms involved in metal-phosphorous interactions. In laboratory experiment, P amendments were added at 120 mg P/kg of soluble KH2PO4 amendment and 9,000 mg P/kg of solid Ca5(PO4)3OH amendment.

Results and discussion

Field-culture results showed the possible food-chain contamination due to Pb, Cd, Cu, and Zn phytoaccumulation by pea and mustard plants from a cultivated agricultural soil. Moreover, P-metal complexes were observed by microscopy in the rhizosphere soil. In laboratory experiments, the application of P amendments significantly increased Pb and Zn level in rhizosphere soil compared to control. Phosphate amendments significantly increased metal-P fraction and decreased “oxides” and “organic matter” fractions of Pb and Zn. Soluble-P amendment was more effective than solid P amendment in changing Pb and Zn speciation. The changes in metal speciation are higher in the rhizosphere soil of pea than tomato. Application of P amendments increased Pb and Zn TF root/soil but decreased TF shoot/root.

Conclusions

The effectiveness of in situ metal immobilization technique varies with the type and quantity of applied P amendment as well as plant and metal type.  相似文献

Ten fruit and kernel traits were measured in 152 Irvingia gabonensis and 293 Dacryodes edulis trees from 6 villages in Cameroon and Nigeria. Frequency distribution curves were used to examine the range of variation of each trait of each species in each village and aggregated into national and regional populations. There were differences between the village sub-populations, with regard to the normality (e.g., mean kernel mass of D. edulis) or skewness (e.g., mean flesh depth of D. edulis) of the distribution curves and in the degree of separation between the individual village populations along the x axis, resulting in the development of a bimodal distribution in the regional population. For all traits, populations of both species differed significantly between countries, but only in D. edulis were there significant differences between the Cameroon populations. On the basis of the results of this study, D. edulis can be said to be virtually wild in Nigeria but semi-domesticated in Cameroon, while I. gabonensis is wild in Cameroon and semi-domesticated in Nigeria. These results are discussed with regard to a hypothesis that the range and frequency of variation in the different populations can be used to identify five stages of domestication. From a comparison of the frequency distribution curves of desirable versus undesirable traits, and statistically identifyable changes in skewness and kurtosis, it is concluded that as a result of the farmers’ own efforts by truncated selection, D. edulis is between Stages 2 and 3 of domestication (with a 67% relative gain in flesh depth) in Cameroon, while I. gabonensis in Nigeria is at Stage 2 (with a 44% relative gain in flesh depth). Bacteremia is an important cause of morbidity and mortality in humans. In this study, we focused on the development of an animal model of bacteremia induced by non-typhoidal Salmonella. New Zealand White rabbits were inoculated with a human isolate of non-typhoidal Salmonella strain CVD J73 via the intra-peritoneal route. Blood samples were collected at specific time points and at euthanasia from infected rabbits. Additionally, tissue samples from the heart, lungs, spleen, gastrointestinal tract, liver and kidneys were obtained at euthanasia. All experimentally infected rabbits displayed clinical signs of disease (fever, dehydration, weight loss and lethargy). Tissues collected at necropsy from the animals exhibited histopathological changes indicative of bacteremia. Non-typhoidal Salmonella bacteria were detected in the blood and tissue samples of infected rabbits by microbiological culture and real-time PCR assays. The development of this animal model of bacteremia could prove to be a useful tool for studying how non-typhoidal Salmonella infections disseminate and spread in humans.  相似文献

Since usual processes involve water as absorbent, they appear not always really efficient for the treatment of hydrophobic volatile organic compound (VOC). Recently, absorption and biodegradation coupling in a two-phase partitioning bioreactor (TPPB) proved to be a promising technology for hydrophobic compound treatment. The choice of the organic phase, the non-aqueous phase liquid (NAPL) is based on various parameters involved in both steps of the process, hydrophobic VOC absorption in a gas?Cliquid contactor, and biodegradation in the TPPB. VOC solubility and diffusivity in the selected NAPL, as well as NAPL viscosity, seems to be the main parameters during the absorption step, while biocompatibility, namely the absence of toxic effect of the NAPL towards microorganisms, non-biodegradability and VOC partition coefficient between NAPL and water were revealed as the key factors during the biodegradation step. The screening of the various NAPL available in the literature highlighted two families of compounds matching the required conditions for the proposed integrated process, silicone oils and ionic liquids.  相似文献

This study examined the feeding selectivity of Hippocampus kuda juveniles under captive conditions and evaluates different food organisms that could be used to improve hatchery‐rearing of this species. Newly born H. kuda were reared for 10 days in 60‐L capacity tanks and fed rotifers (Brachionus rotundiformis), zooplankton (mostly Pseudodiaptomus annandalei and Acartia tsuensis) alone or both food sources. The size and amount of food ingested increased as seahorses grew. Selective feeding of seahorses appeared to change as they develop, preferring copepod adults over nauplii and rotifers. A. tsuensis was highly selected by juveniles over P. annandalei. Specific growth rate in terms of body weight (SGR‐BW, 15% day–1) was the highest and mortality rate (9% at day 10) the lowest in seahorses fed a mixed food sources. Slowest growth rate (0.3% day–1) and highest mortality rate (60% at day 7) were observed in seahorses fed rotifers alone. These results indicate that copepods are suitable food for seahorse juveniles, but a mixture of food organisms in the rearing tank environment enhances survivorship and growth of H. kuda, thus potentially providing a source of cultured rather than wild specimens for characterizing the life history of this threatened species.  相似文献
