In recent years, there has been a great demand for artificial materials for biomedical applications, especially to bone growth. In the present study, the in vitro bioactivity of chitosan-acetylated jute blended film was investigated through biomimitic growth of bone-like apatite layer formation in simulated body fluid (SBF) on the surface of the film. Chitosan was prepared from crab shells and the blended film was prepared by solvent casting method using 10 % formic acid aqueous solution of in-house chitosan and mercerized, acetylated jute fiber. Water absorption characteristics (swelling test) and tensile strength (Universal testing machine) of the films were determined. Thermal behavior of the films were examined by TGA analysis. The biocompatibility of the chitosan-acetylated jute (CAJ) film was indicated by the formation of hydroxy apatite (HAp) bone-like layer formed on the surface when soaked in SBF solution for 14 days. The topography of the film was characterized by scanning electron microscope (SEM) and Ca/P ratio of the apatite layer was confirmed to be 50:50 by EDX analysis. The results showed that the CAJ film will be a promising bone substituting material.  相似文献

ObjectiveTo compare the effects of morphine, parecoxib, tramadol and a combination of parecoxib, tramadol and pindolol on nociceptive thresholds in awake animals and their effect on glomerular filtration rate (GFR) in dogs subjected to 30 minutes of anesthesia.AnimalsEight adult mixed breed experimental dogs.Study designRandomized, controlled trial.MethodsDogs received 0.05 mg kg?1 acepromazine subcutaneously (SC) as anaesthetic pre-medication. Thirty to sixty minutes later, they received either tramadol 3 mg kg?1 intravenously, (IV), parecoxib (1 mg kg?1 IV), a combination of tramadol 3 mg kg?1 (IV), parecoxib 1 mg kg?1 (IV) and pindolol 5 μg kg?1 (SC), morphine (0.1 mg kg?1 (IV) or 0.9% saline (2 mL). Anaesthesia was then induced with IV propofol to effect (2.9 ± 0.8 mg kg?1) and maintained with halothane in oxygen for 30 minutes. Systolic arterial blood pressure was maintained above 90 mmHg with IV fluids and by adjusting the inspired halothane concentration. Post-treatment nociceptive thresholds to mechanical stimuli, expressed as percent of pre-treatment values, were compared between the treatments to assess the analgesic efficacy of the drugs. Plasma iohexol clearance (ICL), a measure of GFR, was estimated both before and 24 hours after induction of anaesthesia to study the drugs’ effects on renal perfusion. Nociceptive threshold and GFR data were compared using mixed model analysis in sas®9.1.ResultsBoth tramadol and parecoxib produced similar analgesia, which was less than that of morphine. Their combination with pindolol produced analgesia comparable with morphine. None of the test drugs, either alone or in combination, reduced GFR.ConclusionTramadol and parecoxib (either alone or in combination) can increase nociceptive thresholds in awake dogs and have minimal effects on renal perfusion in normotensive dogs subjected to anaesthesia.  相似文献

Current yeast interactome network maps contain several hundred molecular complexes with limited and somewhat controversial representation of direct binary interactions. We carried out a comparative quality assessment of current yeast interactome data sets, demonstrating that high-throughput yeast two-hybrid (Y2H) screening provides high-quality binary interaction information. Because a large fraction of the yeast binary interactome remains to be mapped, we developed an empirically controlled mapping framework to produce a "second-generation" high-quality, high-throughput Y2H data set covering approximately 20% of all yeast binary interactions. Both Y2H and affinity purification followed by mass spectrometry (AP/MS) data are of equally high quality but of a fundamentally different and complementary nature, resulting in networks with different topological and biological properties. Compared to co-complex interactome models, this binary map is enriched for transient signaling interactions and intercomplex connections with a highly significant clustering between essential proteins. Rather than correlating with essentiality, protein connectivity correlates with genetic pleiotropy.  相似文献

This study was conducted to compare the efficacy of combinations of morphine, dexmedetomidine and maropitant in preventing the changes in electroencephalographic (EEG) indices of nociception in anaesthetized dogs subjected to a noxious electrical stimulus. In a crossover study, eight healthy adult dogs were randomly allocated to four groups: Mor: morphine 0.6 mg/kg; Dex + Mor: morphine 0.3 mg/kg + dexmedetomidine 5 μg/kg; Maro + Mor: morphine 0.3 mg/kg + maropitant 1 mg/kg; and Dex + Maro + Mor: morphine 0.2 mg/kg + dexmedetomidine 3 μg/kg + maropitant 0.7 mg/kg. Following intramuscular administration of test drugs in a minimal anaesthesia model, a supramaximal electrical stimulus (50 V at 50 Hz for 2 s) was applied and the EEG data were recorded. There were significant increases (p < .05) in the poststimulus median frequency (F50) only in groups Mor and Maro + Mor. Dex + Mor group had a significantly lower change in F50 and F95 compared to all other treatment groups. There was no correlation of the changes in EEG frequencies with blood plasma concentration of the drugs during and after noxious stimulation. Combination of dexmedetomidine and morphine was most effective in abolishing the changes in EEG indices in response to a noxious stimulus indicating a supra-additive interaction between these two drugs.  相似文献

This study compared the efficacy of different classes of analgesics in preventing the changes in electroencephalographic (EEG) indices of nociception in anaesthetised dogs, subjected to a standard electrical stimulus. In a crossover study, eight dogs received morphine (0.5 mg/kg) or tramadol (3 mg/kg) or parecoxib (1 mg/kg) or 0.9% saline subcutaneously (SC) at the time of pre-anaesthetic medication. After induction with intravenous propofol, anaesthesia was maintained with halothane at a stable concentration between 0.85% and 0.95%. EEG was recorded in a three electrode montage, using SC needle electrodes, before and after electrical stimulation of dogs during anaesthesia. Post-stimulation median frequency (a reliable indicator of nociception) of the EEG increased significantly in tramadol, parecoxib and saline groups compared to that of morphine. Total EEG power decreased in all treatment groups following stimulation. These results indicate that the changes in EEG responses to noxious stimulation can be used for evaluating anti-nociceptive efficacy of analgesics.  相似文献

Research on marine gastropod breeding in India is in its infancy and scanty literature is only available. To fill the lacunae, marine gastropods of three ornamentally valued and conservation important groups such as Cyprids, Strombids and Muricids were held under captivity and studied for their broodstock maintenance, spawning behaviour, larval rearing and metamorphosis into juveniles at the Shellfish Hatchery of Central Marine Fisheries Research Institute, Tuticorin. Valuable information on the focal themes of the article was obtained for Strombids and Muricids leading to the successful life cycle closing for Lambis lambis and Chicoreus virgineus are reported earlier. In the present study, experiments were conducted on aspects such as post‐metamorphosed juvenile rearing up to 76 days post hatch for L. lambis, spawning, larval development and life cycle closing and reasonable level of juvenile production (8.8%) for Chicoreus ramosus. In addition, the primary observations on the captive breeding of Cypraea tigris with reference to its egg mass, brooding habit and early development is reported. However, while studying these groups few unanswered questions and bottle necks in their breeding nature, larval rearing and metamorphosis arose. The following account details the experiments conducted and results obtained in each of the focal themes of the paper and the constraints faced.  相似文献

Asian cultivated rice shows allelic variation in sodium transporter, OsHKT1;5, correlating with shoot sodium exclusion (salinity tolerance). These changes map to intra/extracellularly-oriented loops that occur between four transmembrane-P loop-transmembrane (MPM) motifs in OsHKT1;5. HKT1;5 sequences from more recently evolved Oryza species (O. sativa/O. officinalis complex species) contain two expansions that involve two intracellularly oriented loops/helical regions between MPM domains, potentially governing transport characteristics, while more ancestral HKT1;5 sequences have shorter intracellular loops. We compared homology models for homoeologous OcHKT1;5-K and OcHKT1;5-L from halophytic O. coarctata to identify complementary amino acid residues in OcHKT1;5-L that potentially enhance affinity for Na+. Using haplotyping, we showed that Asian cultivated rice accessions only have a fraction of HKT1;5 diversity available in progenitor wild rice species (O. nivara and O. rufipogon). Progenitor HKT1;5 haplotypes can thus be used as novel potential donors for enhancing cultivated rice salinity tolerance. Within Asian rice accessions, 10 non-synonymous HKT1;5 haplotypic groups occur. More HKT1;5 haplotypic diversities occur in cultivated indica gene pool compared to japonica. Predominant Haplotypes 2 and 10 occur in mutually exclusive japonica and indica groups, corresponding to haplotypes in O. sativa salt-sensitive and salt-tolerant landraces, respectively. This distinct haplotype partitioning may have originated in separate ancestral gene pools of indica and japonica, or from different haplotypes selected during domestication. Predominance of specific HKT1;5 haplotypes within the 3 000 rice dataset may relate to eco-physiological fitness in specific geo-climatic and/or edaphic contexts.  相似文献

Recovery study was performed at regular intervals to establish the time course of 50% and 100% recovery in neurotransmitter enzyme (acetylcholinesterase, AChE, EC 3.1.1.7) and locomotor behaviour response of mosquito fish, Gambusia affinis exposed to lethal concentration (20.49 mg L−1) of an organophosphorous pesticide, monocrotophos (MCP) for 96 h. In vitro AChE activity studies indicated that MCP could cause 50% inhibition (I50) at 10.2 × 10−5 M. A positive correlation was observed between brain AChE activity and swimming speed during the recovery study. Also, the recovery response of the antioxidant enzymes superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6) and glutathione reductase (GR, EC 1.6.4.2) as well as lipid peroxidation (LPO) as biomarkers of oxidative stress were assessed in viscera of G. affinis. The results showed that the MCP besides its inhibitory effect on target enzyme AChE activity and induction in antioxidant enzyme activities as a characteristic of oxidative stress, which can be used as biomarkers in the pesticide contaminated aquatic streams.  相似文献

Abstract

A study was conducted in the Department of Environmental Sciences, Tamil Nadu Agricultural University, Coimbatore, India, to transform the normal compost into bioactive compost, which has multiple benefits to the crop system. The key players in this transformation process were Azotobacter sp., Pseudomonas sp., Phosphobacteria sp. and the waste materials like poultry litter and spent wash. This enrichment process increases both the quality and nutrient content of the municipal solid waste compost significantly. A study was carried out to evaluate the effect of application of different levels of enriched municipal solid waste compost on the availability of the macronutrient content to the rice field soil. The effect of enriched compost on soil available nutrients was significant. The soil ammonium nitrogen and soil nitrate nitrogen content was found to be high in the plots where the enriched compost was applied along with inorganic fertilizer with the values of 38.87 mg kg?1 and 32.87 mg kg?1, respectively. In addition, the availability decreased towards crop growth. The soil available P and K were also increased with enriched compost application to about 22.46 kg ha?1 and 647 kg ha?1 compared with control values of 19.44 kg ha?1 and 518 kg ha?1, respectively. Both phosphorus and potassium content decreased towards advancement of crop growth.  相似文献
